Molecular cloning,characterization and promoter analysis of LbgCWIN1 and its expression profiles in response to exogenous sucrose during in vitro bulblet initiation in lily

Lily(Lilium spp.) is an important ornamental flower, which is mainly propagated by bulbs. Cell wall invertases(CWINs), which catalyze the irreversibly conversion of sucrose into glucose and fructose in the extracellular space, are key enzymes participating in sucrose allocation in higher plants. Previous studies have shown that CWINs play an essential role in bulblet initiation process in bulbous crops, but the underlying molecular mechanism remains unclear. Here, a CWIN gene of Lilium brownii var. giganteum(Lbg) was identified and amplified from genomic DNA. Quantitative RT-PCR assays revealed that the expression level of LbgCWIN1 was highly upregulated exactly when the endogenous starch degraded in non-sucrose medium during in vitro bulblet initiation in Lbg. Phylogenetic relationship, motif, and domain analysis of LbgCWIN1 protein and CWINs in other plant species showed that all sequences of these CWIN proteins were highly conserved. The promoter sequence of LbgCWIN1 possessed a number of alpha-amylase-, phytohormone-, light-and stress-responsive cis-elements. Meanwhile, β-glucuronidase(GUS) assay showed that the 459 bp upstream fragment from the translational start site displayed maximal promoter activity. These results revealed that LbgCWIN1 might function in the process of in vitro bulblet initiation and be in the response to degradation of endogenous starch.

A mutation in the promoter of the yellow stripe-like transporter gene in cucumber results in a yellow cotyledon phenotype

Leaf color mutants in higher plants are considered to be ideal materials for studying the chlorophyll biosynthesis,photosynthesis mechanism and chloroplast ***,we identified a spontaneous mutant,yc412,in cultivated cucumber that exhibited yellow *** yellow-lethal mutant was diagnosed with an abnormal chloroplast ultrastructure,and reduced photosynthetic capacity and pigment *** bulked segregant analysis-based whole-genome sequencing and fine genetic mapping,we narrowed the yellow cotyledons (yc) locus to a 96.8 kb interval on chromosome *** resequencing and molecular cloning,we showed that Csyc is a potential candidate gene,which encodes a yellow stripe-like (YSL) *** T to C mutation in the promoter region of Csyc caused the yellow cotyledon phenotype in *** to YZU027A (WT),the expression of Csyc was significantly downregulated in the cotyledons of *** of Csyc in cucumber via virus-induced gene silencing resulted in chlorotic leaves,mainly by suppressing the chlorophyll ***,a comparative transcriptome analysis revealed that chloroplast-related genes and chlorophyll biosynthesis genes were significantly downregulated in yc412 *** results provide new insights into the molecular function of the YSL transporter in plant chloroplast development and chlorophyll synthesis.

Analysis of spatial distribution characteristics and driving factors of ethnicminority villages in China using geospatial technology and statistical models

This study aims to reveal the spatial structural characteristics of 1,652 Ethnic-Minority Villages(EMV)in China and to analyze the mechanisms driving their spatial *** are a special type of settlement space that preserve a large number of historical traces of the ethnic culture of ancient *** are important carriers of China’s excellent traditional culture and are key to the implementation of rural revitalization *** this study,1652 EMV in China were selected as the research *** Nearest Neighbor Index,kernel density,and spatial autocorrelation index were employed to reveal the spatial structural characteristics of minority *** network models,spatial lag models,and geographical detectors were used to analyze the formation mechanism of spatial heterogeneity in *** results indicate that:(1)EMV exhibit significant spatial differentiation characterized by“single-core with multiple surrounding sub-centers,”“polarization between east and west,”“decreasing quantity from southwest to east coast to northeast to northwest,”and“large dispersion with small agglomeration.”(2)EMV are mainly distributed in areas rich in intangible cultural heritage,with high vegetation coverage and low altitude,far from central cities,and having limited arable land and an underdeveloped economy and transportation,particularly in shaded or riverbank areas.(3)Distance from the nearest river(X3),distance from central cities(X8),national intangible cultural heritage(X9),and NDVI(X10)were the main driving factors affecting the spatial distribution of EMV,whereas elevation(X1)and GDP(X5)had the weakest *** EMV are a relatively unique territorial spatial unit,the identification of their spatial heterogeneity characteristics not only deepens the research content of settlement geography,but also involves the assessment,protection,and development of Minority Villages,which is of great significance for the inheritance and utilization of excellent

