Land Use Changes in Himalaya and Their Impacts on Environment, Society and Economy: A Study of the Lake Region in Kumaon Himalaya, India

The traditional resource use structure in Himalaya has transformed considerably during the recent past, mainly owing to the growth of population and the resultant increased demand of natural resources in the region. This transformation in resource use practices is particularly significant in the densely populated tracts of Himalaya. As a result, cultivated land, forests, pastures and rangelands have been deteriorated and depleted steadily and significantly leading to their conversion into degraded and non-productive lands. These rapid land use changes have not only disrupted the fragile ecological equilibrium in the mountains through indiscriminate deforestation, degradation of land resources and disruption of the hydrological cycle, but also have significant and irreversible adverse impacts on the rural economy, society, livelihood and life quality of mountain communities. It has been observed that the agricultural production has declined, water sources are drying up fast due to decreased ground water recharge and a large number of villages are facing enormous deficit of critical resources, such as food, fodder, firewood and water, mainly due to unabated deforestation. As a result, the rural people, particularly the women, have to travel considerably long distances to collect fodder and firewood and to fetching water. It is therefore highly imperative to evolve a comprehensive and integrated land use framework for the conservation of the biophysical environment and sustainable development of natural resources in Himalaya. The land use policy would help local communities in making use of their natural resources scientifically and judiciously, and thus help in the conservation of the biophysical environment and in the increasing of the productivity of natural resources. Influence of short chain ceramides and lipophilic penetration enhancers on the nano-structure of stratum corneum model membranes studied using neutron diffraction

Oriented stratum corneum model lipid membranes were used to study the influence of the short chain ceramides （CER）[NP] and [AP] as well as the impact of the lipophilic penetration enhancer molecules oleic acid （OA） and isopropyl myristate （IPM） on the lipid nanostrueture. The influence of the enhancer molecules were studied using specifically deuterated OA and IPM and neutron diffraction. 2H NMR spectroscopy was used to study the impact of the ceramides＇ degree of order within the stratum corneum model lipid membranes. It was found that CER[NP] forms two very stable phases with high resistance against temperature increase. Phase B showed unusual hydration behavior as no water uptake of this phase was observed. The 2H NMR spectroscopic measurements showed that CER[NP] based ternary model system had a higher state of lamellar order in comparison to CER[AP] based lipid matrix. The studies confirmed that the short chain ceramides, particularly CER[NP], have a very high impact on the integrity of the Stratum comeum lipid bilayers. The penetration enhancer OA has not influenced the repeat distance of the model membrane based on CER[AP], and was not able to induce a phase separation in the investigated lipid matrix. However, a disorder and a fluidisation of the model membranes were observed when OA was incorporated. IPM showed the same effect but two phases （assigned as phase A and B） appeared, when IPM was used as penetration enhancer and incorporated into the model membrane. Furthermore, two arrangements of IPM were identified in phase A using deuterated IPM. A model of the nanostructure of the Stratum corneum lipid membranes is presented.

Machine learning guided high-throughput search of non-oxide garnets

Association of late-life changes in blood pressure and cognitive status

BackgroundDisagreement exists on the association between changes in blood pressure and cognitive impairment. We aimed to ex-amine whether 4-year changes in systolic and diastolic blood pressure （SBP and DBP） are associated with cognitive status in a representative sample of older men and *** of longitudinal data from 854 participants of a population-based German sample （aged 60-87 years） was performed with standard cognitive screening and blood pressure measurements. Effects of changes in SBP and DBP （10 mmHg and 5 mmHg respectively as unit of regression effect measure） on cognitive status were evaluated using non-parametric and lin-ear regression *** clear associations were seen between changes in SBP or in DBP and cognitive scores. Small effects were found after stratification for sex and hypertension awareness. Specifically, larger decreases in SBP were associated with higher cogni-tive scores in those men aware of their hypertension （10 mmHg decrease in SBP,b =-0.26, 95% CI：-0.51 to-0.02） and men with con-trolled hypertension （10 mmHg decrease in SBP,b =-0.44, 95% CI：-0.92 to-0.03）. Additionally larger increases in DBP were associated with higher cognitive scores in men with controlled hypertension （5 mmHg increase in DBP,b = 0.67, 95% CI： 0.19-1.15）. For women aware of their hypertension, larger decreases in DBP were associated with higher cognitive scores （5 mmHg decrease in DBP,b =-0.26; 95%CI：-0.51 to-0.01）.ConclusionsChanges in blood pressure were only weakly associated with cognitive status. Specifically, decreases in SBP were associated with higher cognitive scores in men aware of their hypertension and especially those that were medically controlled.

