Mechanism of Deca-BDE-induced apoptosis in Neuro-2a cells:Role of death-receptor pathway and reactive oxygen species-mediated mitochondrial pathway

Decabromodiphenyl ether（BDE-209） is a prevalent polybrominated diphenyl ether（PBDE）congener known to have neurotoxicity. Effects of BDE-209 on Neuro-2a cells were performed in the present study and the possible apoptotic pathway was discussed. Results indicated that BDE-209 induced Neuro-2a cell apoptosis, increased the protein expression of Fas and Fas-associated death domain-containing protein（FADD） and activated the caspase-8 and-3activities in a concentration-dependent manner, inferring the death-receptor pathway was involved in the apoptotic process. Meanwhile, BDE-209 exposure increased the Bax/Bcl-2 ratio and decreased the cellular mitochondrial membrane potential（MMP） which led to cytochrome C released to the cytoplasm. The intracellular caspase-9 was elevated simultaneously,which caused downstream caspase cascade and triggered cell apoptosis. Moreover, BDE-209 exposure increased cellular reactive oxygen species（ROS） level in a concentration-dependent manner and the addition of N-acetyl-L-cysteine（NAC）, known as ROS scavengers, obviously reduced the apoptotic rate and a positive relationship was observed between the degree of apoptosis blocking and the loss of MMP and ROS production. We thus concluded that BDE-209 induced Neuro-2a cell apoptosis via the combination of the death-receptor signaling pathway and the mitochondrial signaling pathway. The elevated ROS production was considered to magnify the intracellular apoptosis signal and played a crucial role in apoptosis of Neuro-2a cells induced by BDE-209.

Effect of precipitation pH and coexisting magnesium ion on phosphate adsorption onto hydrous zirconium oxide

To understand the effect of precipitation pH and coexisting Mg^(2+) on phosphate adsorption onto zirconium oxide(ZrO_2), ZrO_2 particles precipitated at pH 5.3, 7.1 and 10.5, i.e., ZrO_2(5.3), ZrO_2(7.1)and ZrO_2(10.5), respectively were prepared and characterized, then their adsorption performance and mechanism in the absence and presence of Mg^(2+) were comparatively investigated in this study. The results showed that the Elovich, pseudo-second-order and Langmuir isotherm models correlated with the experimental data well. The adsorption mechanism involved the complexation between phosphate and zirconium. Coexisting Mg^(2+) slightly inhibited the adsorption of phosphate on ZrO_2(5.3), including the adsorption capacity and rate, but coexisting Mg^(2+) greatly increased the adsorption capacity and rate for ZrO_2(7.1)and ZrO_2(10.5). The enhanced adsorption of phosphate on ZrO_2(7.1) and ZrO_2(10.5) in the presence of Mg^(2+) was mainly due to the formation of Mg^(2+)-HPO_4^(2-) ion pair(MgHPO_4~0) in the solution and then the adsorption of MgHPO_4~0 on the adsorbent surface, forming the phosphatebridged ternary complex Zr(OPO_3H)Mg. In the absence of Mg^(2+) , the maximum phosphate adsorption capacity at pH 7 calculated from the Langmuir isotherm model decreased in the order of ZrO 2(7.1)(67.3 mg/g) > ZrO_2(5.3)(53.6 mg/g) ≈ ZrO_2(10.5)(53.1 mg/g), but it followed the order of Zr O2(7.1)(97.0 mg/g) > ZrO_2(10.5)(79.7 mg/g) > ZrO_2(5.3)(51.3 mg/g) in the presence of Mg^(2+) . The results of this work suggest that ZrO_2(7.1) is more suitable for use as an adsorbent for the effective removal of phosphate from municipal wastewater than ZrO_2(5.3) and ZrO_2(10.5),because Mg^(2+) is generally present in this wastewater.

Antioxidant system responses in two co-occurring green-tide algae under stress conditions

Green tides have occurred every year from 2007 to 2014 in the Yellow Sea. Ulva prolifera （Mtiller） J. Agardh has been identified as the bloom-forming alga, co-occurring with U. intestinalis. We observed distinct strategies for both algal species during green tides. *** exhibited a high abundance initially and then decreased dramatically, while U. intestinalis persisted throughout. The antioxidant system responses of these two macroalgae were compared in the late phase of a green tide （in-situ） and after laboratory acclimation. Lipid peroxidation and antioxidant system responses differed significantly between the two. Malondialdehyde and hydrogen peroxide contents increased significantly in-situ in U. prolifera, but not in U. intestinalis. In U. prolifera, we observed a significant decrease in total antioxidant ability （T-AOC）, antioxidant enzymes （SOD and Apx）, and non-enzyme antioxidants （GSH and AsA） in-situ. U. intestinalis showed the same pattern of T-AOC and SOD, but its Gpx, Apx, and GSH responses did not differ significantly. The results suggest that U. prolifera was more susceptible than U. intestinalis to the harsh environmental changes during the late phase of a Yellow Sea green tide. The boom and bust strategy exhibited by U. prolifera and the persistence of U. intestinalis can be explained by differences in enzyme activity and antioxidant systems.

