Comparative analysis of goat and bovine milk proteins in the improvement of sarcopenia based on host-microbial interactions

Introduction: Diet intervention， especially supplementation with high-quality protein， is considered to be a critical strategy in sarcopenia. However， different sources and types of protein have different health impacts. Objectives: The aim of this study is to explore the differences in the ameliorative effects and mechanisms of different sources and types of proteins on sarcopenia， providing an optimal path for the prevention and treatment of sarcopenia. Methods: A sarcopenia model was established by intraperitoneal injection of dexamethasone (5 mg/kg). Sixty male C57BL/6 mice (8 months old) were randomly divided into the normal control， sarcopenia， goat whey protein， goat milk casein， bovine whey protein， and bovine milk casein groups. Animals were treated for eight consecutive weeks. Organism-level and molecular phenotypes， 16S rRNA gene sequencing， and untargeted metabolomics profiling based on GC-TOF/MS were employed to investigate the correlation between host metabolism， microbial metabolism， autophagy and inflammation and their influence on sarcopenia in C57BL/6 male mice. Results: All four proteins increased muscle mass， and goat whey protein improved muscle strength in sarcopenic mice. Goat and bovine milk proteins promoted muscle regeneration by increasing MyoD1 and MyoG expression， and the former had a more distinct effect in inducing autophagy and decreasing inflammation than the latter. In addition， goat whey protein and casein could modulate host-microbial arginine co-metabolism. Notably， goat milk proteins responded well to sarcopenia comorbidities， including sarcopenic obesity， osteosarcopenia， and osteoarthritis. Conclusions: The study confirmed that goat milk proteins were more effective than bovine milk proteins for the control of sarcopenia. Moreover， we found that whey protein and casein could modulate host-microbial arginine co-metabolism， which shows their potential as precision nutritional supplements for the management of sarcopenia. Ambient ozone(O3)exposure may be associated with a reduction of semen quality,yet the potential biological mechanisms remain *** investigated the effects of certain seminal plasma metabolites on mediating the links between O3 exposure and the deterioration of semen *** untargeted metabolomics analysis was performed on semen samples of 200 Chinese adult men to determine candidate metabolites associated with characteristics of semen *** analysis was adopted to examine whether these metabolites modulated the links between O3 exposure and semen *** found a significant reduction in sperm concentration by−28.1%(95%CI:−41.7%,−11.3%),and sperm count by−29.2%(95%CI:−43.7%,−11.0%)associated with each 10μg/m3 increase in ambient O3 concentration during the period of sperm *** delineated 7 metabolites in seminal fluid that substantially mediated the links between O3 exposure and declined semen quality,including myristoleic acid,aspartyl-isoleucine,phenylethyl primeveroside,ACar(18:2),ACar(18:1),FAHFA(22:6/22:3),and LPS(22:5).Among these,myristoleic acid exhibited the most pronounced mediation effects,with its indirect effect of which accounts for 46.4%of the overall *** findings suggested that exposure to ozone decreased sperm quality by disrupting fatty acid metabolism,particularly myristoleic acid.

Green-blue spaces(GBS)are pivotal in mitigating thermal ***,their management lacks guidelines rooted in epidemiological evidence for specific planning and *** we show how various GBS types modify the link between non-optimal temperatures and cardiovascular mortality across different thermal *** merged fine-scale population density and GBS data to create novel GBS exposure index.A case time series approach was employed to analyse temperature-cardiovascular mortality association and the effect modifications of type-specific GBSs across 1085 subdistricts in south-eastern *** findings indicate that both green and blue spaces may significantly reduce high-temperature-related cardiovascular mortality risks(e.g.,for low(5%)***(95%)level of overall green spaces at 99th *** mortality temperature(MMT),Ratio of relative risk(RRR)=1.14(95%CI:1.07,1.21);for overall blue spaces,RRR=1.20(95%CI:1.12,1.29)),while specific blue space types offer protection against cold temperatures(e.g.,for the rivers at 1st vs MMT,RRR=1.17(95%CI:1.07,1.28)).Notably,forests,parks,nature reserves,street greenery,and lakes are linked with lower heat-related cardiovascular mortality,whereas rivers and coasts mitigate cold-related cardiovascular *** spaces provide greater benefits than green *** severity of temperature extremes further amplifies GBS's protective *** study enhances our understanding of how type-specific GBS influences health risks associated with non-optimal temperatures,offering valuable insights for integrating GBS into climate adaptation strategies for maximal health benefits.

Tetrabromobisphenol A(TBBPA)and its derivatives widely exist in various environments and *** the available data indicate that TBBPA exposure is highly associated with the increased incidence of endometrial cancer(EC),the effects of TBBPA and its main derivatives on EC proliferation and the involved crucial mechanism remain *** present study aimed to investigate the effects of TBBPA and its derivatives under environmental concentrations on the proliferation of EC,and the crucial mechanism on the progression of EC caused by bromine fame retardants *** this research,TBBPA and two of the most common TBBPA derivatives including TBBPA bis(2-hydroxyethyl ether)(TBBPABHEE)and TBBPA bis(dibromopropyl ether)(TBBPA-BDBPE)were screened for their capacities in induced EC proliferation and explored the related mechanism by in vitro cell culture model and in vivo mice *** environmental concentrations,TBBPA promoted the proliferation of EC,the main derivatives of TBBPA(TBBPA-BHEE and TBBPA-BDBPE)did not present the similar facilitation *** ubiquitination degradation of p53 was crucial in TBBPA induced EC proliferation,which resulted in the increase of downstream cell cycle and decrease of *** further molecular docking result suggested the high affinity between TBBPA and ubiquitinated *** finding revealed the effects of TBBPA and its derivatives on EC proliferation,thus providing novel insights into the underlying mechanisms of TBBPA-caused EC.
