Nanodrugs alleviate acute kidney injury: Manipulate RONS at kidney

Currently,there are no clinical drugs available to treat acute kidney injury(AKI).Given the high prevalence and high mortality rate of AKI,the development of drugs to effectively treat AKI is a huge unmet medical need and a research *** existing evidence fully demonstrates that reactive oxygen and nitrogen species(RONS)burst at the AKI site is a major contributor to AKI progression,the heterogeneity,complexity,and unique physiological structure of the kidney make most antioxidant and anti-inflammatory small molecule drugs ineffective because of the lack of kidney targeting and side ***,nanodrugs with intrinsic kidney targeting through the control of size,shape,and surface properties have opened exciting prospects for the treatment of *** antioxidant nanodrugs have emerged to address the limitations of current AKI *** this review,we systematically summarized for the first time about the emerging nanodrugs that exploit the pathological and physiological features of the kidney to overcome the limitations of traditional small-molecule drugs to achieve high AKI ***,we analyzed the pathological structural characteristics of AKI and the main pathological mechanism of AKI:hypoxia,harmful substance accumulation-induced RONS burst at the renal site despite the multifactorial initiation and heterogeneity of ***,we introduced the strategies used to improve renal targeting and reviewed advances of nanodrugs for AKI:nano-RONS-sacrificial agents,antioxidant nanozymes,and nanocarriers for antioxidants and anti-inflammatory *** nanodrugs have demonstrated excellent therapeutic effects,such as greatly reducing oxidative stress damage,restoring renal function,and low side ***,we discussed the challenges and future directions for translating nanodrugs into clinical AKI treatment.

Background:Atherosclerosis(AS)has been a potentially life-threatening disease *** decoction(AD)is a famous Chinese formula in treating ***,the active components and potential mechanisms remain ***:In this study,the active compositions of AD were analyzed by ultrahigh-performance liquid chromatography quadrupole-orbitrap high-resolution mass spectrometry(UHPLC-Q-Orbitrap-HRMS).The putative targets were predicted and interpreted by a series of bio-informative tools such as Venn,STRING,NetworkAnalyzer,ClusterMarker,and ***,the results were validated by molecular docking using ***:The results showed that alisol A,alisol B,23-o-Acetylalisol B,atractylenolideⅢand atractylenolideⅡwere the major compositions in *** targets mainly distributed in three gene clusters were obtained after topological and cluster *** and GO analyses showed that genes in cluster 1 mainly participated in adipocytokine signaling pathway and played an important role in lipid metabolism,while genes in cluster 2 had regulatory actions via cAMP signaling pathway and may protect vascular function from AS,and genes in cluster 3 were closely related to ***,RXRA,AGT and CXCL8,the central gene of cluster 1,2 and 3,were verified to be strong binding with some major compositions in ***:The possible mechanisms of AD in the treatment of AS may be closely correlated with the regulation of lipid metabolism,vascular physiology and *** study provided a new insight into the anti-AS effect of AD,but more pharmacological experiments should be performed for verification in the future.

Current challenges in the development of scaffolds for bone regeneration include the engineering of biomaterials that can withstand a natural dynamic physiology on the bone that provides a matrix capable of supporting cell migration and tissue ingrowth. The objective of the present work was to develop and characterize a new biomembrane—Mineralized Exoskeleton Shrimp (MES) developed from the exoskeleton of paleomonetes. The integration of MES as a biomaterial for tissue regeneration relies on the growing evidence that the shrimp is characterized by a hierarchically arranged chitin fiber structure, mineralized predominately by calcium carbonate and/or calcium phosphate, bringing beneficial effects in bone regeneration. Additionally, the tridimensional MES structure, can act as a “tent” for Guided Bone Regeneration (GBR). Recently, our team has characterized the MES biomaterial by in vitro (human osteoblastic cellular cultures and immersion of the membrane in modified synthetic plasma) and in vivo (soft tissue in lab mice and hard tissue in rabbit model). The cellular growth in the MES membrane was very exuberant in cellular culture with osteoblastic colonization on its surface (histophilic and biocompatible). After the immersion in modified synthetic plasma for one week, a mass mineralization occurred throughout the membrane’s surface (bioactive). The analysis of histological samples from experimental surgery in lab mice showed that the MES membrane wasn’t toxic to soft tissues and that it caused a moderate inflammatory response (first reabsorption signs at 8 weeks). The MES could act as a cell-guiding template that contains the necessary cues and adequate three-dimensional set to facilitate cell adhesion and promote tissue regeneration upon implantation and subsequent biodegradation.

Background:Naringin as a bioflavanone glycoside is abundant in citrus fruits,which since ancient times have been utilized as natural herbal treatments in traditional *** has potent antioxidant effect that may mitigate oxidative stress mediated cardiovascular and renal dysfunctions in mammalian ***:To explore probable modulatory actions of naringin on fipronil-induced cardio-renal ***:Twenty-four Wistar rats weighing 140±3.0 g were divided into four groups including normal control(saline),10 mg/kg Fipronil(FIP),FIP+100 mg/kg naringin,and 100 mg/kg *** of naringin and fipronil was by oral gavage for 28 consecutive ***,blood pressure monitoring,biochemical assays,histology,and immunohistochemical evaluations were ***:Naringin significantly(P<0.001)lessened blood pressure parameters,oxidative stress markers but significantly(P<0.001)heightened the systemic ***,naringin prevented fipronil-induced histopathological lesions such as congestion and cellular infiltration in cardiac and renal ***,naringin attenuated the immunohistochemical expression of troponin 1 and matrix metalloperoxidase-2 in cardiac tissues,but heightened angiotensin converting enzyme 2 expression in contrast to that of fipronil ***:The observation in this study shows potent attenuation of fipronil-induced toxicity by naringin,through inhibiting processes related to oxidation and inflammation.
