PBQ[1-(4-chlorophenyl)-3-(pyridin-3-yl)urea],an enormous potent molluscicide,showed excellent Pomacea canaliculata(***)control activity and low toxicity for other aquatic organisms,but its snail-killing mechanisms are still not fully *** employed an optical method to elucidate PBQ action via a novel fluorescent viscosity probe,*** the viscosity in the test solutions increased,compared with that in pure ethanol,a 54-fold fluorescence intensity enhancement of NCV was observed in 310 cP of 90%***,NCV successfully exhibited a selective fluorescence response towards monensin-induced cellular viscosity changes in HepG2 *** liver,stomach,and foot plantar of the tested snails were frozen and sectioned for fluorescent imaging experiments after the treatment with different PBQ concentrations over various times.A significant fluorescent increase in the snail's liver was observed upon exposure to 0.75 mg/L PBQ for 72 h,which highlighted an increase in *** and eosin(HE)staining further supported PBQ-induced liver damage with a viscosity increase in *** study provides a new rapid optical visualization method to study the killing mechanisms of PBQ and may help discover new chemicals that control snail populations.

Unveiling phosphofructokinase as the key target: an innovative strategy combining pull-down assays with optical imaging for snail-killing mechanism exploration

The Pomacea canaliculata, also known as apple snail, is an invasive species in China, posing a serious threat to agricultural production and public health. To prevent its rampant infestation, we developed a highly efficient snail-killing lead compound, 1-(4-chlorophenyl)-3-(pyridin-3-yl)urea (PBQ). However, the killing mechanism of PBQ remains unclear. To elucidate this, two novel compounds, B-PBQ and J-PBQ, were developed through structural modification of PBQ and were used, through both protein pull-down assay and confocal laser scanning microscopy imaging, to identify and confirm that phosphofructokinase(PFK) in the apple snail is the primary target protein of PBQ. Further studies revealed that PBQ successfully inhibits the activity,content, and expression of PFK in apple snail, disrupting the glycolysis pathway and leading to abnormal energy *** disruptions impair the snail’s tolerance to low oxygen, resulting in its efficient eradication. These findings offer a promising direction for developing next-generation PFK-targeted snail-killing pesticides. Furthermore, the strategy of combining pull-down assays with optical imaging used in this work provides a novel approach for the exploration of mechanisms of other pesticide candidates.

Dysbiosis of the gut microbiota may lead to a wide range of metabolic， neurological， intestinal and cardiovascular disorders and even to tumorigenesis. Evidence suggests that the gut-brain axis (GBA) plays a crucial role in the treatment of these diseases. Many plant-derived natural actives modulate the gut microbiota and its metabolites， gut hormones and neurotransmitters through a variety of mechanisms， and these actions contribute to the alleviation of irritable bowel syndrome， type 2 diabetes mellitus， cancer， and brain disorders. This review focuses on how natural products act through GBA to protect the central nervous system， regulate metabolism and promote apoptosis in cancer cells. The role of gut microbiota metabolites and neurotransmitter release in modulating inflammation-related factors， promoting or reducing oxidative stress， and activating or inhibiting related signaling pathways is also discussed. This comprehensive overview of the complex GBA mechanisms deepens the understanding of how natural products can target GBA to treat disease， providing valuable insights into the development and utilization of these natural interventions.

Today, tuberculosis (TB) remains a global public health threat associated with significantly high rates of morbidity and mortality. The World Health Organization's (WHO) Global Tuberculosis Report 2018[1] has reported that in 2017, 10.0 million people across the world had developed TB diseases that resulted in an estimated 1.6 million deaths, and 889, 000 people developed TB in China that led to 39, 000 TB-related deaths. Therefore, rapid and accurate detection of Mycobacterium tuberculosis (MTB) is important for initiating early treatment and reducing mortality. Traditional diagnostic methods for pulmonary TB incorporate chest radiography and sputum smear microscopy;however, several cases of tuberculosis go undiagnosed because of the low sensitivity of smear microscopy[2].

Introduction:This study explored the incidence trends and spatial clustering of typhoid and paratyphoid fever(TPF)in Yunnan Province to provide scientific evidence for developing and improving prevention and control ***:Temporal trends were investigated by calculating the annual percent change(APC)and average annual percent change(AAPC),along with their 95%confidence intervals(CIs).The spatial clustering of TPF across Yunnan Province was examined using global Moran’s I and local indicators of spatial association(LISA)***:A total of 206,066 TPF cases were reported in Yunnan Province from 1989 to 2022,with an average annual incidence of 13.98 per 100,000 population and a case fatality rate of 2.5 per 1,*** greatest number of cases was reported during July and *** 25–34-year age group had the highest incidence,and farmers were prominently *** incidence in Yunnan Province showed a significant decrease and spatial *** 2005 to 2022,13 county-level cities/counties/municipal districts in 5 prefectures(cities)in Yunnan Province were identified as statistically significant H-H spatial clusters of TPF incidence.A total of 24 TPF outbreaks were reported in Yunnan Province from 2005 to ***:The incidence of TPF in Yunnan Province showed a significant decrease and spatial *** strategies should focus on highincidence areas,seasons,and populations to reduce the incidence of TPF.

Obesity is positively Associated with Depression in Older Adults:Role of Systemic Inflammation

Objective We aimed to explore the association between obesity and depression and the role of systemic inflammation in older *** Adults≥65 years old(n=1,973)were interviewed at baseline in 2018 and 1,459 were followed up in *** and abdominal obesity were assessed,and serum C-reactive protein(CRP)levels were measured at *** status was assessed at baseline and at *** regression was used to analyze the relationship between obesity and the incidence of depression and worsening of depressive symptoms,as well as the relationship between obesity and CRP *** associations of CRP levels with the geriatric depression scale,as well as with its three dimensions,were investigated using multiple linear *** General obesity was associated with worsening depression symptoms and incident depression,with an odds ratio(OR)[95%confidence interval(CI)]of 1.53(1.13-2.12)and 1.80(1.23-2.63),especially among old male subjects,with OR(95%CI)of 2.12(1.25-3.58)and 2.24(1.22-4.11),respectively;however,no significant relationship was observed between abdominal obesity and *** addition,general obesity was associated with high levels of CRP,with OR(95%CI)of 2.58(1.75-3.81),especially in subjects free of depression at baseline,with OR(95%CI)of 3.15(1.97-5.04),and CRP levels were positively correlated with a score of specific dimension(life satisfaction)of depression,P<*** General obesity,rather than abdominal obesity,was associated with worsening depressive symptoms and incident depression,which can be partly explained by the systemic inflammatory response,and the impact of obesity on depression should be taken more seriously in the older male population.
