Glucose metabolic phenotype of pancreatic cancer

AIM: To construct a global "metabolic phenotype" of pancreatic ductal adenocarcinoma(PDAC) reflecting tumour-related metabolic enzyme ***: A systematic review of the literature was performed using Ovid SP and Pub Med databases using keywords "pancreatic cancer" and individual glycolytic and mitochondrial oxidative phosphorylation(MOP) enzymes. Both human and animal studies investigating the oncological effect of enzyme expression changes and inhibitors in both an in vitro and in vivo setting were included in the review. Data reporting changes in enzyme expression and the effects on PDAC cells, such as survival and metastatic potential, were extracted to construct a metabolic phenotype. RESULTS: Seven hundred and ten papers were initially retrieved, and were screened to meet the review inclusion criteria. 107 unique articles were identified as reporting data involving glycolytic enzymes, and 28 articles involving MOP enzymes in PDAC. Data extraction followed a pre-defined protocol. There is consistent over-expression of glycolytic enzymes and lactate dehydrogenase in keeping with the Warburg effect to facilitate rapid adenosine-triphosphate production from glycolysis. Certain isoforms of these enzymes were over-expressed specifically in PDAC. Altering expression levels of HK, PGI, FBA, enolase, PK-M2 and LDA-A with metabolic inhibitors have shown a favourable effect on PDAC, thus identifying these as potential therapeutic targets. However, the Warburg effect on MOP enzymes is less clear, with different expression levels at different points in the Krebs cycle resulting in a fundamental change of metabolite levels, suggesting that other essential anabolic pathways are being stimulated. CONCLUSION: Further characterisation of the PDAC metabolic phenotype is necessary as currently there are few clinical studies and no successful clinical trials targeting metabolic enzymes.

The flow characteristics of a dual fluidised bed gasifier(DFBG)are more complex than those of a single fluidised bed *** stable operation and appropriate control,a cold DFBG test facility with both an upper and a lower U-valve was built,and electrical capacitance tomography(ECT)sensors were installed with pressure transducers to investigate the effects of operating conditions on gas-solids flow *** operating parameters included gas velocities in the riser and in the bubbling fluidised bed,aeration velocity in the lower U-valve,bed material inventory,and particle *** is the first time that ECT was applied in different flow zones of a dual fluidised bed gasifier *** experimental results indicated that ECT in the recycle chamber could monitor the performance of the lower U-valve under different operating conditions for early detection of gas shortcut from the riser to the bubbling *** main flow regimes in the riser and the differences between the reactors were identified by two sets of ECT sensors with pressure ***,the effects of the operating conditions on the pressure drop in different parts of the DFBG was investigated.

A customized model for 3D human segmental kinematic coupling analysis by optoelectronic stereophotogrammetry

The study of three-dimensional human kinematics has significant impacts on medical and healthcare technology innovations. As a non-invasive technology, optoelectronic stereophotogrammetry is widely used for in-vivo locomotor evaluations. However, relatively high testing difficulties, poor testing accuracies, and high analysis complexities prohibit its further employment. The objective of this study is to explore an improved modeling technique for quantitative measurement and analysis of human locomotion. Firstly, a 3D whole body model of 17 rigid segments was developed to describe human locomotion. Subsequently, a novel infrared reflective marker cluster for 17 body segments was constructed to calibrate and record the 3D segmental position and orientation of each functional body region simultaneously with high spatial accuracy. In addition, the novel calibration procedure and the conception of kinematic coupling of human locomotion were proposed to investigate the segmental functional characteristics of human motion. Eight healthy male subjects were evaluated with walking and running experiments using the Qualisys motion capture system. The experimental results demonstrated the followings: (i) The kinematic coupling of the upper limbs and the lower limbs both showed the significant characteristics of joint motion, while the torso motion of human possessed remarkable features of segmental motion; (ii) flexion/extension was the main motion feature in sagittal plane, while the lateral bending in coronal plane and the axial rotation in transverse plane were subsidiary motions during an entire walking cycle regarding to all the segments of the human body; (iii) compared with conventional methods, the improved techniques have a competitive advantage in the convenient measurement and accurate analysis of the segmental dynamic functional characteristics during human locomotion. In vitro investigations on the effects of graphene and graphene oxide on polycaprolactone bone tissue engineering scaffolds

Polycaprolactone(PCL)scaffolds that are produced through additive manufacturing are one of the most researched bone tissue engineering structures in the *** to the intrinsic limitations of PCL,carbon nanomaterials are often investigated to reinforce the PCL *** several studies that have been conducted on carbon nanomaterials,such as graphene(G)and graphene oxide(GO),certain challenges remain in terms of the precise design of the biological and nonbiological properties of the *** paper addresses this limitation by investigating both the nonbiological(element composition,surface,degradation,and thermal and mechanical properties)and biological characteristics of carbon nanomaterial-reinforced PCL scaffolds for bone tissue engineering *** showed that the incorporation of G and GO increased surface properties(reduced modulus and wettability),material crystallinity,crystallization temperature,and degradation ***,the variations in compressive modulus,strength,surface hardness,and cell metabolic activity strongly depended on the type of ***,a series of phenomenological models were developed based on experimental results to describe the variations of scaffold’s weight,fiber diameter,porosity,and mechanical properties as functions of degradation time and carbon nanomaterial *** results presented in this paper enable the design of three-dimensional(3D)bone scaffolds with tuned properties by adjusting the type and concentration of different functional fillers.

Influence of Filler on the Microstructure, Mechanical Properties and Residual Stresses in TIG Weldments of Dissimilar Titanium Alloys

The influence of titanium alloy(Ti–5 Al–2.5 Sn) and commercially pure titanium(cp Ti) as fillers on dissimilar pulsed tungsten inert gas weldments of Ti–5 Al–2.5 Sn/cp Ti was investigated in terms of microstructure, mechanical/nano-mechanical properties, and residual stresses. A partial martensitic transformation was observed in the weldments for all the welding conditions due to high heat input. The microstructure evolved in the FZ/cp Ti interfacial region was observed to be the most sensitive to the proportion of α stabilizer in the filler alloy. Furthermore, the addition of filler alloy improved the tensile properties and nano-mechanical response of the weld joint owing to the increased volume of metal in the weld joint. As compared to the Ti–5 Al–2.5 Sn wire, the use of cp Ti filler wire proved to be better in terms of energy absorbed during tensile and impact tests, tensile strength and ductility of the dissimilar welds. An asymmetrical residual stresses profile was observed close to the weld centerline, with high compressive stresses on the Ti–5 Al–2.5 Sn side for both the weldments obtained with and without filler wires. This was attributed to mainly the low thermal conductivity of Ti–5 Al–2.5 Sn. The presence of residual stresses also influenced the nano-hardness profile across the weldments.
