Generation of Hematopoietic Stem Cells from Purified Embryonic Endothelial Cells by a Simple and Efficient Strategy

Recent progress by versatile approaches supports the new hypothesis that multi-potent hematopoietic stein cells （HSCs） are directly formed from a rare population of endothelial cells in mid-gestation mouse embryos. This process is therefore known as the endothelial-to- hematopoietic transition （EHT）. Nevertheless, there is no functional evidence that documents the HSC transition from purified endothelial cells. In this study, we developed an OP9-DLl-based co-culture system that was able to facilitate the HSC specification and/or expansion in vitro of mouse embryonic day 10.5 （El0.5） Tie2~ cells remarkably. Then, the immunophenotypically defined endothelial ceils were harvested by a combination of surface markers （Flkl＋CD31 ~CD41 CD45 Ter119 ） from the caudal half of EI0.0-EI 1.0 mouse embryos. The transplantation of the endothelia/OP9-DL1 co-cultures led to long-term, high-level, multi-lineage, and multi-organ he- matopoietic reconstitution in the irradiated adult recipients. The induced HSC activity was initially observed at El0.5, and a significant increase was detected at El 1.0, which suggests a temporally specific regulation. Taken together, tbr the first time, we provide functional evidence showing the HSC potential of purified embryonic endothelial cells, which is indispensable for the emerging EHT concept. Moreover, the newly defined co-culture system will aid the exploration of the key molecules governing the HSC transition from embryonic and even postnatal endothelial cells, which has enormous significance in basic and translational research.

Dear Editor,Clinical and anatomic investigations strongly suggest that a low orbital cerebrospinal fluid pressure (CSFP), parallel to an elevated intraocular pressure (IOP), may have a role in the pathogenesis of optic neuropathy (Ren et al., 2011, Wang et al., 2012). Optic neuropathy including glaucoma demonstrated structural alterations in the optic nerve head (ONH)and retinal nerve fiber layer (RNFL), as well as functional abnormalities such as visual field (VF) that eventually cause severe visual impairment and blindness.

To date,there has been a dramatic increase in the incidence of adenocarcinomas of the esophagogastric junction （AEG）*** classification of AEG,defined by Siewert and stein,was approved at the second International Gastric Cancer Congress in Munich in April *** accordance with the anatomic cardia,EGJC can be divided into three subtypes：type Ⅰ,adenocarcinoma of the distal esophagus with the center located within 1 cm above and 5 cm above the anatomic esophagogastric junction （EGJ）;type Ⅱ,true carcinoma of the cardia with the tumor center within 1 cm above and 2 cm below the EGJ;type Ⅲ,subcardial carcinoma with the tumor center between 2 and 5 cm below EGJ,which infiltrates the EGJ and distal esophagus from below （1）.

Melanopsin-expressing retinal ganglion cell loss and behavioral analysis in the Thy1-CFP-DBA/2J mouse model of glaucoma

In this study, the role of melanopsin-expressing retinal ganglion cells （mRGCs） in the glaucoma-induced depressive behavioral response pattern was investigated. The CFP-D2 transgenic glaucoma animal model from five age groups was used in this study. Immunohistochemical labeling, quantitative analysis of mRGC morphology, open field test （OFT）, and statistical analysis were used. In comparison with C57 BL/6 mice, the age-matched CFP-D2 mice had significantly elevated intraocular pressure （lOP）. We observed parallel morphological changes in the retina, including a reduction in the density of cyan fluorescent protein- （CFP） expressing cells （cells mm^-2 at 2 months of age, 1309±26; 14 months, 878±30, P〈0.001）, mRGCs （2 months, 48_＋3; 14 months, 19±4, P〈0.001）, Brn3b-expressing RGCs （2 months, 1283±80; 14 months, 950±31, P〈0.001）, Brn-3b expressing mRGCs （5 months, 50.17%±5.5%; 14 months, 12.61%±3.8%, P〈0.001）, and reduction in the dendritic field size of mRGCs （mm^2 at 2 months, 0.077±0.015; 14 months, 0.065±0.015, P〈0.05）. CFP-D2 mice had hyperactive locomotor activity patterns based on OFT findings of the total distance traveled, number of entries into the center, and time spent in the center of the testing apparatus. The glaucoma induced hyperactive response pattern could be associated with dysfunctional mRGCs, most likely Brn-3b-positive mRGCs in CFP-D2 mice.

Ifosfamide, Cisplatin or Carboplatin, and Etoposide （ICE）-based Chemotherapy for Mobilization of Autologous Peripheral Blood Stem Cells in Patients with Lymphomas

Background： High-dose chemotherapy followed by autologous stem cell transplantation （ASCT） is a promising approach tbr lymphomas. This study aimed to evaluate the effect of ifosfamide, cisplatin or carboplatin, and etoposide （ICE）-based regimen as a mobilization regimen on relapsed, refractory, or high-risk aggressive lymphoma. Methods： From June 2001 to May 2013, patients with lymphomas who mobilized by ICE-based regimen for ASCT were analyzed in this retrospective study. The results of the autologous peripheral blood stem cells collection, toxicity, engraftment after ICE-based mobilization regimen were analyzed in this study. Furthermore, risk factors for overall survival （OS） and progression free survival （PFS） were evaluated by univariate analysis. Results： The stem cells were mobilized using ICE-based regimen plus rituximab or ICE-based regimen alone in 12 patients and 54 patients, respectively. The results of stem cell mobilization were excellent. Ninety-seven percentages of the patients had the stein cell collection of at least 2.0 ×10^6, CD34^＋ cells/kg and 68% had at least 5 × 10^6, CD34^＋ cells/kg. Fifty-eight percentage of the patients experienced Grade 4 neutropenia, 20% developed febrile neutropenia, and only 12% had Grade 4 thrombocytopenia. At a median follow-up of 63.8 months, the 5-year PFS and OS were 64.4% and 75.3%, respectively. Conclusion： ICE is a powerful regimen for stem cell mobilization in patients with lymphomas.