Physiological responses of macroalga Gracilaria lemaneiformis(Rhodophyta) to UV-B radiation exposure

This paper aims to evaluate the effects of ultraviolet-Bradiation(UVBR) on Gracilaria lemaneiformis,a commercial red macroalga and an important source of agar. To study the in-vitro effect of UVBR on G. lemaneiformis,this plant was cultivated and exposed to photosynthetically active radiation(PAR) at 40 μmol photons/(m2 ·s) and enhanced UVBR(0,0.36,0.72,1.08,1.44,and 1.80 k J/(m 2 ·d)) for 13 days. The samples were processed for histochemical analysis,and the growth rate,photosynthetic pigment contents,photosynthetic performance,reactive oxygen species levels,membrane permeability,malonyl dialdehyde contents and antioxidant capacity of G. lemaneiformis were investigated. After 13 days of exposure to PAR+UVBR,G. lemaneiformis showed photodamage and photoinhibition of photosynthetic pigments(chlorophylla and phycoerythrin),leading to a decreased photosynthetic efficiency. Further,there was a corresponding decrease in the relative growth rates and depigmentation and partial necrosis of the apical segments were noted after exposure to PAR+UVBR. Additionally,UVBR induced excess production of superoxide radicals and hydrogen peroxide,eliciting a marked cellular membrane damage and antioxidative response.

The response of spring phytoplankton assemblage to diluted water and upwelling in the eutrophic Changjiang(Yangtze River) Estuary

A comprehensive study on the phytoplankton standing stocks, species composition and dominant species in the eutrophic Changjiang（Yangtze River） Estuary（CE） was conducted to reveal the response of phytoplankton assemblage to Changjiang Diluted Water（CDW） and upwelling in the spring. Phytoplankton presented peak standing stocks（13.03 μg/L of chlorophyll a, 984.5×103 cells/L of phytoplankton abundance） along the surface isohaline of 25. Sixty-six species in 41 genera of Bacillariophyta and 33 species in 19 genera of Pyrrophyta were identified, as well as 5 species in Chlorophyta and Chrysophyta. Karenia mikimotoi was the most dominant species, followed by Prorocentrum dentatum, Paralia sulcata, Pseudo-nitzschia delicatissima and Skeletonema costatum. A bloom of K. mikimotoi was observed in the stratified stations, where the water was characterized by low nitrate, low phosphate, low turbidity, and specific ranges of temperature（18–22 °C） and salinity（27–32）. *** and P. dentatum accumulated densely in the upper layers along the isohaline of 25. S. costatum was distributed in the west of the isohaline of 20. Benthonic P. sulcata presented high abundance near the bottom,while spread upward at upwelling stations. CDW resulted in overt gradients of salinity, turbidity and nutritional condition, determining the spatial distribution of phytoplankton species. The restricted upwelling resulted in the upward transport of P. sulcata and exclusion of S. costatum, K. mikimotoi and P. dentatum. The results suggested that CDW and upwelling were of importance in regulating the structure and distribution of phytoplankton assemblage in the CE and the East China Sea.

Genetic diversity of populations and clones of Rhopilema esculentum in China based on AFLP analysis

Amplified fragment length polymorphisms(AFLP) markers were developed to assess the genetic variation of populations and clones of Rhopilema esculentum Kishinouye(Scyphozoa,Rhizostomatidae).One hundred and seventy-nine loci from 56 individuals of two hatchery populations and two wild populations were genotyped with five primer *** polymorphic ratio,Shannon's diversity index and average heterozygosity were 70.3%,0.346 and 0.228 for the white hatchery population,74.3%,0.313,and 0.201 for the red hatchery population,79.3%,0.349,and 0.224 for the Jiangsu wild population,and 74.9%,0.328 and 0.210 for the Penglai wild population,***,all populations had a relatively high level of genetic diversity.A specific band was identified that could separate the white from the red hatchery *** was 84.85% genetic differentiation within *** cluster analysis using unweighted pair-group method with arithmetic mean(UPGMA) suggested that hatchery populations and wild populations could be *** the hatchery populations,the white and red populations clustered separately;however,for the wild populations,Penglai and Jiangsu populations clustered *** genetic diversity at the clone level was also *** data suggest that there are relatively high genetic diversities within populations but low genetic differentiation between populations,which may be related to the long-term use of germplasm resources from Jiangsu Province for artificial seeding and *** findings will benefit the artificial seeding and conservation of the germplasm resources.

