Objective:To investigate the effects of T-2 toxin on expressions of Fas,p53,Bcl-xL,Bcl-2,Bax and caspase-3 on human ***:Human chondrocytes were treated with T-2 toxin(1～20 ng/ml)for 5 ***,p53 and other apoptosis-related proteins such as Bax,Bcl-2,Bcl-xL,caspase-3 were determined by Western blot analysis and their mRNA expressions were determined by reverse transcriptase-polymerase chain reaction(RT-PCR).Results:Increases in Fas,p53 and the pro-apoptotic factor Bax protein and mRNA expressions and a decrease of the anti-apoptotic factor Bcl-xL were observed in a dose-dependent manner after exposures to 1～20 ng/ml T-2 toxin,while the expression of the anti-apoptotic factor Bcl-2 was ***,T-2 toxin could also up-regulate the expressions of both pro-caspase-3 and caspase-3 in a dose-dependent ***:These data suggest a possible underlying molecular mechanism for T-2 toxin to induce the apoptosis sig- naling pathway in human chondrocytes by regulation of apoptosis-related proteins.

Objective To study the allele genetic polymorphism of five short tandem repeat （STR） loci on X-chromosome in Ewenke population of north China and to provide basic data for forensic identification. Methods Genomic DNA was extracted from EDTA-whole blood of Ewenke population by Chelex-100. The DNA samples were amplified by PCR and were analyzed by polyacrylamide gel electrophoresis and silver staining. The sequence length variations of DXS6799, DXS8378, DXS101, HPRTB, and DXS6789 loci on X-chromosome in 98 unrelated Ewenke individuals were investigated. Results All five loci analyzed showed high polymorphism and genetic stability. The data of the five X-chromosome STR loci in Ewenke ethnic group of China was in accordance with Hardy-Weinberg equilibrium by Chi-square test. Conchusion Allele polymorphism of five X-chromosome STR loci can be used as a genetic marker for forensic identification and population genetic research.

A coaxial dielectric barrier discharge plasma jet was designed, which can be operated in atmospheric pressure argon under an intermediate frequency sinusoidal resonant power supply, and an atmospheric pressure glow-like discharge was achieved. Two kinds of typical bacteria, i.e., the Staphylococcus aureus （S. aureus） and Escherichia coil （E. coil）, were employed to study the bacterial inactivation mechanism by means of the non-thermal plasma. The killing log value （KLV） of S. aureus reached up to 5.38 with a treatment time of 90 s and that of E. coil up to 5.36 with 60 s, respectively. According to the argon emission spectra of the plasma jet and the scanning electron microscope （SEM） images of the two bacteria before and after the plasma treatment, it is concluded that the reactive species in the argon plasma played a major role in the bacterial inactivation, while the heat, electric field and UV photons had little effect.

Objective To study the humoral immunity status and distribution pattern of lymphocyte subgroups of peripheral blood mononuclear cell (PBMC) in patients with Kashin-Beck Disease (KBD), and their relationship with erythrocyte selenium. Methods 23 X-ray diagnosed patients, 22 age- and sex- matched healthy children in KBD affected area (KAA), and 25 in KBD non-affected area (KNAA) were randomly selected. Immunohistochemistry with monoclonal antibodies anti-CD4, anti-CD8, anti-CD20 was conducted to analyze the lymphocyte subsets. Serum IgM, IgA, IgG, Complement C3 and C4 were assayed using rate nephelometry (Array 360 System, USA). The contents of erythrocyte selenium was determined by 2,3-diaminonaphthalene fluorescence assay. Results CD4+ and CD8+ cells percentage in PBMCs and serum IgA were significantly lower in KAA than those in KNAA(P 0.05) was found. Conclusion These results suggested that children in KAA had a comparably low cellular immunity level and their humoral immunity status was also in a state of moderate immune suppression. Of this immune disorder in Kashin-Beck disease patients, selenium deficiency probably played a critical role via affecting the distribution pattern of peripheral blood lymphocyte. Selenium-deficiency and immune impairment maybe both have something to do with the cause-effect chain of KBD.

Chromodomain-helicase-DNA-binding protein 1(CHD1)deletion is among the most common mutations in prostate cancer(PCa),but its role remains *** this study,RNA sequencing was conducted in PCa cells after clustered regularly interspaced palindromic repeat(CRISPR)/CRISPR-associated protein 9(Cas9)-based CHD1 *** set enrichment analysis(GSEA)indicated upregulation of hypoxia-related pathways.A subsequent study confirmed that CHD1 deletion significantly upregulated hypoxia-inducible factor 1α(HIF1α)*** investigation revealed that CHD1 deletion upregulated HIF1αby transcriptionally downregulating prolyl hydroxylase domain protein 2(PHD2),a prolyl hydroxylase catalyzing the hydroxylation of HIF1αand thus promoting its degradation by the E3 ligase von Hippel–Lindau tumor suppressor(VHL).Functional analysis showed that CHD1 deletion promoted angiogenesis and glycolysis,possibly through HIF1αtarget *** together,these findings indicate that CHD1 deletion enhances HIF1αexpression through PHD2 downregulation and therefore promotes angiogenesis and metabolic reprogramming in PCa.

