Geographical origin identification of winter jujube(Ziziphus jujuba Dongzao')by using multi-element fingerprinting with chemometrics

Winter jujube(Ziziphus jujuba'Dongzao')is greatly appreciated by consumers for its excellent quality,but brand infringement frequently occurs in the ***,we first determined a total of 38 elements in 167 winter jujube samples from the main winter jujube producing areas of China by inductively coupled plasma mass spectrometer(ICP-MS).As a result,16 elements(Mg,K,Mn,Cu,Zn,Mo,Ba,Be,As,Se,Cd,Sb,Ce,Er,Tl,and Pb)exhibited significant differences in samples from different producing *** linear discriminant analysis(LDA)and orthogonal projection to latent structures discriminant analysis(OPLS-DA)showed better performance in identifying the origin of samples than unsupervised principal component analysis(PCA).LDA and OPLS-DA had a mean identification accuracy of 87.84 and 94.64%in the testing set,*** using the multilayer perceptron(MLP)and C5.0,the prediction accuracy of the models could reach 96.36 and 91.06%,*** on the above four chemometric methods,Cd,Tl,Mo and Se were selected as the main variables and principal markers for the origin identification of winter ***,this study demonstrates that it is practical and precise to identify the origin of winter jujube through multi-element fingerprint analysis with chemometrics,and may also provide reference for establishing the origin traceability system of other fruits.

Non-alcoholic fatty liver disease (NAFLD) has become a global issue and a severe threat to public ***, to date, no approved therapeutic drugs have been developed. Dietary interventions with naturalproducts have shown promise in preventing and treating NAFLD. Sulforaphane (SFN) is a phytocompoundwith antioxidant and anti-inflammatory properties, and previous research has demonstrated that SFN canameliorate hepatic lipid accumulation and inflammation. However, the molecular mechanisms underlying thesebeneficial effects remain unclear. In this study, we confirmed the protective effects of SFN on excessive lipidaccumulation and inflammatory injury in a high-fat, high-fructose diet-induced non-alcoholic steatohepatitis(NASH) mouse model. We found that SFN attenuates the inflammatory injury in a macrophage cell line andthe liver of NASH mice, owing to the promotion of M1-type macrophage polarization toward the M2-type andthe regulation of inflammatory mediators. Further analysis demonstrated that this SFN-induced macrophageM2-type polarization occurs in a Krüppel-like factor 4 (KLF4)-dependent manner. In summary, we uncovereda new mechanism of action underlying SFN activity and provide evidence that dietary intervention with SFNmight be protective against NASH.

An integrated transcriptome and proteomic investigation of the stress responses of VBNC state Salmonella Enteritidis triggered by sodium hypochlorite

Viable but non-culturable （VBNC） bacteria after chlorination disinfection pose a considerable threat to the public health and environment. The current work aimed to uncover the stress adaptation mechanisms of VBNC S. Enteritidis to sodium hypochlorite. In this study， we investigated the morphology， gene expressions and proteomic profile in VBNC S. Enteritidis. It demonstrated that 109cells mL-1of bacteria could transition into the VBNC state after exposure to 50 mg L-1chlorine for 2.5 h. The VBNC state cells exhibited cellular membrane impairment， decreased size， and intracellular ATP depletion compared to the culturable cells. In the VBNC state cells， there was a significant increase in intracellular reactive oxygen species （ROS） levels and NAD+/NADH ratios. This observation suggested an imbalance in the intracellular redox status of the VBNC bacteria. Transcriptome and proteomic analysis revealed significant upregulation of the resistance-nodulation-division （RND） efflux systems （arcA， tolC， ramA）， oxidative stress response genes （soxR， trxA， grxB）， DNA repair genes （lexA， radA， mutH， mutL）， and virulence factor genes （sseA， sseL， eutS， and eutN）. Our findings indicated that it is important to formulate disinfection protocols that are specifically designed for food processing plants and healthcare facilities， focusing on implementing a more thorough disinfection method to effectively eradicate VBNC cells.

Comparative evaluation of commercial Douchi by different molds:biogenic amines,non-volatile and volatile compounds

To provide new insights into the development and utilization of Douchi artificial starters,three common strains(Aspergillus oryzae,Mucor racemosus,and Rhizopus oligosporus)were used to study their influence on the fermentation of *** results showed that the biogenic amine contents of the three types of Douchi were all within the safe range and far lower than those of traditional fermented ***-type Douchi produced more free amino acids than the other two types of Douchi,and its umami taste was more prominent in sensory evaluation(P<0.01),while Mucor-type and Rhizopus-type Douchi produced more esters and pyrazines,making the aroma,sauce,and Douchi flavor more *** to the Pearson and PLS analyses results,sweetness was significantly negatively correlated with phenylalanine,cysteine,and acetic acid(P<0.05),bitterness was significantly negatively correlated with malic acid(P<0.05),the sour taste was significantly positively correlated with citric acid and most free amino acids(P<0.05),while astringency was significantly negatively correlated with glucose(P<0.001).Thirteen volatile compounds such as furfuryl alcohol,phenethyl alcohol,and benzaldehyde caused the flavor difference of three types of *** study provides theoretical basis for the selection of starting strains for commercial Douchi production.

