LRP1 facilitates hepatic glycogenesis by improving the insulin signaling pathway in HFD-fed mice

Background: LDL receptor-related protein-1(LRP1) is a cell-surface receptor that functions in diverse physiological pathways. We previously demonstrated that hepatocyte-specific LRP1 deficiency(hLRP1KO) promotes diet-induced insulin resistance and increases hepatic gluconeogenesis in mice. However, it remains unclear whether LRP1 regulates hepatic ***: Insulin signaling, glycogenic gene expression, and glycogen content were assessed in mice and HepG2 cells. The pcDNA 3.1 plasmid and adeno-associated virus serotype 8 vector(AAV8) were used to overexpress the truncated β-chain(βΔ) of LRP1 both in vitro and in ***: On a normal chow diet, hLRP1KO mice exhibited impaired insulin signaling and decreased glycogen content. Moreover, LRP1 expression in HepG2 cells was significantly repressed by palmitate in a dose-and time-dependent manner. Both LRP1 knockdown and palmitate treatment led to reduced phosphorylation of Akt and GSK3β, increased levels of phosphorylated glycogen synthase(GYS), and diminished glycogen synthesis in insulin-stimulated HepG2 cells, which was restored by exogenous expression of the βΔ-chain. By contrast, AAV8-mediated hepatic βΔ-chain overexpression significantly improved the insulin signaling pathway, thus activating glycogenesis and enhancing glycogen storage in the livers of high-fat diet(HFD)-fed ***: Our data revealed that LRP1, especially its β-chain, facilitates hepatic glycogenesis by improving the insulin signaling pathway, suggesting a new therapeutic strategy for hepatic insulin resistance-related diseases.

Background and Aims:Stress granules(SGs)as membrane-less cytoplasmic foci formed in response to unfavorable external stimuli could promote cancer cells to adapt to hostile *** carcinoma(HCC)is prone to be highly aggressive once diagnosed,which markedly reduces patient survival ***,it is crucial to develop valid diagnostic markers to prognosticate HCC patient prognosis,which promotes individualized precision therapeutics in *** the pro-tumorigenic activity of SGs,it is of great potential value to construct a prognostic tool for HCC based on the expression profiles of SG-related genes(SGGs).Methods:Bioinformatic analysis was employed to establish an SGG-based prognostic *** blotting and realtime polymerase chain reaction assays were used to assess the expression patterns of the related ***-of-function experiments were performed to analyze the effect of the SGGs on SG formation and cell ***:A four-SGG signature(KPNA2,MEX3A,WDR62,and SFN)targeting HCC was established and validated to exhibit a robust performance in predicting HCC ***,all four genes were further found to be highly expressed in human HCC *** important,we demonstrated that individually knocking down the four SGGs significantly reduced HCC cell proliferation and metastasis by compromising the SG formation ***:We developed an SGG-based predictive signature that can be used as an independent prognostic tool for *** strong predictive power of this signature was further elucidated by the carcinogenic activity of KPNA2,MEX3A,WDR62,and SFN in HCC cells by regulating SG formation.

Islet transplantation is a promising strategy for diabetes mellitus treatment as it can recapitulate endogenous insulin secretion and provide long-term glycemic *** models constructed in biomaterial scaffolds that reproduce biological characteristics of native islets is a feasible option to circumvent the dilemma of donor shortage and the requirement of chronic ***,we developed bioinspired artificial microcapsule-based islet models with microvessels for glycemic control using microfluidic electrospray *** electrospray can generate uniform hydrogel microcapsules with core-shell structure for encapsulating islet *** cell-laden microcapsules enabled the efficient transportation of nutrient,oxygen,and insulin;as well as the incorporation with microvessels for prompting glucose responsiveness and molecular *** demonstrated by in vivo experiments that the blood glucose,food intake,and body weight of diabetic mouse models were alleviated,and the glucose tolerance was promoted after the engraftment of islet *** further demonstrated the improved functionality of transplanted islet model in insulin secretion,immune escape,and microcirculation using standard histological and molecular *** results indicated that the microcapsules with microvessels are promising artificial islet models and are valuable for treating diabetes.

Intermittent fasting can benefit breast cancer patients undergoing chemotherapy or ***,it is still uncertain how to select immunotherapy drugs to combine with intermittent *** we observed that two cycles of fasting treatment significantly inhibited breast tumor growth and lung tissue metastasis,as well as prolonged overall survival in mice bearing 4T1 and 4T07 breast *** this process,both the immunosuppressive monocytic-(M-)and granulocytic-(G-)myeloid-derived suppressor cell(MDSC)decreased,accompanied by an increase in interleukin(IL)7R^(+)and granzyme B^(+)T cells in the tumor ***,we observed that Ly6G^(low)G-MDSC sharply decreased after fasting treatment,and the cell surface markers and protein mass spectrometry data showed potential therapeutic *** investigation revealed that glucose metabolism restriction suppressed the splenic granulocytemonocyte progenitor and the generation of colony-stimulating factors and IL-6,which both contributed to the accumulation of *** the other hand,glucose metabolism restriction can directly induce the apoptosis of Ly6G^(low)G-MDSC,but not Ly6G^(high)*** summary,these results suggest that glucose metabolism restriction induced by fasting treatment attenuates the immune-suppressive milieu and enhances the activation of CD3^(+)T cells,providing potential solutions for enhancing immune-based cancer interventions.

