Molecular cytogenetic of the Amoy croaker, A rgyrosomus amoyensis(Teleostei, Sciaenidae)

The family Sciaenidae is remarkable for its species richness and economic importance. However, the cytogenetic data available in this fish group are still limited, especially those obtained using fl uorescence in situ hybridization(FISH). In the present study, the chromosome characteristics of a sciaenid species, Argyrosomus amoyensis, were examined with several cytogenetic methods, including dual-FISH with 18 S and 5 S rDNA probes, and a self-genomic in situ hybridization procedure(Self-GISH). The karyotype of A. amoyensis comprised 2 n=48 acrocentric chromosomes. A single pair of nucleolar organizer regions(NORs) was located at the proximal position of chromosome 1, which was positive for silver nitrate impregnation(AgNO_3) staining and denaturation-propidium iodide(DPI) staining but negative for Giemsa staining and 4',6-diamidino-2-phenylindole(DAPI) staining, and was confi rmed by FISH with 18 S rDNA probes. The 5 S rDNA sites were located at the centromeric region of chromosome 3. Telomeric FISH signals were detected at all chromosome ends with dif ferent intensities, but internal telomeric sequences(ITSs) were not found. Self-GISH resulted in strong signals distributed at the centromeric regions of all chromosomes. C-banding revealed not only centromeric heterochromatin, but also heterochromatin that located on NORs, in interstitial and distal telomeric regions of specifi c chromosomes. These results suggest that the karyotype of Amoy croaker was relatively conserved and primitive. By comparison with the reported cytogenetic data of other sciaenids, it can be deduced that although the karyotypic macrostructure and chromosomal localization of 18 S rDNA are conserved, the distribution of 5 S rDNA varies dynamically among sciaenid species. Thus, the 5 S rDNA sites may have different evolutionary dynamics in relation to other chromosomal regions, and have the potential to be ef fective cytotaxonomic markers in Sciaenidae.

The chromosomes of spinyhead croaker Collichthys lucidus（Richardson,1844） were characterized for the first time by fluorescence staining,self genomic in situ hybridization（self-GISH）,and multicolor fluorescence in situ hybridization（FISH） with 18 SrDNA,5 SrDNA and telomeric sequence *** female karyotype has exclusively 24 pairs of acrocentric chromosomes（2 n=48 a,NF=48）,while the male one consists of 22 pairs of acrocentric chromosomes,2 monosomic acrocentric chromosomes and a metacentric chromosome（2 n=1 m＋46 a,NF=48）.The difference between female and male karyotypes indicates the presence of a sex chromosome of X1 X1 X2 X2/X1 X2 Y type,where Y is the unique metacentric chromosome in the male *** revealed by FISH,5 S r DNA and 18 S r DNA sites were mapped at syntenic position of the largest acrocentric chromosome（X_1）,and the short arms of the Y chromosome as *** X_1-chromosome specific interstitial telomeric signal（ITS） was detected overlapping the 5 S r DNA *** addition,self-GISH revealed that the repetitive DNAs accumulated on all the putative sex *** fusion accompanied by a partial deletion in the ancestral karyotype（2 n=48 a） is hypothesized for the origin of such multiple sex chromosome *** present study,as the first description of differentiated sex chromosome in family Sciaenidae,will give clues to the studies on the sex chromosome of other Sciaenids.

Impacts of dietary supplementation of bamboo vinegar and charcoal powder on growth performance,intestinal morphology,and gut microflora of large-scale loach Paramisgurnus dabryanus

Although accumulating data demonstrate that dietary supplementation of bamboo vinegar and charcoal powder(BVC)can significantly strengthen the immunity and boost the growth of domestic animals,its application potency still awaits verification in commercial fish species such as the large-scale loach Paramisgurnus *** impacts of 90-day dietary supplementation of 1%and 2%BVC on survival and growth performance of the loach,and on the intestinal morphological characteristics and gut microflora were *** data show that the large-scale loach supplied with BVC at the experimental doses had significant higher survival rates and better growth performance(indicated by greater weight gain(1.13–1.14 times),higher specific growth rate(1.04 times),and lower feed conversion ratio(0.88–0.89 times))compared to that of the control(P<0.05).Histological examination revealed significant longer villus(3.22–5.54 times),deeper crypt(1.77–1.87 times),and thicker muscle(1.59–3.17 times)in the intestines of large-scale loach fed with BVC(P<0.05).Furthermore,we found that the gut microflora consisted of significantly fewer proportions of potential pathogenic bacterial species(Aeromonas veronii and Escherichia coli),but significantly greater proportions of beneficial microbes(Lactococus raffinolactis and Faecalibacterium prausnitzii).Therefore,dietary intake of BVC can promote intestinal tract development and optimize gut microflora,by which the survival and growth of large-scale loach may be improved.