Experimental Study on Inactivation of Bacterial Endotoxin by Using Dielectric Barrier Discharge

The low-temperature plasma （LTP） generated by dielectric barrier discharge （DBD） was used to sterilize the *** endotoxin, which is usually difficult to kill by traditional methods. Three different concentrations of bacterial endotoxin （1 EU/mL, 0.5 EU/mL and 0.25 EU/mL） were treated by LTP for different time （20 s, 40 s and 60 s）. Tachypleus amebocyte lysate （TAL） method was employed to detect the concentration variation of bacterial endotoxin before and af- ter the plasma treatment, and endotoxic shock mice model was used to evaluate the inactivation effects of LTP on endotoxin for further study. Experimental results demonstrated that, DBD plasma can inactivate the bacterial endotoxin quickly and effectively, and when the LTP treatment time was increased, the concentrations of bacterial endotoxin decreased gradually （after 60 s plasma treatment, its inactivation effect was beyond the Chinese pharmacopoeia standard）, and the average survival time of mice gradually extended. The possible inactivation mechanisms are proposed to be related to reactive oxygen species （ROSs）.

Objective To analyze the genetic polymorphism of 5 STR loci (D12S304, D12S313, D12S1583, D12S1640 and D12S1708) on chromosome 12 in Chinese Han population. Methods EDTA-anticoagulated blood specimens were collected from unrelated individuals of Chinese Han population in Shaanxi province. DNA samples were extracted with the Wizard Genomic DNA purification Kit and were amplified by polymerase chain reaction (PCR) technique. The PCR products were analyzed by ABI 3100 Genetic Analyzer. Results All 5 loci were in Hardy-Weinberg equilibrium. Allele and genotype frequencies, heterozygosity, power of discrimination, polymorphism information content, probability of paternity exclusion and matching probability of each locus were calculated in Excel 2002. Conclusion They are complex loci with lots of evenly distributed alleles and high heterozygosity in Chinese Han population. Thus they are informative polymorphic loci and valuable DNA marker which represents a superior alternative to many established STRs.

Nerve regeneration following traumatic peripheral nerve injuries and neuropathies is a complex process modulated by diverse factors and intricate molecular *** studies have focused on factors that stimulate axonal outgrowth and myelin ***,recent studies have highlighted the pivotal role of autophagy in peripheral nerve regeneration,particularly in the context of traumatic ***,autophagy-targeting modulation has emerged as a promising therapeutic approach to enhancing peripheral nerve *** current understanding suggests that activating autophagy facilitates the rapid clearance of damaged axons and myelin sheaths,thereby enhancing neuronal survival and mitigating injury-induced oxidative stress and *** actions collectively contribute to creating a favorable microenvironment for structural and functional nerve regeneration.A range of autophagyinducing drugs and interventions have demonstrated beneficial effects in alleviating peripheral neuropathy and promoting nerve regeneration in preclinical models of traumatic peripheral nerve *** review delves into the regulation of autophagy in cell types involved in peripheral nerve regeneration,summarizing the potential drugs and interventions that can be harnessed to promote this *** hope that our review will offer novel insights and perspectives on the exploitation of autophagy pathways in the treatment of peripheral nerve injuries and neuropathies.

Objective To observe the effect of growth differentiation factor-5 (GDF-5) on the growth and anabolic metabolism of articular chondrocytes. Methods The articular chondrocytes isolated from rats were treated with various concentrations of rmGDF-5, and the growth of chondrocytes measured by MTT assay, the cellular cartilage matrices formation detected sulfated glycosaminoglycan by Alcian blue staining and type Ⅱcollagen by RT-PCR. Results After 7 days culture, MTT assay showed that GDF-5 enhanced the growth of chondrocytes in a dose-dependent manner, RT-PCR showed that GDF-5 clearly induced the synthesis of type Ⅱ collagen because of the col2a1 mRNA band more and more strong in a dose-dependent. Chondrocytes were cultured with GDF-5 for 14 days, the intensity of Alcian blue staining was greatly enhanced, especially, at a high concentration of 1000ng/mL, and GDF-5 enhanced the accumulation of the Alcian blue-stainable material in a concentration-dependent manner and in a does-dependent manner. Conclusion GDF-5 enhanced the growth of mature articular chondrocytes, and stimulated the cellular cartilage matrices formation in mono-layer culture.

Objective To discuss the genetic relationship between Chaoxian and the other populations with STR polymorphisms, and study the origination of Chaoxian with historical data, analyze the difference among them. Methods Samples were obtained from 91 unrelated individuals of Jilin province. PCR amplification was performed using PE9700, the results were analyzed with the genescan software and then using the Genetic Analyzer ABI377 and Genotyper2.5. Tests for Hardy Weinberg equilibrium (HWE) and the clustering map were carried out using SPSS11.5. The DA (genetic distance) according to Nei's was calculated and a phylogenetic tree based on the neighbor-joining method using Mega2 software package was constructed. Results 81 alleles and 196 genotypes were observed, with the corresponding frequencies ranging from 0.0055 to 0.4615 and 0.0110 to 0.2747, the observed and expected of genotypes were evaluated using χ 2-test and all of the loci were in accordance with Hardy Weinberg equilibrium (P> 0.05). Chaoxian was clustered with the other Chinese ethnic groups, then the American Black and the White; it was clustered with the Baishan Han, the Beijing Han of China first, then the South Korean. Conclusion The differentiation among races is larger than that among minorities, and, the differentiation among minorities is related to and is consistent with their geographic location.
