Neuronal activity controls the development of interneurons in the somatosensory cortex

BACKGROUND： Neuronal activity in cortical areas regulates neurodevelopment by interacting with defined genetic programs to shape the mature central nervous system. Electrical activity is conveyed to sensory cortical areas via intracortical and thalamocortical neurons, and includes oscillatory patterns that have been measured across cortical regions. OBJECTIVE： In this work, we review the most recent findings about how electrical activity shapes the developmental assembly of functional circuitry in the somatosensory cortex, with an emphasis on intemeuron maturation and integration. We include studies on the effect of various neurotransmitters and on the influence of thalamocortical afferent activity on circuit development. We additionally reviewed studies describing network activity patterns. METHODS： We conducted an extensive literature search using both the PubMed and Google Scholar search engines. The following keywords were used in various iterations：＂intemeuron＂, ＂somatosensory＂, ＂development＂, ＂activity＂, ＂network patterns＂, ＂thalamocortical＂, ＂NmdA receptor＂, ＂plasticity＂. We additionally selected papers known to us from past reading, and those recommended to us by reviewers and members of our lab. RESULTS： We reviewed a total of 132 articles that focused on the role of activity in interneuronal migration, maturation, and circuit development, as well as the source of electrical inputs and pattems of cortical activity in the somatosensory cortex. 79 of these papers included in this timely review were written between 2007 and 2016. CONCLUSIONS： Neuronal activity shapes the developmental assembly of functional circuitry in the somatosensory cortical interneurons. This activity impacts nearly every aspect of development and acquisition of mature neuronal characteristics, and may contribute to changing phenotypes, altered transmitter expression, and plasticity in the adult. 背景:研究发现,过早绝经的女性患痴呆的风险加倍,死干神经功能障碍的风险也增加5倍,但其分子机制还不明确。本研究试图探讨卵巢1 7β-雌二醇(E2)持续降低是否可能促进海马区局部缺血引起的淀粉状蛋白生成。方法:用已建造的早期手术性更年期大鼠模型(双侧卵巢切除术10周),在手术后立即给予E2(短期E2缺乏,STED)或卵巢切除术后期给予E2(长期E2缺乏,LTED)。在连续1周皮下注射E2后,对于动物进行全脑缺血10 min,再评价LTED对缺血诱导的海马CA1区淀粉样蛋白生成的作用。结果:海马区β淀粉样蛋白(Aβ)水平在全脑缺血后没有明显增加,而LTED雌鼠在全脑缺血后内源性Aβ水平快速稳健提高。在STED的雌鼠,我们观察到全脑缺血后减少ADAM10在海马CA1区的表达,而E2治疗能保持ADAM10在海马CA1区的表达水平。同时,全脑缺血后引起BACEl的增强,而E2可以在相同脑区域降低BACE1水平。然而,我们在LTED雌鼠观察到E2治疗失去了在海马CA1区对ADAM 10、ADAM 17和BACE1水平的调节,说明早期内源性雌激素长期缺乏会加重脑缺血后导致的Aβ-生成。同时,我们还观察到,LTED雌鼠全脑缺血后,E2还失去了对tau蛋白高度磷酸化的调节,为体内雌激素水平下降和脑缺血所导致的粉状蛋白增加的机制提供了证据。结论:本研究部分解释了过早绝经的妇女有高风险患痴呆和过早死亡的机制,并认为及时补充E2会使得神经系统得到最大的保护作用。

Driver mutations of cancer epigenomes

Epigenetic alterations are associated with all aspects of cancer, from tumor initiation to cancer progression and metastasis. It is now well understood that both losses and gains of DNA methylation as well as altered chro- matin organization contribute significantly to cancer- associated phenotypes. More recently, new sequencing technologies have allowed the identification of driver mutations in epigenetic regulators, providing a mecha- nistic link between the cancer epigenome and genetic alterations. Oncogenic activating mutations are now known to occur in a number of epigenetic modifiers （i.e. IDHll2, EZH2, DNMT3A）, pinpointing epigenetic path- ways that are involved in tumorigenesis. Similarly, investigations into the role of inactivating mutations in chromatin modifiers （i.e. KDM6A, CREBBPIEP300, SMARCB1） implicate many of these genes as tumor suppressors. Intriguingly, a number of neoplasms are defined by a plethora of mutations in epigenetic regu- lators, including renal, bladder, and adenoid cystic car- cinomas. Particularly striking is the discovery of frequent histone H3.3 mutations in pediatric glioma, a particularly aggressive neoplasm that has long remained poorly understood. Cancer epigenetics is a relatively new, promising frontier with much potential for improving cancer outcomes. Already, therapies such as 5-azacytidine and decitabine have proven that targeting epigenetic alterations in cancer can lead to tangible benefits. Understanding how genetic alterations give rise to the cancer epigenome will offer new possibilities for developing better prognostic and therapeutic strategies.