Growth performance of the clam, Meretrix meretrix, breeding-selection populations cultured in different conditions

A breeding-selection program for the clam, Meretrix meretrix, was conducted since 2004. Two of the selec- tion populations were generated with the shell color pattern as an additional selection criterion and named as SP （purple stripes） population and SB （black dots） population. The third-generation SP and SB popula- tions （08G3SP and 08G3SB, respectively） were cultured at two commercial clam farms and a nursery pond and their shell lengths were compared. 08G3SB clams had significantly larger sizes than 08G3SP clams at commercial clam farms （p 〈 0.05）, yet 08G3SB individuals were significantly smaller than 08G3SP individ- uals at the nursery pond （p 〈 0.05）. Then, we examined the growth of the fourth-generation SP and SB populations （10G4SP and 10G4SB, respectively） at a commercial farm, and found that the shell lengths of the 10G4SB clams increased at a significantly higher growth rate than the 10G4SP clams （p 〈 0.05） from May to September, when the water temperature was between 24.2-27.5℃, while 10G4SB lost the growth advantage in the other months. These results indicated that SP and SB populations responded differently to environmental factors, so it is beneficial for the clam farmers to select a suitable population according to the culture environment. Furthermore, a diallel mating of the SB and SP populations was designed to investigate whether their hybrid population would show heterosis. However, the heterosis was not shown in this study, which might result from the slight genetic divergence between SB and SP populations.

Ethidium bromide(EtBr)is one of the contaminants recorded in aquatic environments whose effects have been investigated;however,there is still limited knowledge about its *** study examined the potential protective effects of Spirulina platensis(SP)against the effects of EtBr toxicity in the Nile tilapia(Oreochromis niloticus)*** were divided to five groups,viz.,a control and four treatment groups of low-dose EtBr(10μg/L),low-dose EtBr with SP(10μg/L EtBr+200 mg/L SP),high-dose EtBr(100μg/L),and high-dose EtBr with SP(100μg/L EtBr+200 mg/L SP);the exposure period was 2 *** and high doses of EtBr induced alterations in some hematological,biochemical,and histopathological *** hepatocytes,degenerated area,vacuolated hepatocytes,pyknotic nuclei,constricted and dilated blood sinusoids,and infiltration of inflammatory cells were *** peroxidation concentration was not significantly different in groups exposed to low doses of EtBr and EtBr with SP,but it was increased in groups exposed to high doses of EtBr and EtBr with SP,compared with the control *** feeding with SP,most histological and histochemical parameters restored to normal ***,SP may possess the ability to preserve the structural integrity of the hepatic and renal membranes.

Ciliates in the class Prostomatea play an important role in the global microbial loop due to their signifcant abundances and broad feeding strategies at the foundation of food *** their importance in ecosystems,the taxonomy and systematics of this group of ciliates has long been poorly understood,with this being especially true for members of the family *** we examine four lagynusids collected from sandy beaches in China,using silver-staining and 18S rRNA gene sequencing *** investigations revealed two new genera and two new species and provided details for two little known forms:Penardella marina ***.,***.,Apolagynus cucumis(as reported by Penard.Études sur les infusoires d’eau *** and Cie,Genève,1922)***.,***.,Lagynus minutus ***.,and Lagynus elegans(Engelmann in Z Wiss Zool 11:347-393,1862)Quennerstedt(Acta Univ Lund 4:1-48,1867).Penardella *** be morphologically distinguished by having more than three dikinetidal perioral *** *** from the closely related genus Lagynus in the absence of a conspicuous neck-like *** ciliature of Apolagynus cucumis is revealed here for the frst time,which demonstrates the classifcation of this species within ***,Apolagynus binucleatus(Jiang et al.,2021)*** established according to the new *** results of our phylogenetic analyses based on the 18S rRNA gene support the establishment of two new genera and indicate that Lagynusidae is monophyletic,which further strengthens its valid taxonomic status.

