Analyzing polysorbate 20(PS20)co.position and the impact of each co.ponent on stability and safety is crucial due to formulation variations and individual *** similar structures and polarities of PS20 co.ponents make accurate separation,identification,and quantification *** this work,a high-resolution quantitative method was developed using single-dimensional high-performance liquid chromatography(HPLC)with charged aerosol detection(CAD)to separate 18 key co.ponents with multiple *** separated co.ponents were characterized by ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UHPLC-Q-TOF-MS)with an identical gradient as the HPLC-CAD *** polysorbate co.pound database and library were expanded over 7-time co.pared to the co.mercial *** method investigated differences in PS20 samples from various origins and grades for different dosage forms to evaluate the co.position-process ***-Q-TOF-MS identified 1329 to 1511 co.pounds in 4 batches of PS20 from different *** method observed the impact of 4 degradation co.ditions on peak co.ponents,identifying stable co.ponents and their tendencies to ***-CAD and UHPLC-Q-TOF-MS results provided insights into fingerprint differences,distinguishing quasi products.

[Objectives]To establish an LC-MS method for the determination of dibromoacetic acid in Cefathiamidine and Cefathiamidine for Injection.[Methods]Shiseido PC HILIC co.umn(2.0 mm ×100 mm,3μ.m)was *** mobile phase was 0.005 Mammonium formate and the mobile phase was acetonitrile with gradient *** flow rate was 0.3-0.8mL/min.[Results]The limit of detection was 0.2500 ng/mL and the limit of quantification was 0.500 ng/mL,which were 2.5%and 5.0%of the impurity limits,*** reco.-ery rate was 103.85%.[co.clusions]This method improved the detection of dibromoacetic acid impurities in Cefathiamidine,and has that advantage of good specificity,low limit of detection and limit of quantification,high sensitivity,high accuracy,interference resistance,can meet the detection requirements of Cefathiamidine,and is suitable for the quality co.trol of Cefathiamidine.

Insufficient therapeutic strategies for acute kidney injury(AKI)necessitate precision therapy targeting its *** study reveals the new mechanism of the marine-derived anti-AKI agent,piericidin glyco.ide S14,targeting peroxiredoxin 1(PRDX1).By binding to Cys83 of PRDX1 and augmenting its peroxidase activity,S14 alleviates kidney injury efficiently in Prdx1-overexpression(Prdx1-OE)***,S14 also increases PRDX1 nuclear translocation and directly activates the Nrf2/HO-1/NQO1 pathway to inhibit ROS *** to the limited druggability of S14 with low bioavailability(2.6%)and poor renal distribution,a pH-sensitive kidney-targeting dodecanaminechitosan nanoparticle system is co.structed to load S14 for precise treatment of AKI.L-Serine co.jugation to chitosan imparts specificity to kidney injury molecule-1(Kim-1)-overexpressed *** developed S14-nanodrug exhibits higher therapeutic efficiency by improving the in vivo behavior of S14 *** encapsulation with micelles,the AUC_(0-t),half-life time,and renal distribution of S14 increase 2.5-,1.8-,and 3.1-fold,*** main factors co.tributing to the improved druggability of S14 nanodrugs include the lower metabolic elimination rate and UDPglyco.yltransferase(UGT)-mediated *** summary,this study identifies a new therapeutic target for the marine-derived anti-AKI agent while enhancing its ADME properties and druggability through nanotechnology,thereby driving advancements in marine drug development for AKI.

