Changes in muscle strength and risk of cardiovascular disease among middle-aged and older adults in China:Evidence from a prospective cohort study

Background:Evidence indicates that low muscle strength is associated with an increased cardiovascular diseases(CVDs)***,the association between muscle strength changes based on repeated measurements and CVD incidence remains ***:The study used data from the China Health and Retirement Longitudinal Study in 2011(Wave 1),2013(Wave 2),2015(Wave 3),and 2018(Wave 4).Low muscle strength was defined as handgrip strength<28 kg for men or<18 kg for women,or chair-rising time≥12 *** on changes in muscle strength from Waves 1 to 2,participants were categorized into four groups of Normal-Normal,Low-Normal,Normal-Low,and *** events,including heart disease and stroke,were recorded using a self-reported questionnaire during Waves 3 and 4 *** proportional hazards models were used to investigate the association between muscle strength changes and CVD incidence after multivariable *** ratios(HRs)and 95%confidence intervals(95%CIs)were estimated with the Normal-Normal group as the ***:A total of 1164 CVD cases were identified among 6608 *** to participants with sustained normal muscle strength,the CVD risks increased progressively across groups of the Low-Normal(HR=1.20,95%CI:1.01-1.43),the Normal-Low(HR=1.35,95%CI:1.14-1.60),and the Low-Low(HR=1.76,95%CI:1.49-2.07).Similar patterns were observed for the significant associations between muscle strength status and the incidence risks of heart disease and *** analyses showed that the significant associations between CVD and muscle strength changes were consistent across age,sex,and body mass index(BMI)***:The study found that muscle strength changes were associated with CVD *** suggests that continuous tracking of muscle status may be helpful in screening cardiovascular risk.

Currently,there is no effective treatment for amyotrophic lateral sclerosis(ALS),despite the limited efficacy of riluzole[1]and edaravone[2].SOD1(coding for the Cu/Zn superoxide dismutase)is the second most frequent genetic cause for ALS only after C9orf72 in patients with European ancestry while being the most frequent in Asian ALS populations[3],Multiple therapeutic approaches have targeted SOD1-related ALS,including the antisense oligonucleotide tofersen with promising results in a recent phase I/II trial[4].

Breast cancerhas been one of the biggest killers of women due to its susceptibility and high *** observations show that malignant cancer cells frequently invade the surrounding normal tissue in collective rather than individual cell migration.1,2 For individual cell migration,it has been found that the Rho/ROCK signaling is upregulated and correlates with disease progression.3 Meanwhile,Rho activates myosin-ll and the actomyosin-mediated contraction creates tension within the cells.4,5 However,the roles of the activation of the Rho/RoCK signal pathway in collective cell migration and the precise mechanisms by which myosin-ll fine-tunes the contractility of the cells to allow for the reorganization of the cytoskeleton that drives collective cell migration,remain *** study investigated whether the high cellular contractility could activate the inside-out signal transductions and how did the intracellular force contribute to the collective cell migration.

Clinical characteristics of critical patients with pandemic influenza A(H1N1) virus infection in Chengdu,China

Objective： The critical illness of pandemic influenza A （H1N1） virus infection may be associated with relatively poor outcomes. The objective of this study is to describe clinical features and factors associated with the deaths of critical patients. Methods： Medical records of 26 critical patients with H1N1 infection admitted from Sept. 1 to Dec. 31, 2009, were retrospectively reviewed. Diagnosis was established by real-time reverse transcdptase- polymerase chain reaction （RT-PCR） assay. Results： The mean age of the patients was （40.4＋18.4） years and 73.1% of them were male. Clinical manifestations included fever, cough, and sputum production. The laboratory findings included leukocytosis, lymphopenia, C-reaction protein, and lactic dehydrogenase elevation. In this series, 17 subjects survived and 9 died. The parameters between the deaths and survivors were compared, which included acute physiology and chronic health evaluation II （APACHE II） scores （23.8＋10.1 vs. 14.3_＋6.6, P〈0.05）, sequential organ failure assessment （SOFA） scores （13.3_＋3.0 vs. 6.6_＋3.3, P〈0.05）, and multiple organ dysfunction syndrome （MODS） scores （7.4＋2.5 vs. 3.3＋1.7, P〈0.05）. The cases of deaths had higher incidences of cardiovascular failure （100% vs. 41.2%, P〈0.05）, renal failure （55.6% vs. 11.7%, P〈0.05）, encephalopathy （44.4% vs. 5.9%, P〈0.05）, hepatic failure （33.3% vs. 5.9%, P〈0.05）, and septic shock （33.3% vs. 17.6%, P〈0.05）. Conclusions： The critical patients with H1 N1 infection have high APACHE II, SOFA, and MODS scores, which may be associated with an increased risk of death and complex clinical courses.

Melatonin alleviates lipopolysaccharide-compromised integrity of blood brain barrier by activation of AMP-activated protein kinase in old mice

The integrity and function dysfunction of the blood brain barrier(BBB)is considered to be an early event in the pathogenesis of a variety of neurological diseases in old patients and dysfunction of BBB could be induced bycommon stress that old people often face,such as sepsis during which lipopolysaccharide(LPS)is released into circulation and BBB is *** decreased melatonin level had been shown in the serum and brain of old people and mice,we aim to investigate whether supplement with melatonin could alleviate LPSinduced BBB damage in old ***(24-28 months old)received one week melatonin(10 mg·kg-1·d-1,intraperitoneally,ip)or saline before challenge with LPS(1 mg·kg-1,ip).Evan′s blue(EB)and immunoglobulin G(Ig G)leakage were used to assess the BBB *** and Western blotting were used to detect the protein expression and *** results showed that LPS significantly increased BBB permeability in old mice accompanied by tight junction protein occludin and claudin-5 degradation,inhibition of AMP-activated protein kinase(AMPK)activation and increase of gp91phox protein ***,one week of melatonin treatment significantly decreased LPS-induced BBB hyperpermeability,activated AMPK and inhibited gp91phox expression ***,activation of AMPK by metformin significantly inhibited LPS-induced gp91phox expression upregualtion in endothelial *** together,our findings demonstrate that melatonin alleviates LPS-impaired integrity of BBB by activation of AMPK and inhibition of gp91phox expression in old mice.

Constructive thinking,rational intelligence and irritable bowel syndrome

AIM： To evaluate rational and experiential intelligence in irritable bowel syndrome （IBS） sufferers. METHODS： We recruited 100 subjects with IBS as per Rome 11 criteria （50 consulters and 50 non-consulters） and 100 healthy controls, matched by age, sex and educational level. Cases and controls completed a clinical questionnaire （including symptom characteristics and medical consultation） and the following tests： rational- intelligence （Wechsler Adult Intelligence Scale, 3rd edition）; experiential-intelligence （Constructive Thinking Inventory）; personality （NEO personality inventory）; psychopathology （MMPI-2）, anxiety （state-trait anxiety inventory） and life events （social readjustment rating scale）. Analysis of variance was used to compare the test results of IBS-sufferers and controls, and a logistic regression model was then constructed and adjusted for age, sex and educational level to evaluate any possible association with IBS. RESULTS： No differences were found between IBS cases and controls in terms of IQ （102.0 ± 10.8 vs 102.8 ± 12.6）, but IBS sufferers scored significantly lower in global constructive thinking （43.7± 9.4 vs 49.6 ± 9.7）. In the logistic regression model, global constructive thinking score was independently linked to suffering from IBS [OR 0.92 （0.87-0.97）], without significant OR for total IQ. CONCLUSION： IBS subjects do not show lower rational intelligence than controls, but lower experiential intelligence is nevertheless associated with IBS.
