For over two decades,the development of B-cell lymphoma-2(Bcl-2)family therapeutics has primarily focused on anti-apoptotic proteins,resulting in the first-in-class drugs called BH3 mimetics,especially for Bcl-2 inhibitor *** pro-apoptotic protein Bcl-2-associated X protein(BAX)plays a crucial role as the executioner protein of the mitochondrial regulated cell death,co.tributing to organismal development,tissue homeostasis,and *** dysregulation of BAX is closely associated with the onset and progression of diseases characterized by pathologic cell survival or death,such as cancer,neurodegeneration,and heart *** addition to co.ducting thorough investigations into the physiological modulation of BAX,research on the regulatory mechanisms of small molecules identified through biochemical screening approaches has prompted the identification of functional and potentially druggable binding sites on BAX,as well as diverse all-molecule BAX *** review presents recent advancements in elucidating the physiological and pharmaco.ogical modulation of BAX and in identifying potentially druggable binding sites on ***,it highlights the structural and mechanistic insights into small-molecule modulators targeting diverse binding surfaces or co.formations of BAX,offering a promising avenue for developing next-generation apoptosis modulators to treat a wide range of diseases associated with dysregulated cell death by directly targeting BAX.

co.lagen,the most abundant structural protein in the human extracellular matrix(ECM),provides essential support for tissues and guides tissue *** its widespread use in tissue engineering,there remains uncertainty regarding the optimal selection of co.lagen ***-derived sources pose challenges such as immunogenicity,while the reco.binant system is hindered by diminished *** this study,we hypothesized that human ECM-like co.lagen(hco.)co.ld offer an alternative for tissue *** this study,a facile platform was provided for generating hco. derived from mesenchymal stem cells with a hierarchical structure and biochemical properties resembling native *** results further demonstrated that hco. co.ld facilitate basal biological behaviors of human adipose-derived stem cells,including viability,proliferation,migration and adipocyte-like ***,it co.ld promote cutaneous wound *** to its high similarity to native co.lagen and good bioactivity,hco. holds promise as a prospective candidate for in vitro and in vivo applications in tissue engineering.

Raman spectrosco.y has found extensive use in monitoring and co.trolling cell culture *** this co.text,the prediction accuracy of Raman-based models is of paramount ***,models established with data from manually fed-batch cultures often exhibit poor performance in Raman-co.trolled ***,there is a need for effective methods to rectify these *** objective of this paper is to investigate the efficacy of Kalman filter(KF)algorithm in co.recting Raman-based models during cell ***,partial least squares(PLS)models for different co.ponents were co.structed using data from manually fed-batch cultures,and the predictive performance of these models was ***,various co.rection methods including the PLS-KF-KF method proposed in this study were employed to refine the PLS ***,a case study involving the auto-co.trol of gluco.e co.centration demonstrated the application of optimal model co.rection *** results indicated that the original PLS models exhibited differential performance between manually fed-batch cultures and Raman-co.trolled *** gluco.e,the root mean square error of prediction(RMSEP)of manually fed-batch culture and Raman-co.trolled culture was 0.23 and 0.40 g·L^(-1).With the implementation of model co.rection methods,there was a significant improvement in model performance within Raman-co.trolled *** RMSEP for gluco.e from updating-PLS,KF-PLS,and PLS-KF-KF was 0.38,0.36 and 0.17 g·L^(-1),***,the proposed PLS-KF-KF model co.rection method was found to be more effective and stable,playing a vital role in the automated nutrient feeding of cell cultures.

Dammarane-type triterpenoids from Gynostemma longipes and their protective activities on hypoxia-induced injury in PC12 cells

Sixteen new dammarane-type triterpenoid saponins(1-16)featuring diverse structural variations in the side chain at C-17,along with twenty-one known analogues(17-37),have been isolated from the rhizomes of Gynostemma longipes ***,a plant renowned for its medicinal and edible *** structural elucidation of these co.pounds was acco.plished through co.prehensive analyses of 1D and 2D NMR and HRMS spectrosco.ic data,supplemented by co.parison with previously reported *** assays on the isolates for their protective effects against hypoxia-induced damage in pheochromocytoma cells(PC12 cells)revealed that nine saponins exhibited significant anti-hypoxic *** investigation into the anti-hypoxia mechanisms of the representative saponins demonstrated that co.pounds 22 and 36 markedly reduced the levels of hypoxia-induced ***,these co.pounds were found to decrease the release of lactate dehydrogenase(LDH)and malondialdehyde(MDA),while increasing the activity of superoxide dismutase(SOD),thereby indicating that the saponins co.ld mitigate hypoxia-induced injuries by ameliorating apoptosis and oxidative *** findings offer substantial evidence for the future utilization and development of ***,identifying dammarane-type triterpenoid saponins as its active anti-hypoxic co.stituents.

OBJECTIVE: To explore the material basis of the difference of efficacy of Dahuang(Radix Et Rhizoma Rhei Palmati)-Taoren(Semen Persicae)(DT) drugs with different proportions. METHODS: Samples of different ratios of Dahuang(Radix et Rhizoma Rhei Palnati, DH) to Taoren(Semen Persicae, TR)(Group A 1∶1, B 2∶3, C 3∶2) were analyzed based on gas chromatography time-of-flight mass spectrometry untargeted metabolomics technique. RESULTS: A total of 240 primary metabolites were detected. Forty-one differential metabolites involved nine differential metabolic pathways, of which four were closely related to the efficacy of DT in the treatment of heat and blood stasis syndrome. These pathways included the biosynthesis of amino acid(phenylalanine tyrosine and tryptophan), flavonoids, unsaturated fatty acids, and the glyco.ysis/glyco.enesis pathway. co.CLUSION: There are significant differences in the efficacy of different ratios of DT drugs, and their optimal ratio for the treatment of heat and blood stasis syndrome should be 1∶1.

Overexpression of PbrGA2ox1 enhances pear drought tolerance through the regulation of GA_(3)-inhibited reactive oxygen species detoxification and abscisic acid signaling

Drought stress is a devastating natural disaster driven by the co.tinuing intensification of global warming,which seriously threatens the productivity and quality of several horticultural crops,including ***(GAs)play crucial roles in plant growth,development,and responses to drought *** studies have shown significant reductions of GA levels in plants under drought stress;however,our understanding of the intrinsic regulation mechanisms of GA-mediated drought stress in pear remains very ***,we show that drought stress can impair the accumulation of bioactive GAs(BGAs),and subsequently identified PbrGA2ox1 as a chloroplast-localized GA deactivation *** gene was significantly induced by drought stress and abscisic acid(ABA)treatment,but was suppressed by GA_(3)***2ox1-overexpressing transgenic tobacco.plants(Nico.iana benthamiana)exhibited enhanced tolerance to dehydration and drought stresses,whereas knock-down of PbrGA2ox1 in pear(Pyrus betulaefolia)by virus-induced gene silencing led to elevated drought *** plants were hypersensitive to ABA,and had a lower BGAs co.tent,enhanced reactive oxygen species(ROS)scavenging ability,and augmented ABA accumulation and signaling under drought stress co.pared to wild-type ***,the opposite effects were observed with PbrGA2ox1 silencing in ***,exogenous GA_(3)treatment aggravated the ROS toxic effect and restrained ABA synthesis and signaling,resulting in the co.promised drought tolerance of *** summary,our results shed light on the mechanism by which BGAs are eliminated in pear leaves under drought stress,providing further insights into the mechanism regulating the effects of GA on the drought tolerance of plants.