Four new diarylheptanoids and two new terpenoids from the fruits of Alpinia oxyphylla and their anti-inflammatory activities

Four previously unreported diarylheptanoids（1a/1b-2a/2b）, one undescribed sesquiterpenoid（8）, one new diterpenoid（12）, and twelve known analogs were isolated from the fruits of Alpinia oxyphylla. The structural elucidation of these co.pounds was achieved through a co.prehensive analysis of spectrosco.ic data, single-crystal X-ray diffraction, electronic circular dichroism（ECD）,and modified Mosher's method. Enantiomeric mixtures（1a/1b, 2a/2b, 3a/3b, 4a/4b, and 5a/5b） were separated on a chiral co.umn using acetonitrile-water mixtures as eluents. Among them, co.pounds 3a/3b and 4a/4b were isolated as optically pure enantiomers in the initial chiral separation. Furthermore, most of the isolates were evaluated for their inhibitory effects against the production of nitric oxide（NO） and interleukin-6（IL-6） in lipopolysaccharide（LPS）-induced RAW264.7 macrophages. Interestingly, 2 and 4 showed significant inhibitory activities against NO production with IC50 values of 33.65 and 9.88 μmol·L-1(hydroco.tisone: IC5034.26 μmol·L-1),respectively. Additionally, they also partially reduced the secretion of IL-6.

Pharmacy is a co.plex process involving many variables. Among them, various proteins produced in the biological reaction process and protein co.plexes of them have an important influence on the co.trol of pharmaceutical process. In addition, detection of protein co.plexes is also the most essential and fundamental problem in most Bionic manufacturing industry. However, most existing clustering technical for protein co.plexes detection focus primarily on the local information of the protein-protein interaction networks(PPI), which leads to the global distribution between proteins is lost and some protein co.plexes are further missed. Therefore, a method namely spectral clustering with improved co.nectivity kernel(SPICK) is proposed to detect protein co.plexes based on both local and global information of the protein-protein interaction networks. In SPICK, the improved co.nectivity kernel co.siders both Euclidean distance and co.nectivity kernel to calculate the affinity matrix, which co.bines local information with global distribution of PPI effectively. Experimental results show that our method performs better in co.parison to existing protein co.plexes techniques.

Effectiveness of Acupoint Application in Patients with Pharyngeal Pain: Evidence from CHUNBO, A Prospective Real-World Study

Objective: To assess the outco.es after acupoint application in patients with pharyngeal pain in a real-world settings, and analyze the characteristics of effective population and prescription characteristics of acupoint application. Methods: Based on CHUNBO platform, patients with pharyngeal pain who were candidates for acupoint application on the basis of physician-evaluation, were enrolled in a nationwide, prospective, 69-week multicenter observational study from August 2020 to February 2022. Propensity sco.e matching(PSM) was used to match the co.founding factors and the association rules were used to analyze the characteristics of effective population and prescription characteristics of acupoint application. Outco.e assessments included the disappearance rate of pharyngeal pain(within 3, 7, and 14 days), disappearance time of pharyngeal pain, as well as adverse events. Results: Of 7,699 enrolled participants, 6,693(86.9%) received acupoint application and 1,450(21.7%) with non-acupoint application. After PSM, there were 1,004 patients each in the application group(AG) and non-application group(NAG). The disappearance rate of pharyngeal pain in the AG at 3, 7, and 14 days were all higher than those in the NAG(Pco.fidence interval: 1.41–1.63). The median age of effective cases was 4 years, mainly 3–6 years old(40.21%). The disappearance rate of pharyngeal pain in the application group with tonsil diseases was 2.19 times higher than that in the NAG(Pco.monly used acupoints for the effective cases were Tiantu(RN 22), Shenque(RN 8) and Dazhui(DU 14). The co.monly used herbs for the effective cases were Natrii sulfas, Radix et Rhizoma Rhei, and Herba Ephedrae. Among them, Natrii sulfas was applied to RN 8 most frequently(support 84.39%). A sustainable and practical process is presented for the direct synthesis of sodium tanshinone IIA sulfonate(STS).Our approach was inspired by the well-established and industrially applied batch synthetic route for STS *** co.structed a telesco.ed two-step co.tinuous flow *** involved a co.tinuous tanshinone IIA sulfonation and in-line salt *** the setup,we co.structed a 3D circular cyclone-type microreactor using femtoseco.d laser *** to the 68%yield for 2 h in batch,the two-step co.tinuous flow had an STS yield of 90%,achieved for a total residence time ofco.tinuous flow method vastly simplified the operation and improved procedural safety,while significantly reducing the required acid co.tent and wastewater production.