Biotic and Abiotic Conditions Can Change the Reproductive Allocation of Zostera marina Inhabiting the Coastal Areas of North China

The relationships between reproductive allocation and different factors have been widely recognized for terrestrial plants, but scarcely for marine angiosperms. Focusing on the seagrass Zostera marina, we examined the change in biomass when it was partitioned to growth, vegetative propagation and reproductive allocation of Z. marina. We also evaluated the relationships between reproductive allocation and biotic and abiotic conditions in the coastal areas of north China. The results indicated that the reproductive allocation of Z. marina at the levels of genet, shoot and spathe was positively correlated with shoot characteristics such as height and biomass(P < 0.01), but was negatively correlated with branch number and leaf number per genet(P < 0.01). In addition, the allocation was also affected by other abiotic factors such as water column phosphate concentration(P < 0.05) and sediment total petroleum(P < 0.01). These findings indicated that both biotic and abiotic conditions significantly influenced the reproductive allocation of Z. marina, which may affect further the population dynamics.

Allelopathic interactions between Prorocentrum micans and Skeletonema costatum or Karenia mikimotoi in laboratory cultures

Algal allelopathy is an ecological/physiological phenomenon that has focused attention on the interactions among algae and the production of algal toxins. We investigated the allelopathic interactions between the dinoflagellate genus Prorocentrum micans and diatom genus Skeletonema costatum and between P. micans and dinoflagellate genus Karenia mikimotoi using bi-algal cultures. Because the effects were species-specific and size-dependent, we evaluated the effect of different initial densities. At low densities of P. mieans and high densities of S. costatum inoculated into the same medium, the growth of R rnieans was weakly restrained, whereas the growth of S. costatum was significantly suppressed. S. costatum and K. mikimotoi were strongly inhibited by P. micans, in both the bi-algal cultures and enriched filtrates. Direct cell-to-cell contact was not necessary to gain a competitive advantage, thus, our results suggest that P. micans inhibited the growth of S. costatum and K. mikimotoi by the release of allelochemical（s）. Last, a mathematical model was used to simulate growth and interactions between P. micans and S. eostatum and between P. micans and K. mikimotoi in bi-algal cultures.

Dietary vitamin A,ascorbic acid and α-tocopherol affect the gonad development and reproductive performance of starry flounder Platichthys stellatus broodstock

The present trial was conducted with starry flounder Platichthys stellatus broodstock to evaluate the effects of dietary vitamin A, ascorbic acid, and a-tocopherol on the gonadal development and reproductive performance. 8 000 IU/kg diet vitaminA （VA group）, 500 mg/kg diet ascorbic acid （Vcpp group）, or 250 mg/kg diet a-tocopherol （a-TA group） was added into basal diet to create 3 vitamin experimental diets, respectively. Each diet was fed to 450 starry flounder broodstock for 104 days. Samples were collected weekly. The gonadosomatic index （GSI） of 3 vitamin experimental groups first decreased and then increased. Maximum GSI of Vcpp group was higher than that of ct-TA group but lower than that of VA group. The spawning periods of 3 vitamin experimental groups lasted 49, 56, and 45 days, respectively. No mature eggs were observed in the control group during the trial. The absolute fecundity （AF） and relative fecundity （RF） of a-TA group was higher than that of Vcpp group but lower than that of VA group. The results suggest that different vitamins play different roles in the fish reproductive process. Vitamin A stimulated the maturation of the ovary, ascorbic acid prolonged the spawning period, and a-tocopherol affected the development of the eggs.

The Effects of Reclamation Activity and Yellow River Runoff on Coastline and Area of the Laizhou Bay,China

Study on morphological changes of a bay can help to identify the effects of anthropogenic activities on coastal environ-ment and guide the exploration of marine *** this paper,morphological data including coastline and water areas in five discrete years between 1968 and 2015 were selected and extracted from the remote sensing images and historical marine charts to study the morphological changes in Laizhou Bay(LZB),one of the bays in the southwest of the Bohai Sea.A systematic analysis on spatial variations of the coastline and the surface areas of different types of waters in LZB was *** results showed that the surface area of LZB was decreased by 1253.2km^(2)in the last half century,which is 17.4%of the total in the *** areas of the natural wetland and the intertidal zone were decreased by 17.2%and 56.1%,respectively,and the average water depth varied from 9.05 m to 8.16m at low tide level from 1968 to *** coastline and shape variations of the bay turned to be complex after the 1980s,and the shape index of LZB showed an increasing trend in more recent *** centroid of the bay generally migrated to the northeast direction,i.e.,the direction of the center of the Bohai Sea,and the shrinking direction of the bay was consistent with the migration direction of the *** reclamation area during 1968-2015 in LZB was 1201.7km^(2),and 94.1%was in the inter-tidal *** overall morphological change of the bay during the last half century was mainly controlled by the coastal reclamation activities,and the Yellow River runoff including the river course change and sediment load variation was also an important control-ling factor.
