Cutaneous neurogenic inflammation （CNI） is inflammation that is induced （or enhanced） in the skin by the release of neuropeptides from sensory nerve endings. clinical manifestations are mainly sensory and vascular disorders such as pruritus and erythema. Transient receptor potential vanilloid 1 and ankyrin 1 （TRPV1 and TRPA1, respectively） are non-selective cation channels known to specifically participate in pain and CNI. Both TRPV1 and TRPA1 are co-expressed in a large subset of sensory nerves, where they integrate numerous noxious stimuli. It is now clear that the expression of both channels also extends far beyond the sensory nerves in the skin, occuring also in keratinocytes, mast cells, dendritic cells, and endothelial cells. In these non-neu- ronal cells, TRPV1 and TRPA1 also act as nociceptive sensors and potentiate the inflammatory process. This review discusses the role of TRPV1 and TRPA1 in the modulation of inflammatory genes that leads to or maintains CNI in sensory neurons and non-neuronal skin cells. In addition, this review provides a summary of current research on the intracellular sensitization pathways of both TRP channels by other endogenous inflammatory mediators that promote the self-maintenance of CNI.

Custard apple is a dry land *** leaves exhibit different pharmacological *** the present study,both silver(Ag)nanoparticles and chitosan-coated Ag(Chi-Ag)nanoparticles were fabricated using the aqueous leaf extract of the custard apple *** preliminary phytochemical analysis,various types of phytocompounds were found in the aqueous leaf extract of the same ***,both nanoparticles were physiochemically *** analysis exhibited the fingerprint vibrational peaks of active bioactive compounds in plant extract,Ag nanoparticles,and Chi-Ag ***/Visible spectral analysis revealed the highest absorbance peak at 419 nm,indicating the presence of Ag *** analysis presented the face-centered cubic(FCC)structure of both prepared ***,the average crystalline size of both Ag nanoparticles and Chi-Ag nanoparticles was calculated to be 23 and 74 nm,*** analysis showed the spherical and cubical shapes of Ag nanoparticles and Chi-Ag nanoparticles,*** analysis indicated a peak around 3.29 keV,conforming to the binding energies of Ag *** biogenic nanomaterial also showed strong antibacterial activity against all tested bacterial pathogens.

用脉搏波传导速度（pulse wave velocity,PWV）评估的动脉僵硬度是心血管病的一项替代指标,是心血管事件和全因死亡率的独立预测因素。弹性动脉的僵硬度随年龄增大而增加,主要是由左心室产生的搏动性机械应力造成动脉壁弹性蛋白层的疲劳和断裂所致。高收缩压增加心脏负荷和动脉应力,因此加速心血管退化和动脉粥样硬化。研究已证明儿童期血压是成年期PWV的独立预测因素,且血压,尤其是血压升高,可从儿童期追踪到成年期。

Abelmoschus is a genus of about fifteen species belongs to the family Malvaceae. The herb, popularly known as Lady’s Finger or Okra (in English) is a nutritional source used for both medicinal and culinary purposes. The plant is widely distributed from Africa to Asia, Southern Europe, and America. This comprehensive account provides a botanical description of the plant, its phytochemical constituents and pharmacological activities focusing anti-diabetic, anti-oxidant, anti-adhesive, gastro-protective, hepatoprotective, and immunomodulating actions. Most of the pharmacological effects can be explained by the constituents like tannins, terpenoids, flavonoids and glycosides present in all plant parts. However, future efforts should concentrate more on in vitro and in vivo studies and also on clinical trials in order to confirm traditional wisdom in the light of a rational phytotherapy. The present review is an overview of phytochemistry and ethnopharmacological studies that support many of the traditional?ethnomedicinal uses of the plant.