In addition to their pivotal roles in energy storage and expenditure,adipose tissues play a crucial part in the secretion of bioactive molecules,including peptides,lipids,metabolites,and extracellular vesicles,in response to physiological stimulation and met-abolic *** secretory factors,through autocrine and paracrine mechanisms,regulate various processes within adipose *** processes include adipogenesis,glucose and lipid metabolism,inflammation,and adaptive thermogenesis,all of which are essential for the maintenance of the balance and functionality of the adipose tissue micro-environment.A subset of these adipose-derived secretory factors can enter the circulation and target the distant tissues to regulate appetite,cognitive function,energy expenditure,insulin secretion and sensitivity,gluconeogenesis,cardiovascular remodeling,and exercise *** this review,we highlight the role of adipose-derived secretory factors and their signaling pathways in modulating meta-bolic ***,we delve into the alterations in both the content and secretion processes of these factors under various physiological and pathological conditions,shedding light on potential pharmacological treatment strategies for related diseases.

The influence of Lactobacillus plantarum fermentation in selenium-enriched Brassica napus L.:changes in the nutritional constituents,bioactivities and bioaccessibility

Selenium(Se)-enriched Brassica napus *** a valuable organic Se *** this study,the fermentation broth enriched with organic Se(FFS)was prepared using Lactobacillus plantarum to ferment the substrate of Se-enriched Brassica napus *** increases were observed after fermentation in total sugars,reducing sugars,soluble proteins,total phenolic content(TPC),and total flavonoid content(TFC).The organic Se was retained at a concentration of 54.75 mg/g in the freeze-dried *** component analysis and cluster analysis showed good separation between the FFS and unfermented(FS)*** 2-ethyloxetane had the highest content among all volatiles,while sinapine had the highest content among all phenolic *** fermentation process showed remarkable improvement in the abundance and concentration of volatile compounds and phenolic contents,making FFS exhibit strong antioxidant activity and inhibitory capacity againstα-glucosidase *** bioaccessibility of phenolic compounds was significantly greater in FFS compared to *** analysis revealed that the majority of phenolic compounds contained in FFS did not exhibit mutagenicity toxicity,hepatotoxicity,skin sensitization,or blood-brain barrier penetration,indicating a favorable level of ***,our study provides a new insight into the further utilization of Se-enriched Brassica napus *** foods.

Fluoro-functionalized plant biomass adsorbent:Preparation and application in extraction of trace perfluorinated compounds from environmental water samples

Perfuorinated compounds(PFCs)are toxic and widely present in the environment,and therefore effective adsorbents are required to remove PFCs from environmental *** the present study,a new type of fuorinated biomass materials was synthesized via an ingenious fuorosilanization *** adsorbents were applied for the adsorption of 13typical PFCs,including perfuorocarboxylic acids(PFCAs)and perfuorosulfonic acids(PFSAs).By comparing their adsorption performance,Fluorinated cedar slag(FCS)was discovered to have the best absorption efficiency and enabled highly efficient enrichment of *** adsorption recovery of FCS with the investigated PFCs is greater than 90%under the optimal adsorption *** to the high affinity of F-F sorbent-sorbate interaction,FCS had good adsorption capacities of PFCs from aqueous solution,with the maximum adsorption capacity of 15.80 mg/g for PFOS and 10.71 mg/g for PFOA,***,the adsorption time could be achieved in a short time(8 min).Using the FCS absorbent,an innovative FCS-solid phase extraction assisted with high performance liquid chromatography-electrospray-tandem mass spectrometry(FCS-SPE-HPLC-ESI-MS/MS)method was first developed to sensitively detect PFCs in the environmental water *** intra-day and inter-day recovery rates of the 13 compounds ranged from 90.7%-104.3%,with the RSD of 2.1%-4.7%(intra-day)and 2.5%-8.5%(inter-day),*** research demonstrates the potential of the newly fuoro-functionalized plant biomass to adsorb PFCs from environmental water,with the advantages of high adsorption efficiencies,high antiinterference,easy operation and low economic cost.

Effect of defatted flaxseed gum powder addition on the quality of sesame paste

The study explored the influence of defatted flaxseed gum powder(DFGP) on the stability and quality of sesame paste by measuring and analyzing its composition, color, texture, particle size, centrifugal oil separation rate,rheological properties, and microstructure. The results showed that the moisture and polysaccharide content of sesame paste was increased as the DFGP increased. Additionally, the hardness, gumminess, and chewiness of the sesame paste was improved, while the presence of particles with small particle size(1–100 μm) was *** rate of oil precipitation was reduced by 28.99% when the amount of DFGP was 6%. The sesame paste samples exhibited pseudoplastic behavior, demonstrating shear thinning. As the shear rate increased, the apparent viscosity of sesame paste gradually decreased. Both the storage modulus(G’) and the loss modulus(G’’) increased as the shear frequency increased.