Milk fat globule membrane(MFGM),which contains abundant glycoproteins and phospholipids,exerts beneficial effects on intestinal health and *** aim of this study was to evaluate the protective effects and possible underlying mechanisms of MFGM on cow’s milk allergy(CMA)in aβ-lactoglobulin(BLG)-induced allergic mice *** was supplemented to allergic mice induced by BLG at a dose of 400 mg/kg body *** demonstrated that MFGM alleviated food allergy symptoms,decreased serum levels of lipopolysaccharide,pro-inflammatory cytokines,immunoglobulin(Ig)E,Ig G1,and Th2 cytokines including interleukin(IL)-4,while increased serum levels of Th1 cytokines including interferon-γand regulatory T cells(Tregs)cytokines including IL-10 and transforming growth factor-β.MFGM modulated gut microbiota and enhanced intestinal barrier of BLG-allergic mice,as evidenced by decreased relative abundance of Desulfobacterota,Rikenellaceae,Lachnospiraceae,and Desulfovibrionaceae,while increased relative abundance of Bacteroidetes,Lactobacillaceae and Muribaculaceae,and enhanced expressions of tight junction proteins including Occludin,Claudin-1 and zonula ***,MFGM increased fecal short-chain fatty acids(SCFAs)levels,which elevated G protein-coupled receptor(GPR)43 and GPR109A *** increased expressions of GPR43 and GPR109A induced CD103+dendritic cells accumulation and promoted Tregs differentiation in mesenteric lymph node to a certain *** summary,MFGM alleviated CMA in a BLG-induced allergic mice model through enhancing intestinal barrier and promoting Tregs differentiation,which may be correlated with SCFAs-mediated activation of *** findings suggest that MFGM may be useful as a promising functional ingredient against CMA.

Lactobacillus delbrueckii subsp. bulgaricus(L. bulgaricus) and Streptococcus thermophilus(S. thermophilus) are commonly used starters in milk fermentation. Fermentation experiments revealed that L. bulgaricus-S. thermophilus interactions(LbSt I) substantially impact dairy product quality and production. Traditional biological humidity experiments are time-consuming and labor-intensive in screening interaction combinations, an artificial intelligence-based method for screening interactive starter combinations is necessary. However,in the current research on artificial intelligence based interaction prediction in the field of bioinformatics, most successful models adopt supervised learning methods, and there is a lack of research on interaction prediction with only a small number of labeled ***, this study aimed to develop a semi-supervised learning framework for predicting LbSt I using genomic data from 362 isolates(181per species). The framework consisted of a two-part model: a co-clustering prediction model(based on the Kyoto Encyclopedia of Genes and Genomes(KEGG) dataset) and a Laplacian regularized least squares prediction model(based on K-mer analysis and gene composition of all isolates datasets). To enhance accuracy, we integrated the separate outcomes produced by each component of the two-part model to generate the ultimate LbSt I prediction results, which were verified through milk fermentation experiments. Validation through milk fermentation experiments confirmed a high precision rate of 85%(17/20; validated with 20 randomly selected combinations of expected interacting isolates). Our data suggest that the biosynthetic pathways of cysteine, riboflavin, teichoic acid, and exopolysaccharides, as well as the ATP-binding cassette transport systems, contribute to the mutualistic relationship between these starter bacteria during milk fermentation. Role of bpfA in adhesion and biofilm formation of Shewanella putrefaciens CN32 under cold stress: a comprehensive transcriptomic analysis

As a specific spoilage organism of seafood under refrigerated temperature conditions， Shewanella spp. tend to form biofilms that exacerbate the occurrence of seafood spoilage. Biofilm-promoting factor A (BpfA) has been reported to promote the adhesion and biofilm formation of Shewanella spp.， but its role in adhesion and biofilm formation of S. putrefaciens under cold stress needs to be further investigated. To better comprehend the effect of BpfA on adhesion and biofilm formation of S. putrefaciens under cold stress (4℃)， bacterial adhesion and biofilm phenotype of S. putrefaciens CN32 WT and ΔbpfA at 4℃ were analyzed and performed transcriptomics. The results showed that the deletion of bpfA had almost no effect on the growth of S. putrefaciens CN32 at 4℃， but weakened the unicellular adhesion capacity of S. putrefaciens CN32 and destabilized the stability of the multicellular adhesion layer. In addition， the biomass of the mature biofilm formed by ΔbpfA was merely around 50% of that observed in the mature biofilm of S. putrefaciens CN32 WT， the average thickness and volume of the biofilm decreased by 3% and 8%， respectively， and the composition of the biofilm changed. Transcriptome analysis demonstrated that the deletion of bpfA led to differential expression of genes involved in metabolic pathways such as bacterial chemotaxis， two-component system， tyrosine metabolism， drug metabolism-other enzymes and biofilm formation-Vibrio cholerae， which in turn influenced bacterial adhesion and biofilm formation. Those results advance our acknowledge of the character of BpfA on adhesion and biofilm formation of S. putrefaciens CN32， which contributes to understand bacterial adhesion and the control of biofilm formation.

Recovery and in-situ reduction of precious metals by graphdiyne and graphdiyne oxide for antibacterial use

Based on the in-situ self-reduction and chemical stability,graphdiyne(GDY)and graphdiyne oxide(GDYO)are used as trapping agents to investigate the ability for recovering Au^(3+),Ag^+,and Pd^(2+)under different pH values and interfering *** strong acidity at pH=1,these two agents demonstrate high select recovery towards the three precious metal ions,which could be insitu reduced to nanoparticles(NPs).In addition,superparamagnetic Fe_(3)O_(4)NPs are deposited on the surfaces of GDY and *** magnetic responses enable GDYFe_(3)O_(4)to recover precious metals conveniently and efficiently by the aid of an external magnetic *** study also confirms the antibacterial activity of the as-recovered NPs deposited on GDY and GDYO against Escherichia coli and Staphylococcus aureus(1×10^(5)colony-forming unit(CFU)·mL^(-1)),and the antibacterial rates are 100%.This strategy of recovering precious metals and subsequently reusing to combat pathogens will be of great significance for environmental remediation and biomedical applications.