Taxonomy and ultrastructure of five species of Tetraselmis (Prasinophyceae) isolated from China seas

Comparative ultrastructural investigations on twenty-three isolates of Tetraselmis from the South China Sea, East China Sea and Huanghai Sea, have revealed that these isolates belong to:(1)Tetraselmis chui Butcher; (2)T. cordiformis (H. J. Carter) stein; (3)T. helgolandica Kylin; (4)T. suecica (Kylin) Butcher; (5)T. guangdongensis sp. nov. Except T. helgolandica, the others are new records in China. T. guangdongensis sp. nov. is a new species. Its external and anatomical features closely resemble those of T. impellucida McLachlan et Parke. As in that species the pyrenoid is penetrated from many directions by cytoplasmic channels delimited by a double membrane. The protoplast withdraws from the apical portion of the theca, that portion which overlaid the trough inverts and pops out in the form of a teat. But unlike that species a starch sheath is present; pyrenoid matrix is surrounded by thylakoids which intervenes between the matrix and the starch sheath surrounding pyrenoid; the theca is stratified.

Let p∈（0, 1], q∈（0, ∞] and A be a general expansive matrix on Rn. Let HAp,q （Rn） be the anisotropic Hardy-Lorentz spaces associated with A defined via the non-tangential grand maximal function. In this article, the authors characterize HAp,q（Rn） in terms of the Lusin-area function, the Littlewood-Paley g-function or the Littlewood-Paley gλ＊-function via first establishing an anisotropic Fefferman-stein vector-valued inequality in the Lorentz space Lp,q（Rn）. All these characterizations are new even for the classical isotropic Hardy-Lorentz spaces on Rn. Moreover, the range of λ in the gλ＊-function characterization of HAp,q （Rn） coincides with the best known one in the classical Hardy space Hp（Rn） or in the anisotropic Hardy space HAp （Rn）.

Self-adaptive spatial image denoising model based on scale correlation and SURE-LET in the nonsubsampled contourlet transform domain

A novel self-adaptive image denoising model based on scale correlation and stein’s unbiased risk estimate-linear expansion of thresholds(SURE-LET)in the nonsubsampled contourlet transform domain is proposed in this *** we implement the multidimensional and translation invariant decomposition for spatial images by the nonsubsampled contourlet transform,and establish the image cross-scale description *** combining the scale correlation,we make improvements for the existing SURE-LET denoising idea and establish the self-adaptive denoising *** scale correlation calculation is needed for the coefficients at different scales and sub-bands to determine whether the coefficients are retained or processed with the adaptive SURE-LET threshold *** meanwhile a new local context self-adaptive threshold strategy is proposed in the process of scale correlation *** results both on spatial images and standard images demonstrate that the proposed algorithm performs significantly better in terms of both the visual subjective evaluation and the quantitative objective *** method can achieve better noise suppression,and effectively retain image edge details.

Obsessive-compulsive disorder(OCD) is a chronic and debilitating psychiatric disorder that affects ～2%–3% of the population globally. Studying spontaneous OCD-like behaviors in non-human primates may improve our understanding of the disorder. In large rhesus monkey colonies, we found 10 monkeys spontaneously exhibiting persistent sequential motor behaviors(SMBs) in individual-specific sequences that were repetitive, time-consuming and stable over prolonged periods. Genetic analysis revealed severely damaging mutations in genes associated with OCD risk in humans. Brain imaging showed that monkeys with SMBs had larger gray matter(GM) volumes in the left caudate nucleus and lower fractional anisotropy of the corpus callosum. The GM volume of the left caudate nucleus correlated positively with the daily duration of SMBs. Notably, exposure to a stressor(human presence) significantly increased SMBs. In addition, fluoxetine, a serotonergic medication commonly used for OCD, decreased SMBs in these monkeys. These findings provide a novel foundation for developing better understanding and treatment of OCD.

Measuring the number concentration of arbitrarily-shaped gold nanoparticles with surface plasmon resonance microscopy

Molar concentration of gold nanoparticles is one of the most critical parameters of gold colloids in order to develop their applications in sensing, diagnostics and nanomedicine. Previous methods often stand just for gold nanoparticles with regular shape and narrow size distribution. In the present work, we proposed an absolute quantification method that determined the molar concentration of gold nanoparticles with arbitrary shapes and polydisperse sizes. This approach involved the real time monitoring and counting of individual nanoparticles collision events, from which the quantification of molar concentration was achieved using a theoretical model consisting of Fiek＇s laws of diffusion and Stokes-Einstein equation. The determination of spherical gold nanoparticles concentration resulted in excellent agreement with traditional spectrometry method. It was further demonstrated that the present approach can be expanded to determine the molar concentration of gold nanoparticles with arbitrary shapes and poly-diversed distributions.