Effect of Temperature on the Carbon, Nitrogen, and Phosphorus Nutrient Budgets of Steelhead Trout (Oncorhynchus mykiss) with Different Sizes

This study was performed to evaluate the effect of temperature on the carbon(C),nitrogen(N),and phosphorus(P)nu-trient budgets of adult steelhead trout(Oncorhynchus mykiss)with different body sizes.A complete two-factor experiment was con-ducted at four temperatures,i.e.,9(T9),13(T13),16(T1℃℃℃6),and 19(T19);and three body sizes,℃i.e.,0.65 kg±0.02 kg(S1),1.22 kg±0.06 kg(S2),and 2.05 kg±0.09 kg(S3).The following properties of steelhead trout were determined:C,N,and P nutrient budgets;feed intake rate;fecal production rate;oxygen consumption rate;ammonia excretion rate;phosphorus excretion rate;scope of the growth of C,N,and P;and absorption efficiency of C,N,and *** showed no significant interaction between temperature and body size in terms of the nutrient budgets of steelhead ***,temperature and body size played important separate roles in the nutrient budgets except for the absorption efficiency of *** feed intake rate,fecal production rate,ammonia excretion rate,phosphorus excretion rate,and scope of the growth of C,N,and P were the highest in the T16 *** absorption effi-ciency of C and P decreased as temperature *** multiple regression results indicated that 15 was the approximate℃optimal temperature for the feeding and growth of steelhead *** feed intake rate,fecal production rate,oxygen consumption rate,ammonia excretion rate,phosphorus excretion rate,and scope of the growth of C,N,and P per unit of the body weight of fish decreased as body size *** absorption efficiency of C increased as body weight increased,and the absorption efficiencies of P in the S2 and S3 treatments were significantly higher than those in the S1 *** the culture of steelhead trout,the daily ratio should be adjusted with changes in the temperature and body size of ***,managers should focus on the risk of hypoxia at high water temperatures.

The potential of ocean acidification on suppressing larval development in the Pacific oyster Crassostrea gigas and blood cockle Area inflata Reeve

We evaluated the effect of pH on larval development in larval Pacific oyster(Crassostrea gigas) and blood cockle(Arca inflata Reeve).The larvae were reared at pH 8.2(control),7.9,7.6,or 7.3beginning 30 min or 24 h post *** to lower pH during early embryonic development inhibited larval shell formation in both *** with the control,larvae took longer to reach the D-veliger stage when reared under pH 7.6 and *** to lower pH immediately after fertilization resulted in significantly delayed shell formation in the Pacific oyster larvae at pH 7.3 and blood cockle larvae at pH 7.6 and ***,when exposure was delayed until 24 h post fertilization,shell formation was only inhibited in blood cockle larvae reared at pH ***,the early embryonic stages were more sensitive to acidified *** results suggest that ocean acidification will have an adverse effect on embryonic development in *** the effects appear subtle,they may accumulate and lead to subsequent issues during later larval development.

Anaerobic protists in general,and ciliates in particular,are important components of anoxic or hypoxic environments,however,their diversity remains *** is a poorly studied genus that is distributed worldwide and is commonly found in anaerobic *** the present study,the taxonomy and phylogeny of three new species,namely Sonderia aposinuata ***.,Sonderia paramacrochilus *** Sonderia steini ***.,collected from China,were investigated based on microscopic observations and SSU rRNA gene sequencing *** aposinuata *** diagnosed mainly by having a relatively large body size,a crescent-shaped oral opening,numerous slender extrusomes,one suture on the ventral side and two on the dorsal side,and a buccal cavity that occupies the anterior third of the *** paramacrochilus *** resembles *** but differs mainly by its oral opening being located closer to the anterior cell margin and its spindle-shaped *** steini *** a freshwater species that can be recognized by its shallow buccal cavity,sparsely distributed rod-shaped extrusomes,and having 68–79 monokinetidal somatic kineties that form sutures on both sides of the *** analyses based on small subunit ribosomal RNA(SSU rRNA)gene sequence data support the monophyly of the family Sonderiidae,however,Sonderia is *** genus Sonderia is briefly revised and a key to the identification of species belonging to this genus is supplied.
