The multifunctional trans-activator Tat is an essential regulatory protein for HIV-1 replication and is characterized by high sequence diversity. Numerous experimental studies have examined Tat in HIV-1 subtype B, but research on subtype C Tat is lacking, despite the high prevalence of infections caused by subtype C worldwide. We hypothesized that amino acid differences contribute to functional differences among Tat proteins. In the present study, we found that subtype B NL4-3Tat and subtype C isolate HIV1084 i Tat exhibited differences in stability by overexpressing the fusion protein Tat-Flag. In addition, 1084 i Tat can activate LTR and NF-κB more efficiently than NL4-3 Tat. In analyses of the activities of the truncated forms of Tat, we found that the carboxylterminal region of Tat regulates its stability and transactivity. According to our results, we speculated that the differences in stability between B-Tat and C-Tat result in differences in transactivation ability.

Here we explored the contribution of atlastin- 1 （ATL 1）to HIV replication for the first time. HIV-1 encodes only15 proteins （Ayinde et al., 2010） and thus must exploitmultiple host cell functions for successful infection. Forexample, HIV-1 envelope （Env） glycoproteins are synthe-sized as a polyprotein precursor, known as gp 160, in theendoplasmic reticulum （ER）（Checkley et al., 2011）。

Dear Editor,In the late stages of the retroviral life cycle,the transcription of viral pre-m RNA is initiated at the 5′long terminal repeat(5′LTR)and terminated at the 3′LTR;the full-length and spliced viral RNAs are transported out of the nucleus and serve as templates for the translation of viral proteins or alternatively as the full-length viral genome to be packaged into virus particles.

The envelope protein(Env) of lentiviruses such as HIV,SIV,FIV and EIAV is larger than that of other retroviruses.The Chinese EIAV attenuated vaccine is based on Env and has helped to successfully control this virus,demonstrating that envelope is crucial for vaccine.We compared Env variation of the four kinds of lentiviruses.Phylogenetic analysis showed that the evolutionary relationship of Env between HIV and SIV was the closest and they appeared to descend from a common ancestor,and the relationship of HIV and EIAV was the furthest.EIAV had the shortest Env length and the least number of potential N-linked glycosylation sites(PNGS) as well as glycosylation density compared to various immunodeficiency viruses.However,HIV had the longest Env length and the most PNGS.Moreover,the alignment of HIV and SIV showed that PNGS were primarily distributed within extracellular membrane protein gp120 rather than transmembrane gp41.It implies that the size difference among these viruses is associated with a lentivirus specific function and also the diversity of env.There are low levels of modification of glycosylation sites of Env and selection of optimal protective epitopes might be useful for development of an effective vaccine against HIV/AIDS.

Perinatal transmission of Human immunodeficiency virus(HIV),also called mother-to-child transmission(MTCT),accounts for 90% of infections in infants worldwide and occurs in 30%-45% of children born to untreated HIV-1 infected mothers.Among HIV-1 infected mothers,some viruses are transmitted from mothers to their infants while others are not.The relationship between virologic properties and the pathogenesis caused by HIV-1 remains unclear.Previous studies have demonstrated that one obvious source of selective pressure in the perinatal transmission of HIV-1 is maternal neutralizing antibodies.Recent studies have shown that viruses which are successfully transmitted to the child have growth advantages over those not transmitted,when those two viruses are grown together.Furthermore,the higher fitness is determined by the gp120 protein of the virus envelope.This suggests that the selective transmission of viruses with higher fitness occurred exclusively,regardless of transmission routes.There are many factors contributing to the selective transmission and HIV replicative fitness is an important one that should not be neglected.This review summarizes current knowledge of the role of HIV replicative fitness in HIV MTCT transmission and the determinants of viral fitness upon MTCT.

Alzheimer's disease (AD),also known as Alzheimer's,is a chronic neurodegenerative disorder with hallmark amyloid plaques in brain tissue.The diseases commences slowly and worsens over time (Sjogren et al.1952).Although it has been investigated for over six decades,the cause of AD is still not completely understood.It was initially thought to be caused by genetic heritability (autosomal mutation leading to aggregation of β-amyloid and misfolded tau proteins)or head injury.However,hundreds of drug trials targeting the accumulation of sticky amyloid plaques and misfolded tau proteins in the brain have failed to yield any results.

The iron chelator deferoxamine has been shown to inhibit ferroptosis in spinal cord injury.However,it is unclear whether deferoxamine directly protects neurons from ferroptotic cell death.By comparing the survival rate and morphology of primary neurons and SH-SY5Y cells exposed to erastin,it was found that these cell types respond differentially to the duration and concentration of erastin treatment.Therefore,we studied the mechanisms of ferroptosis using primary cortical neurons from E16 mouse embryos.After treatment with 50μM erastin for 48 hours,reactive oxygen species levels increased,and the expression of the cystine/glutamate antiporter system light chain and glutathione peroxidase 4 decreased.Pretreatment with deferoxamine for 12 hours inhibited these changes,reduced cell death,and ameliorated cellular morphology.Pretreatment with the apoptosis inhibitor Z-DEVD-FMK or the necroptosis inhibitor necrostain-1 for 12 hours did not protect against erastin-induced ferroptosis.Only deferoxamine protected the primary cortical neurons from ferroptosis induced by erastin,confirming the specificity of the in vitro ferroptosis model.This study was approved by the Animal Ethics Committee at the Institute of Radiation medicine of the Chinese Academy of medical Sciences,China(approval No.DWLL-20180913)on September 13,2018.

Among various genera of free-living amoebae prevalent in nature,some members are identified as causative agents of human encephalitis,in which Naegleria fowleri followed by Acanthamoeba spp.and Balamuthia mandrillaris have been successively discovered.As the three dominant genera responsible for infections,Acanthamoeba and Balamuthia work as opportunistic pathogens of granulomatous amoebic encephalitis in immunocompetent and immunocompromised individuals,whereas Naegleria induces primary amoebic meningoencephalitis mostly in healthy children and young adults as a more violent and deadly disease.Due to the lack of typical symptoms and laboratory findings,all these amoebic encephalitic diseases are difficult to diagnose.Considering that subsequent therapies are also affected,all these brain infections cause significant mortality worldwide,with more than 90%of the cases being fatal.Along with global warming and population explosion,expanding areas of human and amoebae activity in some regions lead to increased contact,resulting in more serious infections and drawing increased public attention.In this review,we summarize the present information of these pathogenic free-living amoebae,including their phylogeny,classification,biology,and ecology.The mechanisms of pathogenesis,immunology,pathophysiology,clinical manifestations,epidemiology,diagnosis,and therapies are also discussed.

Granulocyte macrophage colony stimulating factor(GM-CSF)is generally considered to be a proinflammatory cytokine;GM-CSF-based vaccines have been shown to elicit potent antitumor and antiviral immune responses in preclinical experiments.In clinical trials,however,these effects were not as robust and sometimes even contradicted the findings from animal models.In this commentary,we aim to discuss the latest advances in this field,hoping that an upto-date and comprehensive understanding of this molecule will help others design effective GM-CSF-based strategies for antiviral and anticancer vaccines.
