Bone loss and deterioration of bone microarchitecture would increase the bone fragility and fracture risk,leading to the *** and more evidences proved that plant-derived polysaccharides could have a remarkable influence on osteoblasts and osteoclasts,exerting anti-osteoporosis *** to the previous research,the extract of Cibotium barumoz,Achyranthes bidentata,Curculigo orchioides,Epimedium brevicornum,Angelica sinensis,Polygonatum sibiricum,Dendrobium officinale,Morinda officinalis,Nelumbo mucifera,Diospyros kaki,Hordeum vulgare,Cistanche deserticola,Commiphora Myrrha and other plant-derived polysaccharides could benefit to the osteoblasts and *** essential mechanisms are mainly related to the activation or inhibition of many factors,including runt-related transcription factor 2(Runx2),B-catenin,osterix(Osx),activator protein-1(AP-1),osteocalcin(OCN/BGP),alkaline phosphatase(ALP),osteopontin(OPN),bone morphogenetic protein(BMP),phosphatidylinositol 3-kinase(PI3K)/C-Jun N-terminal kinase(JNK)/extracellular regulated protein kinase(ERK),osteoprotegerin(OPG),receptor activator of nuclear factor-kB(RANK),monocyte/macrophage colony-stimulating factor(M-CSF),tumor necrosis factor receptor-associated factor 6(TRAF-6),receptor activator of nuclear factor(NF)-KB ligand(RANKL),nuclear factor of activated T cells 1(NFATc1),c-Fos,matrix metallopeptidase-9(MMP-9),glycogen synthase kinase 3β(GSK3B)/B-catenin,nuclear factor E2-related factor 2(Nf2),as well as these related pathways,such as Wnt/p-catenin,BMP-2/SMAD1/5/8,PI3K/AKT,OPG/RANKL/RANK,NF-κB,MAPKs,*** plant-derived polysaccharides could improve the dynamic balance of bone formation and resorption through promoting the differentiation and maturation of osteoblast or inhibiting its *** reviewed plant-derived polysaccharides and their regulating mechanisms on the osteoclasts and osteoblasts provide the evidences for the development of osteoporosis therapeutics.

Mycobacterium tuberculosis(Mtb)is responsible for the spread of tuberculosis(TB).The current study employed virtual screening of 2569 natural compounds against the DNA-directed RNA polymerase(RNAP)of Mtb to identify the possible binders that can inhibit its *** in-silico methodology included molecular docking to the compounds,further,the stability and flexibility of the best complexes were studied using molecular dynamics simulation,the MM/GBSA binding free energy technique with energy decomposition,PCA,FEL,steered MD simulation,and umbrella *** virtual screenings were conducted for the five RNAP subunits(chains A,B,C,D,and E)to identify a compound capable of inhibiting RNAP oligomerization.A promising compound,isoestradiol 3-benzoate,exhibited a low binding score(−7.28kcal/mol to−8.21kcal/mol)and showed binding ability with all subunits of the ***,the five complexes with isoestradiol 3-benzoate were selected for molecular dynamics simulation ***,RMSD showed that isoestradiol 3-benzoate bound with chain E showed the lowest RMSD of 0.49nm,while with chains A and B it had the most stable and consistent conformations with RMSD of 1.75nm and 1.2nm,*** H-bond between isoestradiol 3-benzoate and chains C and E showed the highest occupancy(58.27%,45.33%,and 50.80%,42.25%,11.75%).Moreover,the MMPBSA technique showed that isoestradiol 3-benzoate had a strong association with chains B and C(ΔGbind=−126.25±2.03 and−129.27±2.25).Additionally,free energy decomposition,PCA,FEL-steered MD simulation,and umbrella sampling were also performed to validate the association of the ligand with the *** 3-benzoate binds strongly to chains B and E;therefore,it should be considered as viable candidate for inhibiting the formation of RNAP protein complex,concluded in this study.

Camptothecin is a complex monoterpenoid indole alkaloid with remarkable antitumor *** that two C-10 modified camptothecin derivatives,topotecan and irinotecan,have been approved as potent anticancer agents,there is a critical need for methods to access other aromatic ringfunctionalized congeners(e.g.,C-9,C-10,etc.).However,contemporary methods for chemical oxidation are generally harsh and low-yielding when applied to the camptothecin scaffold,thereby limiting the development of modified *** herein,we have identified four tailoring enzymes responsible for C-9 modifications of camptothecin from Nothapodytes tomentosa,via metabolomic and transcriptomic *** consist of a cytochrome P450(Nt CPT9H)which catalyzes the regioselective oxidation of camptothecin to 9-hydroxycamptothecin,as well as two methyltransferases(Nt OMT1/2,converting 9-hydroxycamptothecin to 9-methoxycamptothecin),and a uridine diphosphate-glycosyltransferase(Nt UGT5,decorating 9-hydroxycamptothecin to9-β-D-glucosyloxycamptothecin).Importantly,the critical residues that contribute to the specific catalytic activity of Nt CPT9H have been elucidated through molecular docking and mutagenesis *** work provides a genetic basis for producing camptothecin derivatives through metabolic *** will hasten the discovery of novel C-9 modified camptothecin derivatives,with profound implications for pharmaceutical manufacture.

