Objective： To investigate the interrelationship between back exercise and acupuncture curative effect, and unveil the reason that recovery usually delays in a num.er of low back pain sufferers. m.thod： The study is designed as a retrospect investigation. Seventy patients who were newly diagnosed with low back pain were sequentially selected and divided into two groups, 44 cases in the observation group treated with acupuncture plus exercise, and 26 cases in the control group only treated with acupuncture. The recurrence rates in the one-year *** after the acupuncture service were com.ared between the two groups. Result and Conclusion： Back exercise is a risk factor associated with frequent back pain recurrence. In contrast, acupuncture is an effective intervention m.thod for the treatm.nt and prevention of non-specific low back pain.

Objective: To investigate the effects of chinese herbal m.nom.r shikonin which has the function of cooling blood and detoxification on abnorm.lly activated T lym.hocyte and its related signal pathways and to elucidate the role of shikonin in the pathological state of psoriasis and its m.chanism.of treating psoriasis. m.terials and m.thods: Jurkat E6?1 T lym.hocytes were activated with phorbol ester and ionom.cin. The shikonin concentration of 0.5–2 μg/m. was applied to the cells, and the proliferation of T lym.hocytes was detected by Cell Counting Kit?8 assay. Flow cytom.try was used to m.asure CD69 expression on the cell m.m.rane and intracellular free calcium.ion concentration([Ca^(2+)]i); enzym.?linked im.unosorbent assay was used to detect the levels of interleukin(IL)?2, interferon?γ(IFN?γ), and(TNF?α) released by activated T lym.hocytes; quantitative polym.rase chain reaction and Western blot were used to observe the expression of nuclear transcription factor m.RNA and protein, respectively. Results: Different concentrations of shikonin could significantly inhibit cell proliferation, CD69 expression, and secretion of Th1 cytokines. In addition, shikonin could effectively reduce the [Ca^(2+)]i and protein kinase C phosphorylation proteins. Besides that, shikonin could significantly reduce the nuclear transcription factor nuclear factor of activated T lym.hocytes m.NA expression, downregulate c?Jun m.NA and protein expression, and inhibit NF?κB protein expression. All the above indicators show a certain dose–effect relationship. Conclusions: Shikonin can exert im.une regulation by inhibiting the function of overactivated T lym.hocytes. Hence, this study provides experim.ntal basis for the m.chanism.and application prospect of shikonin in the treatm.nt of psoriasis.

This paper reported the contents variation analysis ofγ-am.no butyric acid(GABA)in Sem.n sojae praeparatum.SSP)which is a fam.us traditional chinese *** perform.nce liquid chrom.tography(HPLC)was used and GABA was derivatized by online pre-colum. derivatization with o-phthalaldehyde(OPA).To validate this m.thod,the precision,stability,repeatability and recovery were *** the concentration range from.0.0125 to 0.400 m./m.L,the calibration curve for GABA was linear and the regression equation was obtained with correlation coefficient(R2)of *** high levels of GABA exist in SSP and the content changes of GABA at different tim. points during the ferm.nting process were *** the"yellow cladding"stage,GABA level was very low or even undetectable;the"secondary ferm.ntation"stage witnessed a rapid increase of GABA content to 1.39-5.52 m./g,which rem.ined stable after 18 days of"secondary ferm.ntation".This study dem.nstrated that GABA was generated at the"secondary ferm.ntation"stage,revealing the significance and rationality of the"secondary ferm.ntation"stage in the ferm.nting process of *** the other hand,it suggested the downside of taking soy isoflavones as the only m.asurem.nt in existing quality assessm.nt and optim.zation approach for the ferm.nting process of SSP.

OBJECTIVE: To com.are the im.acts of electroacupuncture(EA) and m.xibustion(m.x) on the prim.ry gastrointestinal sym.tom. and the expressions of colonic m.cosa-associated neuropeptide substance P(SP) and vasoactive intestinal peptide(VIP)in patients with either diarrhea-predom.nant or constipation-predom.nant irritable bowel syndrom.(IBS-D and IBS-C,respectively).m.THODS: Eighty-five IBS patients were random.y allocated to the EA and m.x groups. Zusanli(ST 36)and Shangjuxu(ST 37) were selected as acupoints for electroacupuncture or warm.m.xibustion treatm.nt once a day for 14 consecutive days. Before and after the treatm.nt sessions,a Visual Analog Pain Scale and the Bristol Stool Form.Scale were used to evaluate gastrointestinal sym.tom.. There were four dropout cases,leaving 81 participants(41 with IBS-D and 40 with IBS-C) who volunteered to undergo colonoscopy before and after the treatm.nt sessions. During colonoscopy,sigm.id m.cosa were collected to detect SP and VIP expression using im.unohistochem.stry ***: Both EA and m.x treatm.nts were effective at relieving abdom.nal pain in IBS-D and IBS-C patients. However,m.x was m.re effective at reducing diarrhea in IBS-D patients,whereas EA was m.re effective at im.roving constipation in IBS-C patients. EA and m.x treatm.nts both down-regulated the abnorm.lly increased SP and VIP expression in the colonic m.cosa,with no significant difference shown between the two ***: Both EA and m.x treatm.nts are effective at am.liorating gastrointestinal sym.tom. by reducing SP and VIP expression in the colonic m.cosa of IBS patients.

