Receptor recognition and subsequent membrane fusion are essential for the establishment of successful infection by *** these steps can cure *** we have identified and characterized a potent human monoclonal antibody,HB27,that blocks SARS-CoV-2 attachment to its cellular receptor at sub-nM ***,HB27 can also prevent SARS-CoV-2 membrane ***,a single dose of HB27 conferred effective protection against SARS-CoV-2 in two established mouse *** macaques showed no obvious adverse events when administrated with10 times the effective dose of ***-EM studies on complex of SARS-CoV-2 trimeric S with HB27 Fab reveal that three Fab fragments work synergistically to occlude SARS-CoV-2 from binding to the ACE2 *** of the antibody also restrains any further conformational changes of the receptor binding domain,possibly interfering with progression from the prefusion to the postfusion *** results suggest that HB27 is a promising candidate for immuno-therapies against COVID-19.

Plant secondary metabolites play critical roles in plant-environment interactions. They are synthesized in different organs or tissues at particular developmental stages, and in response to various environmental stimuli, both biotic and abiotic. Accordingly, corresponding genes are regulated at the transcriptional level by multiple transcription factors. Several families of transcription factors have been identified to participate in controlling the biosynthesis and accumulation of secondary metabolites. These regulators integrate internal （often developmental） and external signals, bind to corresponding cis-elements -- which are often in the promoter regions -- to activate or repress the expression of enzyme-coding genes, and some of them interact with other transcription factors to form a complex. In this review, we summarize recent research in these areas, with an emphasis on newly-identified transcription factors and their functions in metabolism regulation.

Coffee arabica *** an economical crop cultivated in many countries including *** present study evaluated the phytochemical contents of the methanolic extracts,prepared by maceration,of six Yemeni green coffee beans varieties namely Esmaeli,Hamadi,Harazi,Mattari,Odaini and ***(phosphomolybdenum,antiradical,reducing power and ferrous chelating),and enzyme inhibition activity of the extracts against acetylcholinesterase butyrylcholinesterase,tyrosinase,α-glucosidase,andα-amylase were also *** of total phenols revealed that all green beans varieties were richer in their total polyphenolic(63.77-110.98 mg gallic acid equivalent per g of extract)content than their flavonoids one(2.86-5.57 mg rutin equivalent per g of extract).HPLC-MS/MS analysis of 30 selected bioactive compounds showed that all varieties had the same types of phytoconstituents with differences in their relative ***(1613.89-2466.38μg/g)followed by 5-caffeoylquinic acid(1017.63-1313.39μg/g)were the dominant compounds in all varieties with caffeine more abundant in Esmaeli variety while 5-caffeoylquinic acid in Odaini *** varieties displayed a pronounced antioxidant property in all the in vitro assays with Odaini variety significantly(p<0.05)exerted the highest anti-DPPH radicals(253.96 mg Trolox equivalent(TE)/g),metal chelating(19.73 mg Disodium edetate equivalents/g),Cu^(2+)(441.11 mg TE/g)and Fe3+(221.04 mg TE/g)reducing ***,the majority of green coffee beans varieties showed comparable enzyme inhibition property with highest activity recorded against tyrosinase(39.35-46.96 mg kojic acid equivalents/g)and acetylcholinesterase(1.80-2.17 mg galanthamine equivalents/g)*** conclusion,all Yemeni green coffee beans varieties have proven to be rich source of biochemicals with beneficial impact on human health and could be of significant applications in food,pharmaceutical and cosmetics industry.

Induction and transmission of wheat-Haynaldia villosa chromosomal translocations

In order to develop more wheat-Haynaldia villosa translocations involving different chromosomes and chromosome segments of H. villosa, T. durum-H, villosa amphiploid was irradiated with ^60Co γ-rays at doses of 800, 1,200, and 1,600 rad. Pollen collected from the spikes 1, 2, and 3 days after irradiation were transferred to emasculated spikes of the common wheat cv. ‘Chinese Spring＇. Genomic in situ hybridization was used to identify wheat-H, villosa chromosome translocations in the M1 generation. Transmission of the identified translocation chromosomes was analyzed in the BC1, BC2, and BC3 generations. The results indicated that all three irradiation doses were highly efficient for inducing wheat-alien translocations without affecting the viability of the M1 seeds. Within the range of 800-1,600 rad, both the efficiency of translocation induction and the frequency of interstitial chromosome breakage-fusion increased as the irradiation dosage increased. A higher translocation induction frequency was observed using pollen collected from the spikes 1 day after irradiation over that of 2 or 3 days after irradiation. More than 70% of the translocations detected in the M1 generation were transmitted to the BC1 through the female gametes. All translocations recovered in the BC1 generation were recovered in the following BC2, and BC3 generations. The transmission ability of different translocation types in different genetic backgrounds showed an order of ‘whole-arm translocation 〉 small alien segment translocation 〉 large alien segment translocation＇, through either male or female gametes, In general, the transmission ability through the female gametes was higher than that through the male gametes. By this approach, 14 translocation lines that involved different H. villosa chromosomes have been identified in the BC3 using EST-STS markers, and eight of them were homozygous.

