Subcellular m.tochondria serve as sensors for energy m.tabolism.and redox balance, and the dynam.c regulation of functional and dysfunctional m.tochondria plays a crucial role in determ.ning cells' fate. Selective rem.val of dysfunctional m.tochondria at the subcellular level can provide chondrocytes with energy to prevent degeneration, thereby treating osteoarthritis. Herein, to achieve an ideal subcellular therapy, cartilage affinity peptide (WYRGRL)-decorated liposom.s loaded with m.tophagy activator (urolithin A) were integrated into hyaluronic acid m.thacrylate hydrogel m.crospheres through m.crofluidic technology, nam.d Hm.WY-Lip/UA, that could efficiently target chondrocytes and selectively rem.ve subcellular dysfunctional m.tochondria. As a result, this system.dem.nstrated an advantage in m.tochondria function restoration, reactive oxygen species scavenging, cell survival rescue, and chondrocyte hom.ostasis m.intenance through increasing m.tophagy. In a rat post-traum.tic osteoarthritis m.del, the intra-articular injection of Hm.WY-Lip/UA am.liorated cartilage m.trix degradation, osteophyte form.tion, and subchondral bone sclerosis at 8 weeks. Overall, this study indicated that Hm.WY-Lip/UA provided a protective effect on cartilage degeneration in an efficacious and clinically relevant m.nner, and a m.tochondrial-oriented strategy has great potential in the subcellular therapy of osteoarthritis.

Effect of Sm.ll Needle Knife on Autophagy and TGF-β in Synovial Fluid of KOA Rats

Objective:To explore the effects of sm.ll needle knife on the autophagy function of synovium.and the expression of TGF-βin synovial fluid of KOA rats,so as to provide a new theoretical basis for the treatm.nt of ***:48 SD rats were random.y divided into blank control group,m.del group,drug group and acupotom. *** blank control group m.intained the original feeding without any intervention,the m.del group adopted the sodium.m.noiodoacetate injection m.thod to prepare the KOA rat m.del without any intervention,the drug group prepared the KOA rat m.del and then treated with tripterine by gavage,and the needle knife group treated the KOA rat m.del with sm.ll needle *** expression levels of autophagy-related proteins in synovial tissues were detected by im.unohistochem.stry and Western blotting,and the content of TGF-βin synovial fluid was m.asured by ***:Com.ared with the blank control group,Beclin-1,Caspase-3 and Bax were up-regulated and m.TOR was down-regulated in the m.del ***-1,m.TOR,Caspase-3 and Bax were up-regulated in the drug *** was no significant difference in the expression of autophagy-related proteins Beclin-1(0.28±0.09)and m.TOR(0.45±0.12)between the acupotom. group and the blank control group,while Caspase-3(0.17±0.03)and Bax(0.30±0.01)were *** expression level of TGF-βin synovial fluid in the sm.ll needle knife group was(0.29±0.09),which was sim.lar to that in the blank control group,and was significantly higher than that in the m.del ***:Sm.ll needle knife treatm.nt can prom.te the recovery of synovial autophagy in KOA rats,and reduce the level of TGF-βin synovial fluid,and can play a clinical role in the treatm.nt of KOA by regulating the expression of synovial autophagy and TGF-β.

基于酶的生化检测方法具有准确性高、检测速度快等优势,但生物酶的不稳定性限制了这一方法的应用.因此,开发一种活性高、稳定性强、反应效率高的酶包封体系是提高胆固醇生化检测的关键.为此,本研究发展了由二棕榈酸磷脂酰胆碱包覆中空多壳层结构的仿生人造细胞,Hom.的区室化结构和磷脂膜的选择透过性实现了酶的稳定包封和信号的高效传输.本研究通过信号的级联放大实现了胆固醇的快速准确检测.即利用负载胆固醇氧化酶的Hom.@DPPC引发胆固醇氧化反应,产生的中间产物H;2;O;2;作为信号分子,在负载辣根过氧化物酶的Hom.@DPPC催化作用下,促进荧光红染料生成试卤灵,再进行比色检测胆固醇的含量.由级联催化介导的胆固醇信号放大使得该系统可以应用于胆固醇检测,并且该方法表现出高的选择性和准确性,检测范围为0.05~0.3m.,检测限可达4.9μm.该方法可用于动物血清中胆固醇的检测,回收率为90.8%~104.9%,检测值与商业检测方法的结果高度吻合.此外,该系统可与智能设备集成,构建智能比色平台,实现图像分析与胆固醇浓度检测的一体化,为准确、快速的生化检测开辟了新途径.

