星形胶质细胞是大脑炎症过程的重要介质,可能在包括癫痫在内的几种神经系统疾病中起重要作用。星形胶质细胞中可生产几种miRNA,是炎症通路的关键调节因子,还可能用作治疗靶标。本研究将探索在体外IL-1β介导的炎症条件下,星形胶质细胞内生成的miRNA及其功能,并在人癫痫脑组织中验证。我们通过测序评估IL-1β刺激的人胎儿星形胶质细胞培养物中的miRNA和mRNA表达。使用miRNA模拟物在细胞培养物中过表达miRNA。使用原位杂交技术检测患有结节性硬化症复合体或颞叶癫痫伴海马硬化患者的切除脑组织中miRNA的表达。我们发现了2种差异表达的miRNA:miR146a和miR147b,它们与免疫/炎症反应相关基因的表达增加有关。既往有研究报道,在星形胶质细胞和结节性硬化复合细胞培养物中使用IL-1β刺激后,miR147b的过表达降低了促炎介质IL-6和COX-2的表达。miR146a和miR147b过表达降低了星形胶质细胞的增殖并促进人神经干细胞的神经元分化。miR147b在致癫痫脑中的星形胶质细胞中表达增加。由于其抗炎和恢复异常星形细胞增殖和促进神经元分化的能力,miR146a和miR147b作为与炎症相关的神经疾病(例如癫痫)的潜在治疗靶标值得进一步研究。

Psoriasis (Ps) is an inflammatory skin disease caused by genetic and environmental factors. Previous studies on DNA methylation (DNAm) found genetic markers that are closely associated with Ps, and evidence has shown that DNAm mediates genetic risk in Ps. In this study, Consensus Clustering was used to analyze DNAm data, and 114 Ps patients were divided into three subclassifications. Investigation of the clinical characteristics and copy number variations (CNVs) of DEFB4, IL22, and LCE3C in the three subclassifications revealed no significant differences in gender ratio and in Ps area and severity index (PASI) score. The proportion of late-onset ( ≥ 40 years) Ps patients was significantly higher in type I than in types II and III (P = 0.035). Type III contained the smallest proportion of smokers and the largest proportion of non-smoking Ps patients (P = 0.086). The CNVs of DEFB4 and LCE3C showed no significant differences but the CNV of IL22 significantly differed among the three subclassifications (P = 0.044). This study is the first to profile Ps subclassifications based on DNAm data in the Chinese Han population. These results are useful in the treatment and management of Ps from the molecular and genetic perspectives.

Objective:To evaluate the anti-inflammatory effect of black cumin seed oil(BCSO)and its nanoemulsion on lipopolysaccharide(LPS)-induced liver injury in ***:LPS-induced acute liver injury mouse model was used to evaluate the effects of BCSO and its nanoemulsion formulation on liver *** inflammatory markers including Toll-like receptor 4(TLR4),interleukin(IL)-1β,heme-oxygenase 1,BAX,and BCL-2 were assessed using real-time ***,protein levels of reduced glutathione,tumor necrosis factor-α,and IL-6 were measured using ELISA,and histological analysis was *** was used as a standard positive control for ***:BCSO reduced LPS-induced liver injury and exhibited strong anti-inflammatory effects by downregulating the expression of TLR4,IL-1β,IL-6,tumor necrosis factor-α,and heme-oxygenase ***,BCSO demonstrated antioxidant properties by increasing reduced glutathione protein levels and decreasing key apoptotic markers BAX and BCL-2 in *** nanoemulsion formulation further enhanced these anti-inflammatory,antioxidant,and anti-apoptotic effects,and histological examination confirmed this *** BCSO with indomethacin at a lower dose improved efficacy,thereby reducing its potential side ***:The investigation reveals the anti-inflammatory impact of BCSO and its nanoemulsion formulation on LPS-induced liver oxidative stress,inflammation,and apoptosis.

Influence of Danshen Injection on airway inflammation and CD4^+ CD25^+ regulatory T cells of asthmatic rats

Objective： To investigate the influence of Danshen Injection on airway inflammation and CD4^＋CD25^＋ regulatory T cells（CD4^＋CD25^＋ Tr） of asthmatic rats, and elucidate the possible mechanism of Danshen Injection in treatment of asthma. Methods： 30 Wister rats were randomly divided into control group, asthma group and Danshen Injection treated group. Bronchoalveolar lavage fluids （BALF） were collected, and cytology studies were conducted. Lung tissues were obtained and pathologic analyses were done with hematoxylin and eosin stain （HE）. Flow cytometry was used to detect the CD4^＋CD25^＋ Tr ratio in peripheral blood mononuclear cells （PBMCs）. Results： Total cell, the percentage of lymphocytes, neutrophils and eosinophils （Eos） in BALF of Danshen Injection-treated group were lower than that in asthma group （P〈0.05, P〈0.01）. Compared with asthma group, less infiltration of inflammatory cells in lung tissues was observed in Danshen Injection-treated group. CD4^＋CD25^＋ Tr of asthma group was lower than that of control and Danshen Injection treated group （P〈0.05）. Conclusion： Danshen Injection can suppress airway inflammation of asthmatic rats, probably by increasing the number of CD4^＋CD25^＋ Tr.

Objectives: To determine whether an organized, citywide asthma management prog ram delivered by primary care providers (PCPs) increases adherence to the Nation al Asthma Education and Prevention Program (NAEPP) Asthma Guidelines and whether adherence to the guidelines by PCPs decreases medical services utilization in l ow-income, minority children. Study design: Analysis of the utilization of medi cal services for a cohort of 3748 children with asthma who presented for care at one of six primary care urban clinics in Hartford, Connecticut, and whowere enr olled in a disease management program (Easy BreathingTM) between June 1, 1998 an d August 31, 2002. Results: Of the 3748 children with physician confirmed asthma , 48%had persistent disease. Paid claims for inhaled corticosteroids increased 25%(P inflammatory therapy increased from 38%to 96%. Easy Br eathing children with asthma experienced a 35%decrease in overall hospitalizati on rates (P disease management program increased adherence to the NAEPP guidelines for anti-inflammatory use by PCPs in urban clinics. Adherence to this element of t he guidelines by PCPs reduced hospitalizations, ED visits, and outpatient visits for children with asthma.

Rheumatoid arthritis(RA) is an inflammatory autoimmune disease triggered by antigenic peptides with environmental and genetic risk factors. It has been shown that antigen-specific targeting could be a promising therapeutical strategy for RA by restoring immune tolerance to self-antigens without compromising normal immunity. Citrullination of antigens enhances antigenic properties and induces autoimmune responses. Here, we showed that citrullinated antigenic(citAg) vaccine ameliorated collageninduced arthritis with decreased T-helper 1(Th1) and Th17 cells, downregulated proinflammatory cytokines including interlukin-6 and tumor necrosis factor-a, and inhibited antigen recall responses. B cell receptor sequencing further revealed that citAg vaccine could dampen the dysregulated V(D)J recombination, restoring the immune repertoire. Taken together, the results demonstrated that citAg vaccine might have a therapeutic effect on RA.
