Protein profiles of enzymatically isolated rumen epithelium in sheep fed a fibrous diet

Background: The rumen wall plays a major role in efficient transfer of digested nutrients in the rumen to peripheral tissues through the portal venous system. Some of these substrates are metabolised in the epithelium during this process. To identify the specific proteins involved in these processes, we used proteomic technologies. Protein extracts were prepared from ventral rumen tissue of six sheep fed a fibrous diet at 1.5× maintenance energy *** a newly developed method, we were able to enzymatically isolate the epithelial cells from underlying tissue layers, thus allowing cytosol and membrane fractions to be independently analysed using liquid chromatography tandem mass spectrometry(LC MS/MS).Results: Using our procedure we identified 570 epithelial proteins in the Ovis aries sequence database. Subcellular locations were largely cytosolic(n = 221) and extracellular(n = 85). However, a quarter of the proteins identified were assigned to the plasma membrane or organelle membranes, some of which transport nutrients and metabolites. Of these 91 were transmembrane proteins(TMHMM), 27 had an N-terminal signal peptide(signalP) and TMHMM motif,13 had a glycosylphosphatidylinositol(GPI) anchor and signalP sequence, 67 had beta(β) strands or 17 β strands and a transit peptide sequence, indicating the identified proteins were integral or peripheral membrane proteins. Subunits of the 5 protein complexes involved in mitochondrial cellular energy production were well represented. Structural proteins(15%), proteins involved in the metabolism of lipids and proteins(26%) and those with steroid or cytokine action were a feature of the ***: Our research has developed a procedure to isolate rumen epithelium proteins from the underlying tissue layers so that they may be profiled using proteomic technologies. The restriction of antibiotic growth promoters in livestock nutrition has led to a growing interest in plant feed additives. This study was designed to assess the production performance of broiler chickens fed on graded levels of Pentadiplandra brazzeana powder in drinking water. A total of 448 day-old Cobb 500 broiler chicks were randomly assigned to seven treatment groups with 4 replicates of 16 birds each (08 males and 08 females). Pentadiplandra brazzeana root was grounded and incorporated at 1, 2, 3, 4 and 5 g/L of drinking water. Control rations consisted of diet without any supplement (T0) and antibiotic (1 g/kg) medicated diet (T0+). Throughout the experimental period (49 days) feed intake was not significantly (p > 0.05) affected by graded levels of Pentadiplandra brazzeana except for 5 g/L of water that significantly (p Pentadiplandra brazzeana/L recorded a higher (p 0.05) affected irrespective of the level of incorporation of the test spice in drinking water. Serum content in urea was significantly lower (p Pentadiplandra brazzeana/L compared to all other treatments. ASAT, ALAT, creatinine, total cholesterol, triglycerides, HDL and LDL-cholesterol were not significantly affected by the inclusion of Pentadiplandra brazzeana powder in drinking water. Feeding chickens with up to 4 g of Pentadiplandra brazzeana significantly increased (p < 0.05) lactic acid bacteria count. Background: This study investigated changes in rumen protozoal and methanogenic communities, along with the correlations among microbial taxa and methane(CH_4) production of six Belmont Red Composite beef steers fed tea seed saponins(TSS). Animals were fed in three consecutive feeding periods, a high-grain basal diet for 14 d(BD period) then a period of progressive addition of TSS to the basal diet up to 30 g/d for 20 d(TSS period), followed by the basal diet for 13 d without TSS(BDP post-control period).Results: The study found that TSS supplementation decreased the amount of the protozoal genus Entodinium and increased Polyplastron and Eudiplodinium genera. During BDP period, the protozoa community of steers did not return to the protozoal profiles observed in BD period, with higher proportions of Metadinium and Eudiplodinium and lower Isotricha. The addition of TSS was found to change the structure of methanogen community at the subgenus level by decreasing the abundance of methanogens in the SGMT clade and increasing the abundance of methanogens in the RO clade. The correlation analysis indicated that the abundance of SGMT clade methanogens were positively correlated with Isotricha, and Isotricha genus and SGMT clade methanogens were positively correlated with CH_4 production. While RO clade were positively correlated with the proportion of Metadinium genus, which was negatively correlated with CH_4 ***: These results suggest that different genera of rumen protozoa ciliates appear to be selectively inhibited by TSS, and the change in methanogen community at the subgenus level may be due to the mutualistic relationships between methanogens and rumen ciliates.

The Keta Lagoon and its catchment areas in Ghana are influenced by intensive agriculture and the use of agro-chemicals. It has therefore, become necessary to assess the quality of water in the lagoon and the surrounding fresh water aquifers. In this study, a water quality index (WQI), indicating the water quality has been adopted. The WQI was determined on a basis of various physico-chemical parameters like pH, conductivity, turbidity, dissolved oxygen, calcium, magnesium, chloride, nitrates, ammonium and sodium. The index was used both for tracking changes at one site over time, and for comparisons among sites. The WQI was also employed to wells used for irrigation on farms along the Keta Sand Spit as well as that of the Keta Lagoon Complex and its surrounding floodplains, in order to ascertain the quality of water for public and livestock consumption, irrigation, recreation and other purposes. The WQI of the wells, Keta lagoon and its floodplains showed various degrees of poor water quality and therefore considered unsuitable for drinking and recreation. By WHO standards, this calls for intensive physical and chemical treatment of the water for human consumption.

The diversity of aquatic resources in Thi Nai Lagoon decreased due to the lack of harmonious use and integrated long-term planning on the use of living *** remained a lack of mechanisms and regulations for coordination of activities in the lagoon by Binh Dinh Provincial People’s Committee(PPC).The aim of the study was to investgate the aquatic species and water quality in Thi Nai Lagoon,*** were collected at 23 different points on Thi Nai Lagoon,with three replicates at each *** were 106 species of fish presented on the lagoon and in different sizes,including 17 mollusc species and 15 crustaceans *** the dry season,there were 167 phytoplankton species in five divisions,10 classes in 39 orders in 59 families including 77 *** had eight chordata in 10 classes of 18 orders including 44 families,64 genus and 102 *** had the largest number of species,78 species,accounting for 76.5%of total *** the rainy season,only 48 species of Arthropoda,Cnidaria,Annelida,Chaetognatha and Rotifer were affected and some were not encountered in the wet season,such as mollusca and *** water quality variables were still good for aquaculture,within permitted limits of TCVN standard and criteria,there were still problems caused by production discharges and living activities of inhabitants around the lagoon,such as sedimentary matters.