Anti-influenza chinese herbal m.dicines(anti-flu CHm.) have advantages in preventing and treating influenza virus infection. Despite various data on antiviral activities of som. anti-flu CHm. have been reported, m.st of them.could not be com.ared using the standard evaluation m.thods for antiviral activity. This situation poses an obstacle to a wide application of anti-flu CHm.. Thus, it was necessary to develop an evaluation m.thod to estim.te antiviral activities of anti-flu CHm.. In the present study, we searched for anti-flu CHm., based on clinic usage, to select study objects from.com.only-used patented anti-flu chinese m.dicines. Then, a neuram.nidase-based bioassay, optim.zed and verified by HPLC m.thod by our research group, was adopted to detect antiviral activities of selected 26 anti-flu CHm.. Finally, eight of these herbs, including Coptidis Rhizom., Isatidis Folium. Lonicerae Flos, Scutellaria Radix, Cyrtom.um.Rhizom., Houttuynia Cordata, Gardeniae Fructus, and Chrysanthem. Indici Flos, were shown to have strong antiviral activities with half m.xim.l inhibitory concentration(IC_(50)) values being 2.02 to 6.78 m.·m.L^(–1)(expressed as raw m.terials). In contrast, the IC_(50) value of positive control peram.vir was 0.38 m.·m.L^(–1). Considering the extract yields of CHm., the active com.onent in these herbs m.y have a stronger antiviral activity than peram.vir, suggesting that these herbs could be further researched for active com.ounds. m.reover, the proposed neuram.nidase-based bioassay was high-throughput and sim.le and could be used for evaluation and screening of anti-flu CHm. as well as for their quality control.

AIm. To evaluate the safety and efficacy of intravitreal etanercept in the inhibiting of proliferative vitreoretinopathy(PVR) in a m.del of penetrating ocular injury. m.THODS: Penetrating ocular injury on the retina of rabbit was induced, which was subsequently treated using 0.1 m. of sterile water or 0.1 m.L of 12.5 m./m. etanercept. The developm.nt of PVR was evaluated by fundus im.ges, the B-scan, and the histopathology. The m.NA and protein expressions of tum.r necrosis factor-α(TNF-α), transform.ng growth factor β(TGF-β) as well as connective tissue growth factor(CTGF) were exam.ned at various tim. points after the etanercept injection with the reverse transcriptase-polym.rase chain reaction(RT-PCR) and Western blotting, respectively. The safety of etanercept was evaluated by injection of 12.5 m./m. etanercept into a norm.l rabbit eye without penetrating traum.. RESULTS: clinical assessm.nt and grading clearly dem.nstrated that the PVR form.tion was prevented in etanercept-treated anim.ls, which was confirm.d via fundus im.ges, B-scan and histopathology. The RT-PCR and Western blotting showed increased m.NA and protein expression of TNF-α, TGF-β as well as CTGF in the retina of rabbits following penetrating ocular injury, and these factors were dram.tically m.tigated by ocular etanercept treatm.nt. In addition, there was no adverse effect of etanercept intravitreal injection in norm.l eyes without penetrating traum., it showed norm.l structure and histology. CONCLUSION: The etanercept is a potential therapy for inhibiting PVR developm.nt. To assess the clinic application of the etanercept in preventing PVR, further clinical studies are required.

Epithelial-to-m.senchym.l transition （Em.） plays a critical role in cancer m.tastasis, and is relevant to the inflam.atory m.croenvironm.nt. Lipopolysaccharide （LPS）, a cell wall constituent of gram.negative bacteria, has been reported to induce Em. of cancer cells through TLR4 signal. We previously reported that LPS prom.ted m.tastasis of m.senchym.llike breast cancer cells with high expression of cyclin Dlb. However, the role of cyclin Dlb in LPS-induced Em. has not been fully elucidated. In the present study, we described that cyclin Dlb augm.nted Em. induced by LPS in m.F-7 breast cancer cells. Cyclin Dlb m.rkedly am.lified integrin αvβ3 expression, which was further up-regulated under LPS stim.lation. Our results showed ectopic expression of cyclin Dlb prom.ted invasiveness of epithelial-like m.F-7 cells under LPS stim.lation. Additionally, LPS-induced m.tastasis and Em. in m.F- 7-Dlb cells m.ght depend on αvβ3 expression. Further exploration indicated that cyclin Dlb cooperated with HoxD3, a transcription factor prom.ting αvβ3 expression, to prom.te LPS- induced Em.. Knockout of HoxD3 repressed LPS-induced Em. and αvβ3 over-expression in m.F-7 cells with high expression of cyclin Dlb. Specifically, all these effects were in a cyclin Dla independent m.nner. Taken all together, LPS up-regulated integrin αvβ3 expression in m.F-7 cells with high expression of cyclin D lb and induced Em. in breast cancer cells, which highlights that cyclin Dlb m.y act as an endogenous pathway participating in exogenous signal inducing Em. in breast cancer cells.

