The lactylation index predicts the immune microenvironment and prognosis of pan-cancer patients

Background:Protein lactylation is a new way for the“metabolic waste”lactic acid to perform novel ***,our understanding of the contribution of protein lactylation to both tumor progression and therapeutic interventions remains *** construction of a scoring system for lactylation to predict the prognosis of pancancer patients and to evaluate the tumor immune microenvironment(TIME)would improve our understanding of the clinical significance of ***:Consensus clustering analysis of lactylation-related genes was used to cluster 177 pancreatic adenocarcinoma(PAAD)***,a scoring system was developed using the least absolute shrinkage and selection operator(LASSO)*** validation and external validation were both conducted to assess and confirm the predictive accuracy of the scoring ***,leucine rich repeat containing 1(LRRC1),a newly discovered lactylation-related gene,was analyzed in PAAD in ***:Utilizing the profiles of 332 lactylation-related genes,a total of 177 patients with PAAD were segregated into two distinct ***^(high) patients had a poorer prognosis than LacCluster^(low) *** the differential analysis between the LacCluster^(high) and LacCluster^(low) groups,we identified additional genes associated with *** genes were then integrated to construct the LacCluster-enhanced system,which enabled more accurate prognosis prediction for patients with ***,a lactylation index containing three genes(LacI-3)was constructed using LASSO *** was done to enhance the usability of the LacCluster-enhanced system in the *** to those in the LacI-3^(high) subgroup,patients in the LacI-3^(low) subgroup exhibited increased expression of immune checkpoint-related genes,more immune cell infiltration,lower tumor mutation burdens,and better prognoses,indicating a“hot tumor”***,knocking down the expression of LRRC1,

Allogeneic red blood cell(RBC)transfusion is commonly performed in medical practice because of its efficacy and low-risk ***,pre-transfusion tests are susceptible to monoclonal antibody(mAb)interference.1 Currently,mAb therapies are being developed to treat many diseases,such as ***,certain mAbs,such as anti-CD38mAb and anti-CD47mAb,can bind to RBC membranes;this binding interferes with pre-transfusion tests.2 CD47 has gained considerable attention in recent years because of its potential as a therapeutic target for hematologic malignancies and solid tumors.3 The binding of anti-CD47mAb to RBCs may lead to false-positive results in pan-agglutination tests and cause delays and risks in establishing compatible RBCs for transfusion.

Therapeutic targeting FOxO3A(a forkhead transcription factor)represents a prom-ising strategy to suppress acute myeloid leukemia(AML).However,the effective inhibitors that target FOXO3A are lacking and the adaptive response signaling weakens the cytotoxic effect of FOxO3A depletion on AML ***,we show that FOxO3A deficiency induces a compensa-tory response involved in the reactive activation of mTOR that leads to signaling rebound and adaptive *** metabolism acts downstream of mTOR to provoke activa-tion of JNK/c-JUN via reactive oxygen species(ROS).At the molecular level,FOXO3A directly binds to the promoter of G protein gamma subunit 7(GNG7)and preserves its expression,while GNG7 interacts with mTOR and restricts phosphorylated activation of ***,combinatorial inhibition of FOXO3A and mTOR show a synergistic cytotoxic effect on AML cells and prolongs survival in a mouse model of *** a structure-based virtual screening,we report one potent small-molecule FOxO3A inhibitor(Gardenoside)that exhibits a strong effect of anti-FOXO3A DNA *** synergizes with rapamycin to substantially reduce tumor burden and extend survival in AML patient-derived xenograft *** results demonstrate that mTOR can mediate adaptive resistance to FOxO3A inhibition and validate a combinatorial approach for treating AML.

Stroke Risk Assessment Decision-Making Using a Machine Learning Model:Logistic-AdaBoost

Stroke is a chronic cerebrovascular disease that carries a high *** risk assessment is of great significance in preventing,reversing and reducing the spread and the health hazards caused by *** to objectively predict and identify strokes,this paper proposes a new stroke risk assessment decision-making model named Logistic-AdaBoost(Logistic-AB)based on machine ***,the categorical boosting(CatBoost)method is used to perform feature selection for all features of stroke,and 8 main features are selected to form a new index evaluation system to predict the risk of ***,the borderline synthetic minority oversampling technique(SMOTE)algorithm is applied to transform the unbalanced stroke dataset into a balanced ***,the stroke risk assessment decision-makingmodel Logistic-AB is constructed,and the overall prediction performance of this new model is evaluated by comparing it with ten other similar *** comparison results show that the new model proposed in this paper performs better than the two single algorithms(logistic regression and AdaBoost)on the four indicators of recall,precision,F1 score,and accuracy,and the overall performance of the proposed model is better than that of common machine learning *** Logistic-AB model presented in this paper can more accurately predict patients’stroke risk.

