Human salivary histatin 1(Hst1)exhibits a series of cell-activating properties,such as promoting cell spreading,migration,and metabolic *** recently have shown that fluorescently labeled Hst1(F-Hst1)targets and activates mitochondria,presenting an important molecular ***,its regulating signaling pathways remain to be *** investigated the influence of specific inhibitors of G protein-coupled receptors(GPCR),endocytosis pathways,extracellular signal-regulated kinases1/2(ERK1/2)signaling,p38 signaling,mitochondrial respiration and Na+/K+-ATPase activity on the uptake,mitochondria-targeting and-activating properties of *** performed a si RNA knockdown(KD)to assess the effect of Sigma-2 receptor(S2R)/Transmembrane Protein 97(TMEM97)—a recently identified target protein of *** also adopted live cell imaging to monitor the whole intracellular trafficking process of *** results showed that the inhibition of cellular respiration hindered the internalization of *** inhibitors of GPCR,ERK1/2,phagocytosis,and clathrin-mediated endocytosis(CME)as well as siRNA KD of S2R/TMEM97 significantly reduced the uptake,which was accompanied by the nullification of the promoting effect of F-Hst1 on cell metabolic *** the inhibitor of CME and KD of S2R/TMEM97 significantly compromised the mitochondria-targeting of *** further showed the intracellular trafficking and targeting process of F-Hst1,in which early endosome plays an important ***,phagocytosis,CME,GPCR,ERK signaling,and S2R/TMEM97 are involved in the internalization of Hst1,while only CME and S2R/TMEM97 are critical for its subcellular *** inhibition of either internalization or mitochondria-targeting of Hst1 could significantly compromise its mitochondria-activating property.

Advancements in computational technologies for residential energy management have become widespread due to increased energy usage and peak demand in *** switching of appliances during peak periods for energy reduction is one of such ***,this technique may constrict the consumer’s flexibility to utilize desired appliances to fulfil their *** study proposes a peak load control technique based on an end-use appliance prioritization and event detection algorithm.A noteworthy feature of the technique is the users’preferred appliance(UPA),which provides the occupants with the flexibility to use any electric load to fulfill their needs at any time whether peak demand control has been initiated or ***,to address the challenging issue around the uncertain and varying nature of the users’energy usage pattern,an empirical bottom-up approach is adopted to characterize the consumers’diverse end-use *** results obtained show good performance in terms of peak demand and energy consumption reduction,with 3%to 20%and at least 14.05%in demand reduction for time of use(ToU)periods and energy savings,*** offer a further perspective on demand side management,affording load users a new cost-saving energy usage pattern.

This study reports the morphological characteristics of anomalous variations in Global Navigation Satellite System Total Electron Content(GNSS-TEC) prior to the strong local earthquakes(EQ) that occurred during the period of *** have analyzed 20 earthquakes of magnitude M> 5.6.A statistical technique is implemented on the data of six GNSS stations located in Tashkent,Kitab,and Maidanak in Uzbekistan,and Islamabad,Multan,Quetta in *** results show continuous anomalous variations in TEC during 24 h befo re the occupancy of local *** is shown that the precursors before the occurrence of strong earthquakes,in particular of magnitude 5.7,7.7,7.5,7.8 and 7.3 are detected near Eastern Uzbekistan(26 May 2013),Southwestern Pakistan(24 September 2013),Hindukush region of Afghanistan(26 October 2015),and Central Nepal(25 April 2015) and(12 May 2015),*** ionospheric anomalies appearing before the strong earthquakes at six GNSS stations are registered in 14cases(70%) out of 20 selected *** is depicted that anomalies referred to as ionospheric precursors appeared about 1-7 days prior to the occurrence of strong earthquakes.

BACKGROUND:Infectious complications are common during the postoperative course of a liver transplant recipient. Malaria, however, is a rare complication in such a setting. METHOD:We report post-transplantation malaria causing elevation of liver enzymes in two recipients. RESULTS:Both patients who had undergone living donor liver transplantation showed elevated levels of liver enzymes and fever during the postoperative course. Investigations (including liver biopsy in one patient) were initially inconclusive in determining the cause of liver dysfunction. The diagnosis of malaria was established in both cases by peripheral blood smear. Liver function transiently worsened with antimalarial treatment but subsequently became normal. CONCLUSION:This report highlights the importance of excluding such uncommon causes of post-transplantation liver dysfunction, especially when either the recipient or the donor comes from a region endemic for malaria.