An ethnobotanical study of medicinal plants in Güce district,north-eastern Turkey

This study aims to identify medicinal plants traditionally used to treat diseases by local people living in Güce district,north-eastern *** study was carried out between 2018 and 2021 using a two-part semi-structured,open-ended questionnaire with 165 local *** were analysed using use-report,frequency of citation,and informant consensus *** identified 128 vascular medicinal plant taxa belonging to 54 families and 106 *** most common plant taxa belong to Rosaceae(16 taxa/12.5%),Asteraceae(12 taxa/9.4%),and Lamiaceae(9 taxa/7%)*** most frequently used preparation method reported was decoction(39.8%);the most commonly utilized plant parts were leaves(40.3%).Statistical analysis reveals that women in Güce district(df?163,p?0.043medicinal plants for the first time,and documented a total of 293 new therapeutic ***,the study indicates that the transfer of traditional knowledge to future generations is limited(F?3.355,p?0.020).Action should be taken as soon as possible to preserve existing traditional knowledge and to ensure its transfer to future generations.

Evaluation of hypolipidemic Marrubium vulgare effect in Triton WR-1339-induced hyperlipidemia in mice

Objective: To evaluate the hypocholesterolemic and hypotriglyceridemic activities of four Marrbium vulgare(M. vulgare) herb extracts using Triton WR-1339-induced hyperlipidemia in mice. Methods: Hyperlipidemia was developed by intraperitoneal injection of Triton(200 mg/kg body weight). The animals were divided into main four groups of eight mice each: normal control group, hyperlipidemic control group, hyperlipidemic plus tween-40 control and treated group. The fourth one was divided into four subgroups, petroleum ether extract group, chloroform extract group, ethyl acetate extract group and methanol extract treated group each of them contains two sub-sub group for treating animals with two doses at 0.1 and 0.25 LD50. Results: After 7 h and 24 h of treatment, the intragastric administration of all extracts caused a significant decrease of plasma total cholesterol. Triglyceride levels were also significantly lowered by all extracts while petroleum ether produced the lowest decreasing level. Similar results were observed for LDL-cholesterol concentrations. Furthermore, more polar extracts(methanol and ethyl acetate)-soluble fractions showed a significant ameliorative action on elevated atherogenic index(AI) and LDL/HDL-C ratios, while these atherogenic markers were not statistically suppressed by the chloroform and petroleum ether-soluble extract. Conclusion: The findings indicated that Marrubium may contain polar products able to lower plasma lipid concentrations and might be beneficial in treatment of hyperlipidemia and atherosclerosis.

Herbal medicines are popular natural medicines that have been used for *** use of alternative medicines continues to expand rapidly across the *** World Health Organization suggests that quality assessment of natural medicines is essential for any therapeutic or health care applications,as their therapeutic potential varies between different geographic origins,plant species,and *** of herbal medicines based on a limited number of secondary metabolites is not an ideal *** quality should be considered based on a complete metabolic profile,as their pharmacological activity is not due to a few specific secondary metabolites but rather a larger group of bioactive compounds.A holistic and integrative approach using rapid and nondestructive analytical strategies for the screening of herbal medicines is required for robust *** this study,a rapid and effective quality assessment system for geographical traceability,species,and variety-specific authenticity of the widely used natural medicines turmeric,Ocimum,and Withania somnifera was investigated using Fourier transform near-infrared(FT-NIR)spectroscopy-based metabolic *** different geographical origins of turmeric,five different Ocimum species,and three different varieties of roots and leaves of Withania somnifera were studied with the aid of machine learning *** good discrimination(R^(2)>0.98,Q^(2)>0.97,and accuracy=1.0)with sensitivity and specificity of 100%was achieved using this metabolic fingerprinting *** study demonstrated that FT-NIR-based rapid metabolic fingerprinting can be used as a robust analytical method to authenticate several important medicinal herbs.

In this study,four biochars prepared from different crop residue waste *** bagasse(SBB),coconut shell(CNB),paddy straw(PDB),and distilled waste of lemongrass(LGB)were evaluated for removal of Remazol Brilliant Blue R from the aqueous *** RBBR adsorption capacities of biochar were 97-79%for SBB,99.9-99.47%for CNB,66.1-48%for PDB,and 78-68%for LGB,dominantly controlled by their aromaticity and mineral *** Langmuir and Freundlich isotherms and pseudo-second-order kinetic models have described the chemisorption of RBBR on biochar *** thermodynamic data suggested that adsorption was spontaneous and *** biochars demonstrated excellent reusability(till four cycles with 50-61%regeneration).The purified water and biochar dye sludge demonstrated no *** findings obtained in this study may provide supports for the potential of biochars for anionic dye removal from water and utilization of generated sludge for zero waste-producing technologies in the future.

Ten ring-B aromatized ergosterols from Aspergillus spectabilis

Spectasterols F−O(1−10),ten interesting ergosterols with an aromatized B ring,were obtained from Aspergillus *** structures and absolute configurations were determined using a combination of high-resolution electrospray ionization mass spectrometry(HR-ESI-MS),nuclear magnetic resonance(NMR)spectroscopy,single-crystal X-ray diffraction analyses,and electronic circular dichroism(ECD)***,these aromatic ergosterols feature versatile side ***,compound aromatic ergosterols featured versatile side chains,and compound 4 is an unusual C23 ergosterol characterized by a shorter side chain due to oxidative cleavage between C-23 and *** compounds were evaluated for their neuroprotective activities,with compound 8 showing a dose-dependent ability to reduce apoptosis and protect mitochondrial function in glutamate-induced SH-SY5Y cells.