Dual Targeting of a Mitochondrial Protein： The Case Study of Cytochrome C1

As a result of the endosymbiotic gene transfer, the majority of proteins of mitochondria and chloroplasts is encoded in the nucleus and synthesized in the cytosol as precursor molecules carrying N-terminal transit peptides for the transport into the respective target organelle. In most instances, transport takes place into either mitochondria or chloroplasts, although a few examples of dual targeting into both organelles have been described. Here, we show by a combination of three different experimental strategies that also cytochrome c1 of potato, a component of the respiratory electron transport chain, is imported not only into mitochondria, but also into plastids. In organello import experiments with isolated mitochondria and chloroplasts, which were analyzed in both single and mixed organelte assays, demonstrate that the processing products accumulating after import within the two endosymbiotic organelles are different in size. Dual targeting of cytochrome c1 is observed also in vivo, after biolistic transformation of leaf epidermal cells with suitable reporter constructions. Finally, Western analyses employing cytochrome c1-specific antiserum provide evidence that the protein accumulates in significant amounts in mitochondria and chloroplasts of both pea and spinach. The possible consequences of our findings on the relevance of the dual targeting phenomenon are discussed.

Utilizing melt crystallization fundamentals in the development of a new tabletting technology

Increasing production effeciency and lowering costs are some of the many advantages melt crystallization technology offers over the conventional methodology of tabletting. A normal tablet consists of a pure shell or a coat and a separate core constituting the pharmaceutical active ingredient. Great emphasis is put on the purity of the shell since its purpose is to solely protect and deliver the active ingredient to its target. Melt crystallization is a purification （separation） process. It is discussed here for its ability to produce coated tablets, by separating the ＂coating＂ material from the ＂to be coated＂ material coming from one molten mixture. Molten drops of lutrol-ibuprofen mixture are produced using the drop forming technique. The subsequent analysis involves proving and quantifying the phase separation （coat purity）. The mechanism of a crystallizing drop is shown as direct evidence of the ongoing process. Moreover, solidified tablet batches are analyzed for the purity of their coating by measuring the ibuprofen concentration. This optimization process is carried out through multiple stages of development and condition enhancements in order to produce the most pure tablet coating. As a result, a trial showing an almost purely coated tablet is presented here.

Purification of L-asparaginase II by crystallization

Here a case study ofL-asparaginase II out of a recombinant Escherichia coli is presented. The target protein was obtained by simple cell disintegration and acetone precipitation. The L-asparaginase II has been crystallized in three different forms in the following microbatch crystallization. The rod-shaped crystals （-400 μm edge length） were obtained at either 8℃ or 22℃ after 17h by addition of PEG6ooo. The rectangular-shaped crystals were obtained after further recrystallization of the rod-shaped crystals. The rhombic-shaped crystals formed at 8℃ after 12 days when cold ethanol was used instead of PEG6000. All crystallizations were performed in tris-acetate buffer （50mmol-L-1, pH 5.1）. By crystallization, the specific activity of L-asparaginase II has increased 5-fold. The protein content and the purity of the crystals were evaluated by sodium dodecyl sulfate polyacrylamide gel electrophoresis （SDS-PAGE）. The more concentrated L- asparaginase II out of an extract mixture and the presence of only less minor proteins after crystallization demonstrates that crystallization is an effective and mild method to purify the target protein. The single crystal X-ray diffraction pattern reveals that the crystals are proteins and the X-ray powder diffraction （XRPD） pattern shows clearly that the crystals forming in PEG600o and ethanol have different crystal structures.

Problems, potentials and future of industrial crystallization

This review discusses important research developments and arising challenges in the field of industrial crystallization with an emphasis on recent problems. The most relevant areas of research have been identified. These are the prediction of phase diagrams; the prediction of effects of impurities and additives; the design of fluid dynamics; the process control with process analytical technologies （PAT） tools; the polymorph and solvate screening; the stabilization of non-stable phases; and the product design. The potential of industrial crystallization in various areas is outlined and discussed with particular reference to the product quality, process design, and control. On this basis, possible future directions for research and development have been pointed out to highlight the importance of crystallization as an outstanding technique for separation, purification as well as for product design.

Molecular level simulations on multi-component systems --a morphology prediction method

The crystal morphology grown from a solution composed of an organic solvent, solute and additive can be predicted reliably by a computational method. Modeling the supersaturated solution as liquid phase is achieved by employing commercial software. The molecular composition of this solution is a required input parameter. The face specific diffusion coefficient of the solid （crystal surface） and liquid （solution） system is determined using the molecular dynamics procedure. The obtained diffusion coefficient is related to the specific face growth rate via the attachment energy of the pure morphology. The significant improvements are achieved in the morphology prediction because the investigation on the face growth rates in a complex growth environment （as multi-component solutions with additives） can be carried out based on the diffusion coefficients.

Importance of emulsions in crystallization --applications for fat crystallization

Emulsions and crystallization are two independent research topics which normally do not overlap although a combination of the two could be applicable to many areas. Here, the importance of emulsions in the field of fat crystallization is described. Three applications with industrial relevance were chosen for investigation： fat fractionation, the solidification of phase change materials and solid lipid nanoparticles. For fat fractionation and phase change materials, emulsification can be applied as a tool to improve the fat crystallization process, and thus the product quality of the crystallized fat. Furthermore, the use of emulsification creates new solid lipid nanoparticles in the application fields such as area of fat crystallization.