Acyl-CoA synthetase long chain family member 5(ACSL5),is a member of the acyl-CoA synthetases(ACSs)family that activates long chain fatty acids by catalyzing the synthesis of fatty *** dysregulation of ACSL5 has been reported in some cancers,such as glioma and colon ***,little is known about the role of ACSL5 in acute myeloid leukemia(AML).We found that the expression of ACSL5 was higher in bone marrow cells from AML patients compared with that from healthy ***5 level could serve as an independent prognostic predictor of the overall survival of AML *** AML cells,the ACSL5 knockdown inhibited cell growth both in vitro and in ***,the knockdown of ACSL5 suppressed the activation of the Wnt/β-catenin pathway by suppressing the palmitoylation modification of ***,triacsin c,a pan-ACS family inhibitor,inhibited cell growth and robustly induced cell apoptosis when combined with ABT-199,the FDA approved BCL-2 inhibitor for AML *** results indicate that ACSL5 is a potential prognosis marker for AML and a promising pharmacological target for the treatment of molecularly stratified AML.

目的　利用人类免疫缺陷病毒 (HIV)低变异的调节蛋白Rev与Rev反应区 (RRE)的高亲和性 ,尝试合成一种不受变异影响的特异性病毒表达检测结构。方法　采用分子克隆技术合成含有RRE和增强的绿荧光蛋白 (EGFP)的Rev依赖性荧光表达质粒 ,采用流式细胞仪检测新质粒的表达功能。结果　新合成的质粒pDM 12 8 EGFP (pE12 8)含氨苄青霉素受体 (AmpR)选择子、SV40 启动子、EGFP基因及HIV 1 RRE片段 ,BssSI酶切后产生 3 .6、1.9和 1.5kb片断 ,HindⅢ酶切后产生 3 470、2 72 5和 879bp 3个片断 ,PCR扩增提示EGFP基因完整插入。pE12 8在Hela细胞中显示Rev依赖性表达 ,pRev不存在时 ,几乎没有EGFP表达 ;pRev存在时 ,EGFP显著表达 (P <0 .0 1)。绿荧光Hela细胞百分比随pRev比例的增加而渐次增加。结论　pE12 8能特异、敏感、快速和直观地检测HIV的表达 ,对反映HIV的感染性及病情的转归及对获得性免疫缺陷综合征 (AIDS)

Acute kidney injury in patients with chronic liver disease

Acute kidney injury(AKI) is a frequent clinical event in patients with liver disease, compounding their prognosis. Furthermore, it is likely that the occurrence of AKI has a detrimental impact on the subsequent renal function and the long-term survival of these patients. Recently, some authors advocated the use of new diagnostic criteria for detecting acute kidney injury in patients with cirrhosis. These criteria are based on the rapidity and extent of the creatinine increase comparing to the basal creatinine and also on the kinetics of diuresis decrease. Although their validity in this population requires further studies to be clearly established, these new criteria could have two advantages:(1) to allow earlier diagnosis of AKI and, thus, hepatorenal syndromefor which earlier intervention could improve patients' survival; and(2) to promote more intensive monitoring of renal function in these patients with high risk of AKI. Finally, recent practice guidelines about the prevention and treatment of general AKI have been published which should be useful in optimising the management of AKI in cirrhotic patients.

Understanding hydrological responses to rising levels of greenhouse gases are essential for climate and impact *** is,however,often limited by a lack of long record of observational data to provide a basis for understanding the longterm behavior of the climate *** reconstructed data and(global climate and hydrological)model simulations will help us to better understand the variability of climate and hydrology over timescales ranging from decades to *** this study,we proposed an integrated approach to study flood variability in the upper reach of the Yangtze River over the last millennium to the end of the 21st *** accomplish this,we first drove hydrological models using the precipitation and temperature from four Global Climate Models(GCM),BCC-CSM1.1,MIROC,MRI-CGCM3,and CCSM4,to simulate daily discharge for the upper reach of the Yangtze River during the period of the last millennium(850–1849),historical period(1850–2005),and a future period(2006–2099).Then,we evaluated whether the modeled precipitation,temperature,and extreme discharge had statistical properties similar to those shown in the documented dry-wet periods,temperature anomalies,and paleoflood ***,we explored the extreme discharge variability using model *** results indicate that:(1)The MIROC-ESM model,differing from the other three GCM models,revealed positive temperature changes from the warm period(Medieval Climate Anomaly;MCA)to the cold period(Little Ice Age;LIA),while the temperature variability of the other models was similar to the records.(2)The BCC-CSM1.1 model performed better than the others regarding correlations between modeled precipitation and documented dry-wet periods.(3)Over most of the subbasins in the upper Yangtze River,the magnitude of extreme discharge in the BCC-CSM1.1 model results showed that there was a decrease from the MCA to the LIA period and an increase in the historical period relative to the cold period,whil
