Dysregulation of CDK8 and Cyclin C in tumorigenesis

Appropriately controlled gene expression is fundamental for normal growth and survival of all living organisms. In eukaryotes, the tran- scription of protein-coding mRNAs is dependent on RNA polymerase II （Pol II）. The multi-subunit transcription cofactor Mediator complex is proposed to regulate most, if not all, of the Pol II-dependent transcription. Here we focus our discussion on two subunits of the Mediator complex, cyclin-dependent kinase 8 （CDK8） and its regulatory partner Cyclin C （CycC）, because they are either mutated or amplified in a variety of human cancers. CDK8 functions as an oncoprotein in melanoma and colorectal cancers, thus there are considerable interests in developing drugs specifically targeting the CDK8 kinase activity. However, to evaluate the feasibility of targeting CDK8 for cancer therapy and to understand how their dysregulation contributes to tumorigenesis, it is essential to elucidate the in vivo function and regulation of CDKS-CycC, which are still poorly understood in multi-cellular organisms. We summarize the evidence linking their dysregulation to various cancers and present our bioinformatics and computational analyses on the structure and evolution of CDK8. We also discuss the implications of these observations in tumorigenesis. Because most of the Mediator subunits, including CDK8 and CycC, are highly conserved during eukaryotic evolution, we expect that investigations using model organisms such as Drosophila will provide important insights into the function and regulation of CDK8 and CycC in different cellular and developmental contexts.

The interest of consumers in farming activities is evident and agricultural producers have responded to this increasing interest in a number of ways, including inviting visitors to farm operations through tourism or farm visits. Livestock industries, in particular, are interacting with the public in numerous ways, including via marketing channels for their products, interacting with community leaders and community members in regard to location siting and regulations, and inviting people onto their operations via agritourism. An improved understanding of who visits agricultural locations (particularly livestock operations) and consumers’ perceptions about livestock agriculture is needed in order to begin to understand the possible relationships between on-farm experiences and consumers’ perceptions. Thus, it is important to gain a deeper understanding in consumers’ interests, including their levels of concern for production process attributes, such as animal welfare, relative to product attributes like price and taste. This analysis investigates the demographics of consumers who have (and have not) visited various agritourism operations, studies differences in preferences for (or perceptions of) animal welfare relative to other pork attributes between consumers who have or have not visited a livestock operation, as well as amongst consumers with varying levels of involvement in household food production. Indeed the relationships explored are between having visited a livestock operation and the perceptions. This study found that 69% of participants had visited a livestock operation (pig farm, dairy farm, and/or horse farm) at some point. The majority of respondents agreed that agriculture was an important industry, yet, those who had visited livestock operations were more likely to note concerns about the impact of livestock operations on water quality in their county.

The motivation for cost-effective management of highway pavements is evidenced not only by the massive expenditures associated with these activities at a national level but also by the consequences of poor pavement condition on road *** paper presents a state-of-the-art review of multi-objective optimization(MOO)problems that have been formulated and solution techniques that have been used in selecting and scheduling highway pavement rehabilitation and maintenance ***,the paper presents a taxonomy and hierarchy for these activities,the role of funding sources,and levels of *** paper then describes how three different decision mechanisms have been used in past research and practice for project selection and scheduling(historical practices,expert opinion,and explicit mathematical optimization)and identifies the pros and cons of each *** paper then focuses on the optimization mechanism and presents the types of optimization problems,formulations,and objectives that have been used in the ***,the paper examines various solution algorithms and discusses issues related to their ***,the paper identifies some barriers to implementing multi-objective optimization in selecting and scheduling highway pavement rehabilitation and maintenance activities,and makes recommendations to overcome some of these barriers.

People’s attitudes towards animals are likely to impact their quality of care offered. Understanding perceptions of cats is important to identify potential cat welfare issues and to facilitate best care practices. An online survey was developed to gather information from 1172 U.S. residents on perceptions of cats and sources of cat welfare information. Demographic variables, information regarding respondents’ characterization, knowledge and beliefs of cats, their behavior, and welfare needs were collected. A subsample of cat owning households was surveyed regarding their opinions on cat behavior. Veterinarians were also surveyed to determine if they differed from general respondents in their views of and knowledge of cats and their care. The highest percentage of respondents in both groups identified veterinarians as the most accessible (52% veterinarian, 29% general public respondents) and most credible sources (veterinarians 76%, general public 29%) of information on cat behavior. In response to being asked about acceptable reasons to relinquish cats, more general respondents selected “convenience” (13% veterinarians, 3% general population). Several inconsistencies in perceptions and practices between and within members of the general public and veterinarians were observed. General respondents and veterinarians both indicated a lack of confidence in addressing behavioral issues in cats. Both groups disagreed that they (in the case of veterinarians) or their veterinarians provided useful resources on cat behavior or health. Opportunities exist for improvement in consistency between knowledge, attitudes and practices or services relative to cats. Improved competence and information transfer on both the behavioral and physical health aspects of cat welfare are also needed.

