Assessing decontamination practices at a medical microbiology research laboratory

To our knowledge,this is the first study to conduct an objective assessment of the routine decontamination practices at a medical microbiology research laboratory(MRL)a year after a biosafety training was provided to all laboratory *** March 28th and June 28th,2021,unobtrusive observations were carried out to identify-three high-touch surfaces at the MRL during different working *** was used to evaluate the effectiveness of the disinfectant used in the *** three high-touch surfaces were sampled before and after decontamination with 200 ppm of 5%sodium hypochlorite(household bleach)to quantify the microbial load and identify the types of organisms residing on the laboratory surfaces.A higher concentration(500 ppm)of 5%sodium hypochlorite was employed after refresher training was provided to housekeeping staff,and resampling of the three surfaces was carried out during a 4-week follow-up period using the same *** three high-touch surfaces identified were the two sides of the workbench(22%–24%)and the front surface of one incubator(14%).Anthracoid bacilli and Staphylococcus aureus were the most commonly found organisms on laboratory surfaces preintervention(100%and 89%,respectively)and post-intervention(56%and 44%,respectively).Other microorganisms detected included Salmonella spp.(27.7%),Proteus spp.(5.6%),Escherichia coli(5.6%),and Klebsiella spp.(33.3%).Employing a higher concentration(500 ppm)of sodium hypochlorite significantly(plaboratory,which will

Association networks are widely applied for the prediction of bacterial interactions in studies of human gut ***,the experimental validation of the predicted interactions is challenging due to the complexity of gut microbiomes and the limited number of cultivated *** this study,we addressed this challenge by integrating in vitro time series network(TSN)associations and cocultivation of TSN taxon *** samples were collected and used for cultivation and enrichment of gut microbiome on YCFA agar plates for 13 *** cells were harvested for DNA extraction and metagenomic sequencing.A total of 198 metagenome-assembled genomes(MAGs)were *** dynamics of bacteria growing on the YCFA agar were used to infer microbial association *** experimentally validate the interactions of taxon pairs in networks,we selected 24 and 19 bacterial strains from this study and from the previously established human gut microbial biobank,respectively,for pairwise *** co-culture experiments revealed that most of the interactions between taxa in networks were identified as neutralism(51.67%),followed by commensalism(21.67%),amensalism(18.33%),competition(5%)and exploitation(3.33%).Genome-centric analysis further revealed that the commensal gut bacteria(helpers and beneficiaries)might interact with each other via the exchanges of amino acids with high biosynthetic costs,short-chain fatty acids,and/or *** also validated 12 beneficiaries by adding 16 additives into the basic YCFA medium and found that the growth of 66.7%of these strains was significantly *** approach provides new insights into the gut microbiome complexity and microbial interactions in association *** work highlights that the positive relationships in gut microbial communities tend to be overestimated,and that amino acids,short-chain fatty acids,and vitamins are contributed to the positive relationships.

Toxic arsenic(As)and trace element selenium(Se)are transformed by microorganisms but their complex interactions in soil-plant systems have not been fully *** Asand Se-oxidizing bacterium,Agrobacterium sp.T3F4,was applied to a native seleniferous As-polluted soil to investigate As/Se uptake by the vegetable Brassica rapa *** As-Se interaction as mediated by strain *** Se content in the aboveground plants was significantly enhanced by 34.1%,but the As content was significantly decreased by 20.5% in the T3F4-inoculated pot culture compared to the control(P<0.05).Similar result was shown in treatment with additional 5 mg/kg of Se(IV)in *** addition,the As contents in roots were significantly decreased by more than 35% under T3F4 or Se(IV)treatments(P<0.05).Analysis of As-Se-bacterium interaction in a soil simulation experiment showed that the bioavailability of Se significantly increased and As was immobilized with the addition of the T3F4strain(P<0.05).Furthermore,an As/Se co-exposure hydroponic experiment demonstrated that As uptake and accumulation in plants was reduced by increasing Se(IV)*** 50% growth inhibition concentration(IC50)values for As in plants were increased about one-fold and two-fold under co-exposure with 5 and 10μmol/L Se(IV),*** conclusion,strain T3F4 improves Se uptake but decreases As uptake by plants via oxidation of As and Se,resulting in decrease of soil As bioavailability and As/Se competitive absorption by *** provides a potential bioremediation strategy for Se biofortification and As immobilization in As-polluted soil.

The purpose of this study was to characterize mulberry leaf instant tea(MLIT)powder prepared from the'Longsang No.1'(Morus abla *** 1)mulberry leaves in Heilongjiang Province(China)and assess its obesity-preventing/relieving effects.A total of 174 compounds including quercetin,chlorogenic acid,1-deoxyecomycin(1-DNJ)related to antihyperlipidemia effects were identified from the MLIT *** treatment reversed the Lee's index,fat coefficient,and serum biochemical parameters in both the obesity relieving and obesity preventing mice fed with high-fat *** the obesity relieving experiment,the relative abundance of Desulfovibrio in mouse feces decreased after both 0.5%and 1%MLIT *** obesity preventing experiments,mouse with different amount of MLIT treatments showed increased relative abundance of Akkermansia,Bifidobacterium and Lactobacillus,while Deferribacteres,Desulfobacterota *** beneficial bacteria in the intestinal tract of mice treated with MLIT *** study proved that MLIT had antihyperlipidemia potential via modulating intestinal microbiota in mice.