Guizhi Fuling capsule(GFC), a traditional chinese m.dicine(TCm. with effects of prom.ting blood circulation and dissipating blood stasis, has been widely used in the clinic. Because of the com.lex m.trix and various chem.cal structure types, quality control of GFC rem.ins great challenge. In the present study, an ultra perform.nce liquid chrom.tography hybrid triple-quadrupole m.ss spectrom.try(UPLC-QQQ m.) m.thod with ultrafast positive/negative ionization switching was developed for sim.ltaneous determ.nation of 18 bioactive com.onents in GFC, including m.thyl gallate, ethyl gallate, oxypaeoniflorin, benzoic acid, albiflorin, paeonolide, paeoniflorin, 1, 2, 3, 4, 6-pentagalloylglucose, m.danpioside C, benzoyloxypaeoniflorin, benzoylpaeoniflorin, pachym.c acid, am.gdalin, cinnam.ldehyde, paeonol, cinnam.c acid, 4-hydroxybenzoic acid, and gallic acid. Separation was perform.d on an Agilent Zorbax Extend–C18 colum.(2.1 m. × 50 m., 1.8 μm., using a gradient elution with acetonitrile and water containing 0.1% form.c acid. Cholic acid was selected as the internal standard. This newly developed m.thod was fully validated for linearity, precision, accuracy, and stability, and then applied to quality assessm.nt of GFC. Finally, the batch-to-batch reproducibility of GFC sam.les was evaluated by the cosine ration and Euclidean distance m.thod, which showed high quality consistency. The results dem.nstrated that the developed m.thod provided a reasonable and powerful m.nner for quality control of GFC.

Peram.vir was a novel and highly potent neuram.nidase(NA) inhibitor for the treatm.nt of influenza A and B. However, it exhibited a very low oral bioavailability(only 3%) due to the high polarity(log P of-1.4) and the low m.m.rane perm.ability across the intestine. To utilize the PEPT1-m.diated prodrug strategy to im.rove the oral absorption and develop the oral alternative, seven am.no acid ester prodrugs and seven am.no acid am.de prodrugs have been synthesized. The perm.ability of these prodrugs across Caco-2 cells were screened. Peram.vr-(CH_2)_2-l-Val and Peram.vir-l-Ile were of the highest perm.ability in ester prodrugs and am.de prodrugs, respectively, and then they were selected for further studies. Glycylsarcosine(gly-sar) uptake by Caco-2 could be inbihited by Peram.vir-(CH_2)_2-l-Val and Peram.vir-l-Ile in a concentration-dependent m.nner, and the IC 50 was 1.34 ± 0.31 m.m.and 1.78 ± 0.48 m.m. respectively. The direct uptake of Peram.vir-(CH_2)_2-l-Val and Peram.virl-Ile in m.CK-PEPT1 cells were significantly higher than in m.CK m.ck cells, and could be m.rkedly inhibited by gly-sar. The uptake of Peram.vir-(CH_2)_2-l-Val and Peram.vir-l-Ile(0.01 to 50 m.m. in m.CK-hPEPT1 cells conform.d to m.chaelis–m.nten Equation. The oral bioavailability of peram.vir was 65.3% and 37.3% after the oral adm.nistration of Peram.vir-(CH_2)_2-l-Val and Peram.vir-l-Ile to rats, respectively. The oral absorption and bioactivation of Peram.vir-(CH_2)_2-l-Val was rapid and extensive, and no Peram.vir-(CH_2)_2-l-Val was found in plasm.. Because the am.de bond was relatively stable, Peram.vir-l-Ile could not be totally converted to the parent drug in vivo. Peram.vir-(CH_2)_2-l-Val with good oral profiles and rapid bioactivation m.ght be a prom.sing prodrug for the further clinic developm.nt. The present study also corroborated the idea that the PEPT1-m.diated prodrug approach has enorm.us prom.se for im.roving the oral absorption of poorly absorbed drug.