Denitrification Rate and Controlling Factors for Accumulated Nitrate in the Deep Subsoil of Intensive Farmlands: A Case Study in the North China Plain

Denitrification in subsoil(to a depth of 12 m) is an important mechanism to reduce nitrate(NO3^-) leaching into ***, regulating mechanisms of subsoil denitrification, especially those in the deep subsoil beneath the crop root zone, have not been well documented. In this study, soil columns of 0–12 m depth were collected from intensively farmed fields in the North China Plain. The fields had received long-term nitrogen(N) fertilizer inputs at 0(N0), 200(N200) and 600(N600) kg N ha^-1 year^-1. Main soil properties related to denitrification, i.e., soil water content, NO3^-, dissolved organic carbon(DOC), soil organic carbon(SOC),pH, denitrifying enzyme activity(DEA), and anaerobic denitrification rate(ADR), were determined. Statistical comparisons among the treatments were performed. The results showed that NO3^- was more heavily accumulated in the entire soil profile of the N600 treatment, compared to the N0 and N200 treatments. The SOC, DOC, and ADR decreased with increasing soil depth in all treatments,whereas considerable DEA was observed throughout the subsoil. The long-term fertilizer rates affected ADR only in the upper 4 m soil layers. The ADRs in the N200 and N600 treatments were significantly correlated with DOC. Multiple regression analysis indicated that DOC rather than DEA was the key factor regulating denitrification beneath the root zone. Additional research is required to determine if carbon addition into subsoil can be a promising approach to enhance NO3^- denitrification in the subsoil and consequently to mitigate groundwater NO3^- contamination in the intensive farmlands.

Comparison of Supervised Clustering Methods for the Analysis of DNA Microarray Expression Data

Several typical supervised clustering methods such as Gaussian mixture model-based supervised clustering （GMM）, k- nearest-neighbor （KNN）, binary support vector machines （SVMs） and multiclass support vector machines （MC-SVMs） were employed to classify the computer simulation data and two real microarray expression datasets. False positive, false negative, true positive, true negative, clustering accuracy and Matthews＇ correlation coefficient （MCC） were compared among these methods. The results are as follows：（1） In classifying thousands of gene expression data, the performances of two GMM methods have the maximal clustering accuracy and the least overall FP＋FN error numbers on the basis of the assumption that the whole set of microarray data are a finite mixture of multivariate Gaussian distributions. Furthermore, when the number of training sample is very small, the clustering accuracy of GMM-Ⅱ method has superiority over GMM- Ⅰ method. （2） In general, the superior classification performance of the MC-SVMs are more robust and more practical, which are less sensitive to the curse of dimensionality, and not only next to GMM method in clustering accuracy to thousands of gene expression data, but also more robust to a small number of high-dimensional gene expression samples than other techniques. （3） Of the MC-SVMs, OVO and DAGSVM perform better on the large sample sizes, whereas five MC-SVMs methods have very similar performance on moderate sample sizes. In other cases, OVR, WW and CS yield better results when sample sizes are small. So, it is recommended that at least two candidate methods, choosing on the basis of the real data features and experimental conditions, should be performed and compared to obtain better clustering result.

Genetic variation and selection of introduced provenances of Siberian Pine(Pinus sibirica) in frigid regions of the Greater Xing'an Range,Northeast China

Siberian Pine （Pinus sibirica） is an ecologically and eco-nomically important species in pristine forests throughout northern Rus-sia. Four provenances of P. sibirica were introduced from Mongolia and Russia to the Greater Xing’an Range （the Daxing’anling）, northeast China in 1993. The aim of this research was to study genetic variation and selection of the introduced four Pinus sibirica provenances. Heights （H）, basal diameters （BD）, survival rates （SR） and crown lengths （CL） of different families were measured as primary outcomes in different growth years. Results of data analyses demonstrated high coefficients of phenotypic variation （PCV） and heritability （H2） for H, BD and CL at 18 years after introduction. PCV and H2 increased with age. Correlations of＆amp;nbsp;growth traits between any two growth years were all significantly positive, but the correlation coefficient was smaller when the growth year interval was larger. Correlations between H and the original environment factors decreased gradually, indicating that with long-term subsistence in the new environment, the influence of the source environment declined. Colligation of multiple traits to estimate provenances showed that Novosibirsk, Tomsk, and Altai Mountains had higher survival rates and biomass, and proved more suitable for introduction and plantation in the Greater Xing’an Range in China.