Discovered in pig brains in 1982,the brain-derived neurotrophic factor(BDNF)is one of the most studied and characterized neurotrophins in the central nervous system.In recent years,BDNF has received considerable attention for its importance in the development and maintenance of normal brain function.This is because BDNF plays an important role in crucial functions of the nervous system,such as the survival,differentiation,and maturation of neurons and glial cells as well as the actions of neuroprotection in adverse conditions,such as glutamatergic overstimulation,cerebral ischemia,hypoglycemia,and neurotoxicity(Kowiański et al.,2018).

Androgenic alopecia is a frequent condition. In western countries, more than 90% of hair loss consultaitons in men are motivated by such condition. In both males and females, the incidence of alopecia increases with age. During a French national program (called SUVIMAX) done on 4050 men, aged between 42 and 64 years and 6231 women aged between 35 and 60 years, we have collected numerous data on hair and scalp features on the whole cohort. Based on the Hamilton scale, the prevalence of androgenic alopecia (AGA) is significantly lower in the South Eastern region of France with 40% of men without alopecia in comparison to an average of 32.4% ir the rest of France. The significant lower prevalence in the South Eastern region could be linked to nutritional habits.

Acute myeloid leukemia(AML)is the most common form of acute leukemia in adults and the second most common form of acute leukemia in children.Despite this,very little improvement in survival rates has been achieved over the past few decades.This is partially due to the heterogeneity of AML and the need for more targeted therapeutics than the traditional cytotoxic chemotherapies that have been a mainstay in therapy for the past 50 years.In the past 20 years,research has been diversifying the approach to treating AML by investigating molecular pathways uniquely relevant to AML cell proliferation and survival.Here we review the development of novel therapeutics in targeting apoptosis,receptor tyrosine kinase(RTK)signaling,hedgehog(HH)pathway,mitochondrial function,DNA repair,and c-Myc signaling.There has been an impressive effort into better understanding the diversity of AML cell characteristics and here we highlight important preclinical studies that have supported therapeutic development and continue to promote new ways to target AML cells.In addition,we describe clinical investigations that have led to FDA approval of new targeted AML therapies and ongoing clinical trials of novel therapies targeting AML survival pathways.We also describe the complexity of targeting leukemia stem cells(LSCs)as an approach to addressing relapse and remission in AML and targetable pathways that are unique to LSC survival.This comprehensive review details what we currently understand about the signaling pathways that support AML cell survival and the exceptional ways in which we disrupt them.

The Institute for the History of Natural Sciences in Beijing celebrated the sixtieth anniversary of its founding in 1957 with an international symposium devoted to“China and the World in the Global History of Science and Technology.”2 This contribution to that symposium focuses on the roles that historians of mathematics,East and West,have played in bringing the importance of mathematics in China to the attention of the world at large.It concludes by discussing a controversy surrounding the diagram and purported“proof”given in the ancient mathematical classic,the Zhoubi suanjing周髀算經(Mathematical classic of Zhou’s gnomon),of the gou-gu theorem that appeared in volume 3 of Joseph Needham’s Science and Civilisation in China,published in 1959.

Comprehensive Flow Cytometry Analysis of PEI-Based Transfections for Virus-Like Particle Production

The generation of stable clones for biomolecule production is a common but lengthy and labor-intensive process.For complex molecules,such as viruses or virus-like particles(VLPs),the timeline becomes even more cumbersome.Thus,in the early stages of development,transient production methods serve as a reasonable alternative to stable clone construction.In this work,an investigation of a polyethylenimine-(PEI-)based transfection method for the transient production of Chikungunya(Chik)VLPs,a vaccine candidate molecule,was undertaken.This effort focuses on tracking cell population responses during transfection,understanding how process changes affect these responses,and monitoring patterns in cell performance over the culture duration.Plasmid labeling and VLP staining were employed to comprehensively track cells via flow cytometry and to draw correlations between plasmid DNA(pDNA)uptake and the resulting VLP expression.The method detected high transfection efficiency(≥97%)in all samples tested and demonstrated the capability to track kinetics of plasmid-cell binding.With varied transfection cell concentrations,the pDNA binding kinetics are altered and saturation binding is observed in the lowest cell concentration sample tested in less than 3 hours of incubation.Interestingly,in all samples,the flow cytometry analysis of relative pDNA amount versus VLP expression staining showed that cells which contained fewer pDNA complexes resulted in the highest levels of VLP stain.Finally,to determine the potential breadth of our observations,we compared daily expression patterns of ChikVLP with a reporter,monomeric GFP molecule.The similarities detected suggest the interpretations presented here to likely be more broadly informative and applicable to PEI-based transient production of additional biological products as well.