Assays that measure steroid hormones in patient care, public health, and research need to be both accurate and precise, as these criteria help to ensure comparability across all clinical and research applications. This review addresses major issues relevant to assay variability and describes recent activities by the US Centers for Disease Control and Prevention （CDC） to improve assay performance. Currently, high degrees of accuracy and precision are not always met for testosterone and estradiol measurements; although technologies for steroid hormone measurement have advanced significantly, measurement variability within and across laboratories has not improved accordingly. Differences in calibration and specificity are discussed as sources of variability in measurement accuracy. Ultimately, a combination of factors appears to cause inaccuracy of steroid hormone measurements, with nonuniform assay calibration and lack of specificity being two major contributors to assay variability. Within-assay variability for current assays is generally high, especially at low analyte concentrations. The CDC Hormone Standardization （HoSt） Program is improving clinical assays, as evidenced by a 50% decline in mean absolute bias between mass spectrometry assays and the CDC reference method from 2007 to 2011. This program provides the measurement traceability to CDC reference methods and helps to minimize factors affecting measurement variability.

BACKGROUND Overexpression of heat shock proteins(HSPs) is associated with several malignancies and contributes to the development, progression, and metastasis of cancer, in addition to the inhibition of cellular death. In recent years, there has been active research into using HSP inhibitors in several malignancies. Due to the poor prognosis of esophageal adenocarcinoma(EAC), it would be valuable to find new biomarkers for the development of cancer *** To evaluate the expressions of HSP27 and HSP70 and their effect on survival in *** Immunohistochemical analyses and evaluations of HSP27 and HSP70 expression were performed on all available samples from 93 patients diagnosed with EACbetween 1990 and 2007 at two university hospitals. Fifteen cases with Barrett's metaplasia and 5 control cases from the same patient population were included in the analysis. HSP expression was quantitatively assessed and classified as high or low. Kaplan-Meier analyses and Cox regression models adjusting for age and sex as well as tumor site, stage, and grade were used to evaluate the effect on *** Tumor stage and surgical treatment were the main prognostic factors. High HSP27 expression in cancer cases was a strong negative predictive factor, with a mean survival of 23 mo compared to the 49 mo in cases with a low expression(P= 0.018). The results were similar for HSP70, with a poorer survival of 17 mo in cases with high HSP70 expression, in contrast to 40 mo(P = 0.006) in cases with a low expression. A Cox regression survival analysis was performed, adjusting for possible confounding factors, and higher HSP27 and HSP70 expressions remained an independent negative prognostic factor. The HSPs' correlation with survival was not affected by cancer treatments. The discovery and development of novel therapeutic agents has become a more challenging endeavor due to the increasing costs associated with newer technologies, greater regulatory requirements for drug registration, the relatively high rate of failure in drug development which has not changed appreciably in the last decade and the increased competition for market share. As a result of the latter two issues, increased focus has been placed on discovering clinical candidates with increased probabilities of success in clinical practice, and with the ability to differentiate and demonstrate therapeutic advantage in clinical outcomes compared to existing therapy. Drug disposition sciences play a critical role in this effort.

Melioidosis, an infection caused by Gram-negative Burkholderia pseudomallei (Bp), has high clinical recurrence and mortality rates associated with pneumonia and sepsis. With the limitations in current therapeutic options and the lack of available human vaccines, development of novel countermeasures against Bp infection is vital. In this study, we evaluated the efficacy of an aminoalkyl glucosaminide 4-phosphate (AGP), a synthetic toll like receptor 4 agonist (CRX-527), in conferring protection against melioidosis in a murine model. Survival data showed 66% of mice treated with AGP prior to lethal intranasal Bp challenge survived and presented no signs of illness over a 3 months period. In contrast, all control mice succumbed to infection within 4 days. Kinetic study on organ bacterial burden demonstrated mice treated with AGP had dramatically reduced bacterial loads in both the lungs and spleens as compared to control mice. Notably, all but one AGP-treated mouse had no Bp growth in the blood as compared to overwhelming bacteraemia found in all control mice. The protective effect of CRX-527 was associated with a transient increase in pulmonary cytokine/ chemokine levels, which boosted the host’s innate immunity. This enabled rapid clearance of the pulmonary and systemic bacterial burden and prevented the development of sepsis. This study demonstrated the potential use of TLR4 agonist as a prophylactic immunotherapy in preventing melioidosis.