The Spatio-temporal Evolution of Ecological Risk and Its Drivers in the Weihe River Basin landscape from 1990 to 2020

The Weihe River Basin has a significant number of tributaries and a delicate ecological *** the spatial and temporal evolution and determinants of landscape ecological risk in the Weihe River Basin(WRB)can improve the scientific protection and development of its watershed *** study is based on land use statistics from the WRB for a 30-year period represented by 1990,2000,2010,and *** initial model for the assessment of landscaping ecological hazards was created using the software that was also used to generate the landscape ecological risk index,such as ArcGIS 10.4 and Fragstats ***,the spatial and temporal evolution of landscape ecological risk in the vicinity of the study area was characterized by the trajectory of the center of gravity migration and the spatial autocorrelation of ***,Geodetector was used to analyze ecological risk drivers in the *** to the findings,the high-risk and relatively high-risk regions are steadily expanding,while the low-risk and relatively low-risk areas dominate the ecological risk landscape in the *** the Weihe River Basin,Xianyang and Xi'an are the areas to which the high-risk centers of gravity are *** spatial correlations were found between the landscape ecological hazards in the study area,most prominently in the form of high-high and low-low *** primary drivers are the interplay between the GDP component,temperature,and elevation as a single factor.

FLOWERING LOCUS T(FT)in Photosensitive Type Chrysanthemum Accelerates Flowering in Arabidopsis

FLOWERING LOCUS T(FT)is an important factor for integrating flowering signals through the photoperiod pathway,which significantly promotes *** chrysanthemum varieties are short-day plants,sensitive to the photoperiod,which limits their ornamental and annual production *** this study,we aim to analyze the nutrients and flowering-related genes of chrysanthemums with different photoperiod types and to clone and verify the function of the flowering-related gene *** found that the formation of floral buds requires the accumulation of starch while consuming soluble sugars and the expression patterns of flowering-related genes GIGANTEA(GI),CONSTANS(CO),and FT in ***‘Zilian’and ***‘Zihongtuogui’had a certain synchronization during floral buds differentiation according to our quantitative validation,and the expression levels of CmGI,CmCO and CmFT in ***‘Zihongtuogui’were higher than those in ***.‘Zilian’in the later stage of *** was cloned from photosensitive ***‘Zihongtuogui’and polypeptide alignment and phylogenetic analysis showed that CmFT was clustered in FT-like *** further functional verification,we obtained two Arabidopsis transgenic *** results showed that CmFT transgenic ft mutant lines can significantly accelerate flowering in ***,we can initially confirm that CmFT plays an important role in promoting flowering,which may be the key reason for the photosensitivity of ***‘Zihongtuogui’.Overall,the results of this study are of great importance in revealing the flowering mechanism of different photoperiod types of chrysanthemums.

Migration and fate of polycyclic aromatic hydrocarbons in bioretention systems with different media:experiments and simulations

Polycyclic aromatic hydrocarbons(PAHs)present significant risks to human health owing to their carcinogenic,teratogenic,and mutagenic *** contamination of surface water with PAHs via runoff has become a prominent source of water *** the capacity of bioretention systems to remove PAHs from runoff is recognized,the dynamics of PAH migration and degradation in these systems are not *** study aims to explain the migration and fate of PAHs in bioretention systems through a series of experiments and model *** study constructed bioretention systems with three different media types and found that these systems achieved PAH load reductions exceeding 92%.Notably,naphthalene(NAP),fluoranthene(FLT),and pyrene(PYR)tended to accumulate in the media’s upper layer,at depths of 10 to 40 *** further analyze the migration and fate of PAHs during multi-site rainfall events and across prolonged operation,we applied the HYDRUS-1D model under three distinct *** findings of this study indicated that NAP degraded in 40 d,whereas FLT and PYR showed incomplete degradation after 120 *** continuous rainfall events,there was no clear pattern of PAH accumulation;however,FLT and PYR persisted in the bioretention *** combination of experimental and simulation findings highlights the inevitable accumulation of PAHs during extended use of bioretention *** research provides a theoretical basis for improving operational efficiency,advancing PAH degradation in bioretention systems,and reducing their toxicity.

