Zinc homeostasis plays an essential role in maintaining immune function and is tightly regulated by zinc transporters. We previously reported that the zinc transporter SLC39A10, located in the cell membrane, critically regulates the susceptibility of macrophages to inflammatory stimuli; however, the functional role of SLC39A10 in T cells is currently unknown. Here, we identified two SNPs in SLC39A10that are associated with inflammatory bowel disease(IBD). We then generated transgenic mice with T cell-specific deletion of Slc39a10(cKO) and found that its loss not only protects against disease progression in IBD and experimental autoimmune encephalomyelitis(EAE),but also induces massive apoptosis via a p53/p21-and Bcl2-independent process. Mechanistically, we show that Slc39a10 serves as a key zinc importer upon activation of T cell receptor to safeguard DNA replication. Together, these findings provide new mechanistic insights and potential targets for the development of new therapeutic strategies for the treatment and/or prevention of T cell-mediated autoimmune diseases.

Robotic-assisted surgery for pediatric choledochal cyst: Case report and literature review

Our paper describes the key surgical points of pediatric choledochocystectomy performed completely by Da Vinci robotic system. A choledochocystectomy was safely carried out for a girl at our hospital, and without any complication. Then systematic literature review was done to discuss the methods of intestine surgery and intestinal anastomosis, the use of 3 rd robotic arm, the surgical safety and advantages comparing open and laparoscopic surgery. We systematically reviewed choledochocystectomy for children performed by robotic surgery. We included a total of eight domestic and foreign reports and included a total of 86 patients, whose average age was 6.3(0.3-15.9) years; the maleto-female ratio was 1:3.5(19:67). Seven patients experienced conversion to open surgery, and the surgery success rate was 91.9%(79/86). The average total operation time was 426(180-520) min, the operation time on the machine was 302(120-418) min, 11 cases used the number 3 arm, and the remaining mainly used the hitch-stitch technique to suspend the stomach wall and liver. Forty-seven patients underwent pull-through intestine and intestinal anastomosis, and 39 patients underwent complete robotic intestine surgery and intestinal anastomosis. The hospitalization time of roboticassisted choledochocystectomy was 8.8 d. Eight patientshad biliary fistula and were all cured by conservative treatment and continuous observation. One patient had anastomotic stenosis, and one patient had wound dehiscence, both cured by surgery. Choledochocystectomy for children performed by completely robotic surgery and Roux-en-Y hepaticojejunostomy is safe and feasible. The initial experience shows that this surgical approach has a clearer field than the traditional endoscopy, and its operation is more flexible, the surgery is more accurate, and the injury is smaller. With the advancement of technology and the accumulation of surgeons' experience, robotic surgery may become a new trend in this surgical procedure.

The gut microbiota and the associated metabolism play a pivotal role in maintaining human health,yet their assessment is challenging due to their inherent diversity,variety,and ***,we developed a multichannel sensor array based on a single fluorescent probe,allowing for rapid and robust profiling gut microbiota and their key metabolites including cysteine(Cys),glutathione(GSH),and homocysteine(Hcy).The assay leverages the single fluorescent probe that is with multiple binding sites and the cross-reactive sensing principle to specifically recognize different biological *** analyzing the pattern of biological thiols,the assay is capable of rapidly identifying six gut-derived bacteria including probiotics,neutral,and pathogenic strains based on fluorescent fingerprints within 5 min,and also discriminating bacteria and their mixtures with different *** the assay that enables the simultaneous measurement of multiple gut-derived bacteria and their metabolites,the designed array achieved an accuracy of 0.99 when discriminating colorectal cancer(CRC)patient feces samples from healthy ***,the as-prepared sensor array can also be used to identify various stages of *** simplicity,rapidness,and cost-effectiveness of our approach render it a robust platform for the analysis of gut microbiota.

Objective: To evaluate the clinical efficacy and safety of Yangxue Qingnao Pills(YXQNP) combined with amlodipine in treating patients with grade 1 hypertension. Methods: This is a multicenter, randomized, double-blind, and placebo-controlled study. Adult patients with grade 1 hypertension of blood deficiency and Gan(Liver)-yang hyperactivity syndrome were randomly divided into the treatment or the control groups at a 1:1 ratio. The treatment group received YXQNP and amlodipine besylate, while the control group received YXQNP's placebo and amlodipine besylate. The treatment duration lasted for 180 days. Outcomes assessed included changes in blood pressure, Chinese medicine(CM) syndrome scores, symptoms and target organ functions before and after treatment in both groups. Additionally, adverse events, such as nausea, vomiting, rash, itching, and diarrhea, were recorded in both groups. Results: A total of 662 subjects were enrolled, of whom 608(91.8%) completed the trial(306 in the treatment and 302 in the control groups). After 180 days of treatment, the standard deviations and coefficients of variation of systolic and diastolic blood pressure levels were lower in the treatment group compared with the control group. The improvement rates of dizziness, headache, insomnia, and waist soreness were significantly higher in the treatment group compared with the control group(Pmin-to-creatinine ratio were improved in both groups, with no statistically significant differences(P>0.05). OBJECTIVE: To evaluate the efficacy and safety of Yunpiqiangshen gel（运脾强肾浸膏） which composed of eleven herbs on quality of life（QOL） improvement. METHODS: We enrolled 180 patients lasted from January 2020 to December 2021. Seventy-eight patients received standards of care（control group） and 76 patients received Yunpiqiangshen gel [Traditional Chinese Medicine,（TCM） group] for 6 months in statistical analysis. The primary outcome assessed using Kidney Disease Quality of Life Short Form Questionnaire version 1.3(KDQOL-SFTM1.3), including Short Form Health Survey（SF-36） and Kidney Disease Targeted Areas（KDTA） scores. The second outcomes included the TCM syndrome score, Fatigue Assessment Instrument（FAI） score, Modified Quantitative Subjective Global Assessment（MQSGA） score, anthropometric indicators, and blood chemical testing. Adverse events, including decompensated gastrointestinal symptoms, were evaluated. RESULTS: The results showed that the mean change of SF-36（75 ± 13 vs 64 ± 16, F = 6.070, P = 0.015） and KDTA（76 ± 7 vs 70 ± 9, F = 4.118, P = 0.044） scores in the TCM group had an improvement（increase） from baseline, as compared with the control group. Yunpiqiangshen gel also resulted in a significant improvement in almost dimensions of QOL. At the end of follow-up, the imputed percentage of patients in the response rate of TCM syndrome was greater in the TCM group than in the control group（76.32% vs 20.51%, χ2= 48.02, P = 0.000）. The fatigue, soreness of waist, anorexia, abdominal distension, loose stool, and constipation were alleviated after Yunpiqiangshen gel therapy. The FAI（98.58 ± 25.08 vs 131.21 ± 31.85, F = 8.745, P = 0.004） and MQSGA（10.13 ± 2.84 vs 12.83 ± 3.85, F = 11.396, P = 0.001） scores in the TCM group had an improvement（reduce） from baseline compared with the control group.