Translational control of gene expression,including recruitment of ribosomes to messenger RNA(mRNA),is particularly important during the response to *** of ribosomeassociated mRNAs using translating ribosome affinity purification(TRAP)followed by RNAsequencing facilitates the study of mRNAs undergoing active transcription and better proxies the translatome,or protein response,to *** identify plant responses to Magnesium(Mg)deficiency at the translational level,we combined transcriptome and translatome ***,we found 26 previously unreported Mg-responsive genes that were only regulated at the translational level and not the transcriptional level,during the early response to Mg *** addition,mutants of the transcription factor ELONGATED HYPOCOTYL 5(HY5),the H^(+)/CATION EXCHANGER 1 and 3(CAX1 and CAX3),and UBIQUITIN 11(UBQ11)exhibited early chlorosis phenotype under Mg deficiency,supporting their functional involvement in ion ***,our study strongly supports that TRAP-seq combined with RNA-seq followed by phenotype screening could facilitate the identification of novel players during stress responses.

For resolving harvesting technology of fresh rice straw silage and plugging of the round steel-roll baler in China,experimental researches were carried *** harvesting technology,baling silage and chopping silage were *** the round baler,three kinds of feeding rolls equipped for enhancing feeding capability were experimented separately by *** results indicate:harvesting technology of baling fresh rice straw as silage is practicable;slicing-disc feeding rolls can be used to the baler to resolve plugging in straw-baling *** through further experiments by reliability and density,optimal structure of the feeding roll is obtained:big and small slicing-discs arranged in interval and inclination,distance between slicing-discs being 30-40 mm.

A three-stage method with efficient calculation for lot streaming flow-shop scheduling

An important production planning problem is how to best schedule jobs(or lots)when each job consists of a large number of identical *** problem is often approached by breaking each job/lot into sublots(termed lot streaming).When the total number of transfer sublots in lot streaming is large,the computational effort to calculate job completion time can be ***,researchers have largely neglected this computation time *** provide a practical method for production scheduling for this situation,we propose a method to address the n-job,m-machine,and lot streaming flow-shop scheduling *** consider the variable sublot sizes,setup time,and the possibility that transfer sublot sizes may be bounded because of capacity constrained transportation *** proposed method has three stages:initial lot splitting,job sequencing optimization with efficient calculation of the makespan/total flow time criterion,and transfer *** experiments are conducted to confirm the effectiveness of the three-stage *** experiments reveal that relative to results reported on lot streaming problems for five standard datasets,the proposed method saves substantial computation time and provides better solutions,especially for large-size problems.

This paper studied new class of integral equation called the Korteweg-de Vries-Kadomtsev-Petviashvili(KdV-KP)*** equation consist of the well-known fifth-order KdV equation in the context of the Kadomtsev-Petviashvili *** newly gathered class of sixth-order KdV-KP equation is studied using the sub-equation method to obtain several soliton-type solutions which consist of trigonometric,hyperbolic,and rational *** application of the sub-equation approach in this work draws attention to the outstanding characteristics of the suggested method and its ability to handle completely integrable ***,the obtained solutions have not been reported in the previous literature and might have significant impact on future research.

A central role of the endoplasmic reticulum(ER)is the synthesis,folding and quality control of secretory *** proteins usually exit the ER to enter the Golgi apparatus in coat protein complex II(COPII)-coated vesicles before transport to different subcellular ***,in plants there are specialized ER-derived vesicles(ERDVs)that carry specific proteins but,unlike COPII vesicles,can exist as independent organelles or travel to the vacuole in a Golgi-independent *** specialized ERDVs include protein bodies and precursor-accumulating vesicles that accumulate storage proteins in the endosperm during seed *** ERDVs also include precursor protease vesicles that accumulate amino acid sequence KDEL-tailed cysteine proteases and ER bodies in Brassicales plants that accumulate myrosinases that hydrolyzes *** functionally specialized ERDVs act not only as storage organelles but also as platforms for signal-triggered processing,activation and deployment of specific proteins with important roles in plant growth,development and adaptive *** specialized ERDVs have also been exploited to increase production of recombinant proteins and *** we discuss our current understanding of the functional diversity,evolutionary mechanisms and biotechnological application of specialized ERDVs,which are associated with some of the highly remarkable characteristics important to plants.