Beamforming design for covert broadcast communication with hidden adversary

This paper explores covert broadcast communication in a challenging situation in which the transmitter, Alice, faces uncertainty regarding the position of the passive adversary, Willie. In this situation,controlling the signal exposure becomes difficult. Specifically, this paper focuses on the cases where Alice can delimit the suspicious areas where Willie may be located. The suspicious areas can have arbitrary number and shapes. Under the Rician fading model, the analytical expression of Willie's detection performance is derived. Then, the transmit power, the beamforming direction, and the number of channel uses are jointly designed to minimize the optimal Willie's detection performance among all the possible positions of Willie, on the premise of satisfying receivers' quality-of-service requirements. To tackle the formulated knotty problem with an infinite number of irregular and discontinuous Willie's possible positions, the continuous nature of the antenna pattern is exploited to convert the original problem into the one with a finite number of possible positions, by sampling the suspicious areas. Then, different algorithms are developed to address the resultant non-convex optimization problem for single-antenna Willie and multi-antenna Willie, respectively. Numerical results demonstrate significant performance gains of the proposed scheme over the widely-adopted maximum ratio transmission scheme, which indicates that simply adjusting the main lobe towards the receiver is indeed far away from optimum and shows the importance of leveraging Alice's limited knowledge of Willie's position.

Plasmon-assisted mode selection lasing in a lanthanide-based microcavity

Lanthanide-based microlasers have attracted considerable attention owing to their large anti-Stokes shifts, multiple emission bands, and narrow linewidths. Various applications of microlasers, such as optical communication, optical storage, and polarization imaging, require selecting the appropriate laser polarization mode and remote control of the laser properties. Here, we propose a unique plasmon-assisted method for the mode selection and remote control of microlasing using a lanthanide-based microcavity coupled with surface plasmon polaritons(SPPs) that propagate on a silver microplate. With this method, the transverse electrical(TE) mode of microlasers can be easily separated from the transverse magnetic(TM) mode. Because the SPPs excited on the silver microplate only support TM mode propagation, the reserved TE mode is resonance-enhanced in the microcavity and amplified by the local electromagnetic field. Meanwhile, lasingmode splitting can be observed under the near-field excitation of SPPs due to the coherent coupling between the microcavity and mirror microcavity modes. Benefiting from the long-distance propagation characteristics of tens of micrometers of SPPs on a silver microplate, remote excitation and control of upconversion microlasing can also be realized. These plasmon-assisted polarization mode-optional and remote-controllable upconversion microlasers have promising prospects in on-chip optoelectronic devices, encrypted optical information transmission, and high-precision sensors.

The photosynthesis of hydrogen peroxide(H_(2)O_(2))through the selective 2e^(-)oxygen reduction reaction(ORR)from O_(2)and H_(2)O stands out as an environmentally sustainable and cost-effective method for generating this essential ***,the widespread application of most photocatalysts is impeded by their reliance on sacrificial *** this context,we present a noteworthy advancement in the form of a step-scheme(S-scheme)heterojunction involving a donor-acceptor(D-A)-conjugated polymer and manganese cadmium sulfide(BTz@MCS).This innovative configuration enables efficient photocatalytic H_(2)O_(2)production within a non-sacrificial system,showcasing an impressive H_(2)O_(2)yield of 5,368μmol g^(-1)h^(-1)and an apparent quantum yield of 4.5%at 420 *** insights gained from in-situ irradiated X-ray photoelectron spectroscopy(ISIXPS),in-situ diffuse reflectance infrared Fourier transformations spectroscopy(DRIFTS),and density function calculations(DFT),it is revealed that the sp^(2)C between thiazole-thiazole rings at the BTz and the concentrated holes at the MCS function as spatially separated redox *** centers catalyze the selective 2e^(-)ORR and 4e^(-)water oxidation reaction(WOR)*** O_(2)generated from the WOR is subsequently utilized by the ORR cycle,enabling the overall photosynthesis of H_(2)O_(2)with accelerated total reaction ***,the high photocatalytic performance of the developed catalyst is attributed to well-designed S-scheme heterojunctions with tuned band *** spatially separated redox centers and effective transfer of photogenerated charge carriers facilitated by the internal electric field significantly enhance reaction *** study introduces a promising avenue for photocatalytic H_(2)O_(2)production without sacrificial agents,providing valuable insights into the underlying mechanisms in S-scheme heterojunctions.
