This article presents and analyses the modular architecture and capabilities of CODE-DE(Copernicus Data and Exploitation Platform–Deutschland,***),the integrated German operational environment for accessing and processing Copernicus data and products,as well as the methodology to establish and operate the *** March 2017,CODE-DE has been online with access to Sentinel-1 and Sentinel-2 data,to Sentinel-3 data shortly after this time,and since March 2019 with access to Sentinel-5P *** products are available and accessed by 1,682 registered users as of March *** this period 654,895 products were downloaded and a global catalogue was continuously updated,featuring a data volume of 814 TByte based on a rolling archive concept supported by a reload mechanism from a long-term *** November 2017,the element for big data processing has been operational,where registered users can process and analyse data themselves specifically assisted by methods for value-added product *** 195,467 core and 696,406 memory hours,982,948 products of different applications were fully automatically generated in the cloud environment and made available as of March *** features include an improved visualization of available Sentinel-2 products,which are presented within the catalogue client at full 10 m resolution.

In this White Paper we present the potential of the Enhanced X-ray Timing and Polarimetry(eXTP) mission for determining the nature of dense matter; neutron star cores host an extreme density regime which cannot be replicated in a terrestrial laboratory. The tightest statistical constraints on the dense matter equation of state will come from pulse profile modelling of accretion-powered pulsars, burst oscillation sources, and rotation-powered pulsars. Additional constraints will derive from spin measurements, burst spectra, and properties of the accretion flows in the vicinity of the neutron star. under development by an international Consortium led by the Institute of High Energy Physics of the Chinese Academy of Sciences, the eXTP mission is expected to be launched in the mid 2020 s.

Two 21 d-experiments were conducted to determine the optimum standardized ileal digestible(SID)threonine:lysine ratio(Thr:Lys) for weaned piglets reared under clean(Exp. 1) or unclean(Exp. 2) sanitary conditions and fed antibiotic-free diets. In each experiment, 90 mixed-sex pigs(Duroc × [Yorkshire × Landrace]; initial BW 7.2 ± 0.3 kg) were randomly assigned to 5 dietary treatments each with 6 replicates(3 pigs per pen). The dietary treatments were 5 graded levels of SID Thr:Lys(55,59, 63, 67 and 71%). Diets were corn-wheat-soybean meal-based with a constant SID Lys of 1.18% that was set to be second limiting amino acid. In Exp. 1 and Exp. 2, plasma-free Thr increased(P = 0.05) with increasing dietary SID Thr:Lys. In Exp. 1, the SID Thr:Lys for gain-to-feed ratio(G:f) was optimized at 65%.In Exp. 2, the estimated optimal SID Thr:Lys for overall G:f was 66.5%. In conclusion, an average optimal SID Thr:Lys of 65 and 66.5% could be used to optimize feed efficiency for weaned pigs under clean and unclean sanitary conditions, respectively.

Determination of Calcineurin Inhibitors in Dried Blood Spots from Kidney Transplant recipients

Background: Determination of cyclosporine A (CsA) and tacrolimus (Tac) in dried blood spots (DBS) could enable drug monitoring in transplanted patients without the necessity of having to take venous blood samples. Therefore, we have developed a method for quantitative determination of calcineurin inhibitors (CNI) by liquid-chromatography-tandem mass spectrometry (LCMS). Methods: In a study with 68 kidney transplant recipients (KTr, 34 CsA, 34 Tac), we tested the clinical application of LCMS monitoring in DBS in comparison to LCMS in whole blood. results: The measuring range is proven for 27.33 to 1345 ng/ml for CsA and for 1.63 to 39.7 ng/ml for Tac. The requirements for clinical chemical analyses for precision and accuracy are complied with. Stability is documented for a period of 14 days. The study showed the following deviations from LCMS in whole blood for determination of CsA and Tac in DBS after introducing a correction factor by the haematocrit (Hct) value (CsA trough level: mean = 4.7%, ±1.96 standard deviation (SD) -52.1% to 61.4%, N = 96;CsA peak level: mean = 7.3%, ±1.96 SD -39.7% to 54.4%, N = 95;Tac trough level: mean = -0.5%, ±1.96 SD -76.4% to 75.3%, N = 88;Tac peak level: mean = 3.9%, ±1.96 SD -80.1% to 88.7%, N = 92). Conclusions: Our data show comparable results with the reference method by means of LCMS in whole blood. Therefore, DBS of KTr for determination of CNI levels could be transported on filter cards by mail to the respective laboratory resistant to breakage and the hazard of infection.

AIM： TO explore the role of Bcl-XL and Myeloid cell leukaemia （Mcl）-I for the apoptosis resistance of colorectal carcinoma （CrC） cells towards current treatment modalities. METHODS： Bcl-XL and Mcl-1 mrNA and protein expression were analyzed in CrC cell lines as well as human CrC tissue by Western blot, quantitative PCrand immunohistochemistry. Bcl-XL and Mcl-1 protein expression was knocked down or increased in CrC cell lines by applying specific sirNAs or expression plasmids, respectively. After modulation of protein expression, CrC cells were treated with chemotherapeutic agents, an antagonistic epidermal growth factor receptor （EGfr1） antibody, an EGfr1 tyrosine kinase inhibitor, or with the death receptor ligand TrAIL. Apoptosis induction and cell viability were analyzed. rESULTS： Here we show that in human CrC tissue and various CrC cell lines both Bcl-xL and Mcl-1 are expressed. Bcl-xL expression was higher in CrC tissue than in surrounding non-malignant tissue, both on protein and mrNA level. Mcl-1 mrNA expression was significantly lower in malignant tissues. However, protein expression was slightly higher. Viability rates of CrC cells were significantly decreased after knock down of Bcl-xL expression, and, to a lower extent, after knock down of Mcl-1 expression. furthermore, cells with reduced Bcl-xL or Mcl-1 expression was more sensitive towards oxaliplatinand irinotecan-induced apoptosis, and in the case of Bcl-xL also towards 5-fU-induced apoptosis. On the other hand, upregulation of Bcl-xL by transfection of an expression plasmid decreased chemotherapeutic drug-induced apoptosis. EGf treatment clearly induced Bcl-xL and Mcl-1 expression in CrC cells. Apoptosis induction upon EGfr1 blockage by cetuximab or PD168393 was increased by inhibiting Mcl-1 and Bcl-xL expression. More strikingly, CD95- and TrAIL-induced apoptosis was increased by Bcl-xL knock down. CONCLUSION： Our data suggest that Bcl-xL and, to a lower extent, Mcl-1, are important anti-apoptotic