Quantitative trait loci identification reveals zinc finger protein CONSTANS-LIKE 4 as the key candidate gene of stigma color in watermelon(Citrullus lanatus)

Stigma color is a critical agronomic trait in watermelon that plays an important role in ***,there are few reports on the regulation of stigma color in *** this study,a genetic analysis of the F2 population derived from ZXG1553(P1,with orange stigma)and W1-17(P2,with yellow stigma)indicated that stigma color is a quantitative trait and the orange stigma is recessive compared with the yellow *** segregant analysis sequencing(BSA-seq)revealed a 3.75 Mb segment on chromosome 6 that is related to stigma ***,a major stable effective QTL Clqsc6.1(QTL stigma color)was detected in two years between cleaved amplified polymorphic sequencing(CAPS)markers Chr06_8338913 and Chr06_9344593 spanning a~1.01 Mb interval that harbors 51 annotated ***97C06G117020(annotated as zinc finger protein CONSTANS-LIKE 4)was identified as the best candidate gene for the stigma color trait through RNA-seq,quantitative real-time PCR(qRT-PCR),and gene structure alignment analysis among the natural watermelon *** expression level of Cla97C06G117020 in the orange stigma accession was lower than in the yellow stigma accessions with a significant difference.A nonsynonymous SNP site of the Cla97C06G117020 coding region that causes amino acid variation was related to the stigma color variation among nine watermelon accessions according to their re-sequencing *** color formation is often related to carotenoids,and we also found that the expression trend of ClCHYB(annotated asβ-carotene hydroxylase)in the carotenoid metabolic pathway was consistent with Cla97C06G117020,and it was expressed in low amounts in the orange stigma *** data indicated that Cla97C06G117020 and ClCHYB may interact to form the stigma *** study provides a theoretical basis for gene fine mapping and mechanisms for the regulation of stigma color in watermelon.

Spatiotemporal Evolution of Urban Heat Islands and Optimization of Spatial Network Construction in the Central Urban Area of Fuzhou City, China

Although accelerated urbanization has led to economic prosperity,it has also resulted in urban heat island ***,identifying methods of using limited urban spaces to alleviate heat islands has become an urgent *** this study,we assessed the spatiotemporal evolution of urban heat islands within the central urban area of Fuzhou City,China from 2010 to *** assessment was based on a morphological spatial pattern analysis(MSPA)model and an urban thermal environment spatial network constructed us-ing the minimum cumulative resistance(MCR)*** measures for the spatial network were proposed to provide a theor-etical basis for alleviating urban heat *** results show that the heat island area within the study area gradually increased while that of urban cold island area gradually *** core area was the largest of the urban heat island patch landscape elements with a significant impact on other landscape elements,and represented an important factor underlying urban heat island network *** thermal environment network revealed a total of 197 thermal environment corridors and 93 heat island *** locations were then optimized according to the current land use,which maximized the potential of 1599.83 *** based on current land use led to an increase in climate resilience,with effective measures showing reduction in thermal environment spatial network structure and function,contributing to the mitigation of urban heat *** findings support the use of current land use patterns during urban heat island mitigation measure planning,thus providing an important reference basis for alleviating urban heat island effects.

Spatial-temporal Distribution and Influencing Factors of Urban Carbon Emissions under the Background of Carbon Emission Reduction:A Case Study of Guangxi Autonomous Region

Based on the panel data of Guangxi from 2005 to 2017,the spatiotemporal characteristics and determinants of urban carbon emissions in Guangxi were analyzed using the extended STIRPAT model and the Geographically and Temporally Weighted Regression(GTWR)*** main findings of our research can be summarized as *** the total carbon emissions of cities in Guangxi consistently increased from 2005 to 2014,the growth trend slowed after 2014,leading to a stabilization in the total *** addition,there are significant differences in the total carbon emissions among the *** central and northeastern regions have higher emissions,while the southwestern region has lower ***,there are variations in the degrees and directions of the impacts that factors have on carbon emissions among the different time periods and *** land use is a key factor driving carbon emissions,and it has a negative impact on most cities in ***,factors such as industrial structure,population urbanization,population concentration,and economic growth have significant positive effects on carbon emissions in *** influence of urban roads on carbon emissions is generally positive,while the degree of openness to the outside world and environmental regulations has relatively weaker impacts on *** summary,in order to promote the low-carbon transition of Guangxi and achieve high-quality development,the cities in Guangxi should implement differentiated urban carbon reduction strategies that are focused on optimizing urban land use and industrial structure.
