The term glaucoma encompasses a spectrum of eye diseases pathologically characterized by an irreversible loss of retinal ganglion cells and their associated axons. Patients initially suffer from gaps in their visual field, the part of the visual range that can be perceived without eye movement. In the early stages of the disease, this damage can be compensated very well by eye movements. This, in turn, means that glaucoma is often not diagnosed until the disease is more advanced.

Foraminifera are highly diverse and have a long evolutionary *** key bioindicators,their phylogenetic schemes are of great importance for paleogeographic applications,but may be hard to recognize *** phylogenetic relationships within the prominent genus Amphistegina are still *** studies on Amphistegina have so far only focused on genetic diversity within single species and suggested a cryptic diversity that demands for further *** molecular sequencing-based approaches,different mass spectrometry-based proteomics approaches are increasingly used to give insights into the relationship between samples and organisms,especially as these do not require reference *** better understand the relationship of amphisteginids and test different proteomics-based approaches we applied de novo peptide sequencing and similarity clustering to several populations of Amphistegina lobifera,*** and *** also analyzed the dominant photosymbiont community to study their influence on holobiont *** analyses indicate that especially de novo peptide sequencing allows to reconstruct the relationship among foraminiferal holobionts,although the detected separation of *** from *** and *** may be partly influenced by their different photosymbiont *** resulting dendrograms reflect the separation in two lineages previously suggested and provide a basis for future studies.

Bladder cancer(BCa)is the most prevalent urological cancer worldwide[1].A significant proportion of BCa(89%)exhibits molecular alterations in the cell cycle pathway,and targeting cyclin-dependent kinases 4 and 6(CDK4/6)is deemed as a promising therapeutic strategy[2].Selective inhibitors of CDK4/6(CDK4/6is)have been approved by the US Food and Drug Administration(FDA)[3].

Background: Determination of cyclosporine A (CsA) and tacrolimus (Tac) in dried blood spots (DBS) could enable drug monitoring in transplanted patients without the necessity of having to take venous blood samples. Therefore, we have developed a method for quantitative determination of calcineurin inhibitors (CNI) by liquid-chromatography-tandem mass spectrometry (LCMS). Methods: In a study with 68 kidney transplant recipients (KTR, 34 CsA, 34 Tac), we tested the clinical application of LCMS monitoring in DBS in comparison to LCMS in whole blood. Results: The measuring range is proven for 27.33 to 1345 ng/ml for CsA and for 1.63 to 39.7 ng/ml for Tac. The requirements for clinical chemical analyses for precision and accuracy are complied with. Stability is documented for a period of 14 days. The study showed the following deviations from LCMS in whole blood for determination of CsA and Tac in DBS after introducing a correction factor by the haematocrit (Hct) value (CsA trough level: mean = 4.7%, ±1.96 standard deviation (SD) -52.1% to 61.4%, N = 96;CsA peak level: mean = 7.3%, ±1.96 SD -39.7% to 54.4%, N = 95;Tac trough level: mean = -0.5%, ±1.96 SD -76.4% to 75.3%, N = 88;Tac peak level: mean = 3.9%, ±1.96 SD -80.1% to 88.7%, N = 92). Conclusions: Our data show comparable results with the reference method by means of LCMS in whole blood. Therefore, DBS of KTR for determination of CNI levels could be transported on filter cards by mail to the respective laboratory resistant to breakage and the hazard of infection.

Rarely,scientific developments centered around the patient as a whole *** multidisciplinary group,headed by gastrointestinal surgeons,applied this research philosophy considering the most important aspects of thediseases“colon-and rectal cancer”in the long-term *** expertcooperation/knowledge at the Comprehensive Cancer center Ulm(CCCU)wereapplied in several phase III trials for multimodal treatments of primary tumors(MMT)and metastatic diseases(involving nearly 2000 patients and 64 centers),fortreatment individualization of MMT and of metastatic disease,for psychooncology/quality of life involving the patients’wishes,and for disease *** of the targets initially were heavily rejected/discussed in thescientific communities,but now have become standards in treatments andnational guidelines or are topics in modern translational research protocolsinvolving molecular biology for e.g.,“patient centered individualized treatment”.In this context we also describe the paths we had to tread in order to realize ournew goals,which at the end were highly beneficial for the patients from manypoints of *** description is also important for students and youngresearchers who,with an actual view on our recent developments,might want toknow how medical progress was achieved.