Analysis of cohesive particles m.xing behavior in a twin-paddle blender:DEm.and m.chine learning applications

This research paper presents a com.rehensive discrete elem.nt m.thod(DEm.exam.nation of the m.xing behaviors exhibited by cohesive particles within a twin-paddle blender.A com.arative analysis between the sim.lation and experim.ntal results revealed a relative error of 3.47%,dem.nstrating a strong agreem.nt between the results from.the experim.ntal tests and the DEm.*** m.in focus centers on system.tically exploring how operational param.ters,such as im.eller rotational speed,blender's fill level,and particle m.ss ratio,influence the *** investigation also illustrates the significant influence of the m.xing tim. on the m.xing *** gain a deeper understanding of the DEm.sim.lation findings,an analytical tool called m.ltivariate polynom.al regression in m.chine learning is *** m.thod uncovers significant connections between the DEm.results and the operational param.ters,providing a m.re com.rehensive insight into their *** m.ltivariate polynom.al regression m.del exhibited robust predictive perform.nce,with a m.an absolute percentage error of less than 3%for both the training and validation sets,indicating a slight deviation from.actual *** m.del's precision was confirm.d by low m.an absolute error values of 0.0144(80%of the dataset in the training set)and 0.0183(20%of the dataset in the validation set).The study offers valuable insights into granular m.xing behaviors,with im.lications for enhancing the efficiency and predictability of the m.xing processes in various industrial applications.

Rem.deling Tum.r Im.une m.croenvironm.nt by Using Polym.r-Lipid-m.nganese Dioxide Nanoparticles with Radiation Therapy to Boost Im.une Response of Castration-Resistant Prostate Cancer

Despite substantial progress in the treatm.nt of castration-resistant prostate cancer (CRPC), including radiation therapy and im.unotherapy alone or in com.ination, the response to treatm.nt rem.ins poor due to the hypoxic and im.unosuppressive nature of the tum.r m.croenvironm.nt. Herein, we exploited the bioreactivity of novel polym.r–lipid m.nganese dioxide nanoparticles (PLm.s) to rem.del the tum.r im.une m.croenvironm.nt (TIm.) by increasing the local oxygen levels and extracellular pH and enhancing radiation-induced im.unogenic cell death. This study dem.nstrated that PLm. treatm.nt sensitized hypoxic hum.n and m.rine CRPC cells to radiation, significantly increasing radiation-induced DNA double-strand breaks and ultim.tely cell death, which enhanced the secretion of dam.ge-associated m.lecular patterns, attributable to the induction of autophagy and endoplasm.c reticulum.stress. Reoxygenation via PLm.s also polarized hypoxic m.rine RAW264.7 m.crophages toward the m. phenotype, enhancing tum.r necrosis factor alpha release, and thus reducing the viability of m.rine CRPC TRAm.-C2 cells. In a syngeneic TRAm.-C2 tum.r m.del, intravenous injection of PLm.s suppressed, while radiation alone enhanced recruitm.nt of regulatory T cells and m.eloid-derived suppressor cells. Pretreatm.nt with PLm.s followed by radiation down-regulated program.ed death-ligand 1 and prom.ted the infiltration of antitum.r CD8+ T cells and m. m.crophages to tum.r sites. Taken together, TIm. m.dulation by PLm.s plus radiation profoundly delayed tum.r growth and prolonged m.dian survival com.ared with radiation alone. These results suggest that PLm.s plus radiation is a prom.sing treatm.nt m.dality for im.roving therapeutic efficacy in radioresistant and im.unosuppressive solid tum.rs.
