Water quality, habitat, and fish assemblage relationships in middle-order agriculture and forest streams of the Mississippi Alluvial Plain

Background Agriculture has greatly influenced water quality, habitats, and fish assemblages in streams of the Mississippi Alluvial Plain(MAP) ecoregion. However, MAP streams have historically been understudied compared to streams in other agricultural regions of the USA. In this study, water quality, habitat, and fish assemblage composition were assessed seasonally(spring, summer, and fall) in eight representative MAP streams located across three U.S. states. The study design included four streams containing highly agricultural watersheds(herein termed “agriculture” streams) and four streams containing mostly forested watersheds(herein termed “forest” streams), which were intended to represent reference conditions for MAP *** In general, forest streams contained significantly better instream and riparian habitats than agriculture streams(P = 0.010–0.040) whereas agriculture streams contained significantly greater levels of primary nutrients(P < 0.001–0.010). Differences between agriculture and forest streams with respect to other physical and chemical variables were intermittent and season dependent. Fish assemblages in agriculture and forest streams were structured primarily along an environmental gradient reflecting instream habitat conditions, water nutrient concentrations, and benthic chlorophyll-a production. Structurally, fish assemblages in both stream types contained many regionally common species, though some species appeared to exhibit affinities for a particular stream type. Functionally, fish assemblages in agriculture streams contained more tolerant species, more omnivores, and fewer insectivores compared to forest stream assemblages, which were nearly all insectivores. Analyses of fin ray types to detect strontium markers in juvenile blunt-snout bream Megalobrama amblycephala

Background Restocking by introducing hatchery-reared fish into wild habitats aids in the restoration of fishery aquatic ecosystems and reefs to increase the abundance of fish resources, restore the ecological balance of water bodies, and enhance ecosystem functioning. Accurately, rapidly, and effectively evaluating the success of restocking using chemical markers（e.g., strontium [Sr]） remains challenging for fisheries management. Consequently, for nonlethal fish sampling, hard tissues, such as fin rays, have received increasing attention as a target for marking method. However, data on the differences in marking different types of fin rays remain limited. Therefore, we exposed juvenile blunt snout bream individuals（Megalobrama amblycephala） to 0（control group） or 800 mg/L of SrCl2·6H2O（marked group） for 5 days and transferred them into normal aerated water for post-immersion culture. We sampled their pectoral, dorsal, ventral, anal, and caudal fin rays. The Sr marks among the fin types were sampled at 0 and 20 days post-immersion and evaluated using an electron probe micro-analyzer（EPMA） for the five-day Sr/Ca ratios, along with line transect and Sr mapping *** Sr marking signatures were observed in all fin types in the marked group, with a success rate of up to 100%. Although marking efficiency varied among the different fin ray types, the highest Sr/Ca ratios were most often detected in the dorsal fin. Cross-sectional Sr concentration maps of all fin rays sampled showed high-Sr domains in the marked group; in contrast, the entire cross-sections of the control group displayed low Sr contents, indicating successful marking *** Fin ray Sr marking is a successful method for juvenile M. amblycephala, with the advantages of nonlethality and negligible sampling injuries, facilitating the rapid and effective evaluation of Sr marking in restocking M. amblycephala.

Water quality and habitat drive phytoplankton taxonomic and functional group patterns in the Yangtze River

Background Although phytoplankton are important primary producers in food webs, they are relatively less studied in large rivers compared to other types of systems. To fill this research gap, we studied phytoplankton taxonomic and functional composition and their relationships with water quality, habitat, climate, and land use across 30 river sections in the middle and lower reaches of the Yangtze River during 2017–*** Major observed phytoplankton groups were cyanobacteria, bacillariophyta, and chlorophyta. Phytoplankton total abundance, total biomass, and species richness significantly decreased in the dry season compared to the wet season, with the species and functional composition differing significantly between seasons. Phytoplankton species differences between seasons were mainly contributed by Oscillatoria sp., Pseudanabaena sp., and Melosira granulata. The dfferences in phytoplankton functional groups between seasons were mainly contributed by P（including Closterium sp., Melosira sp.）, Lo（including Merismopedia sp., Peridinium sp., Ceratium sp., and Gymnodinium sp.）, and J（including Pediastrum sp., Tetraedron sp., Crucigenia sp., Scenedesmus sp., and Coelastrum sp.）. The variance partitioning showed that water quality（NO3-N, total suspended solids, turbidity） and habitat（water flow, river bank and river channel conditions） were critical factors in shaping phytoplankton patterns, followed by climate and land *** Results indicated that there was significant seasonal variation of phytoplankton in the Yangtze River, with water quality and habitat primarily driving phytoplankton patterns. Our study contributes to the understanding of natural and anthropogenic factors that drive seasonal successional processes of phytoplankton in the Yangtze River. These findings have important implications for environmental management as well as towards the ecological restoration of large rivers.

Microplastics(MPs)have become a significant concern for their potential ***,the correlation between the size of plastic particles and their toxicity remains ***,we investigate the toxic effects of different sizes(80 nm,800 nm,8μm and 80μm)polystyrene MPs(PS-MPs)on the model organism Nile tilapia(Oreochromis niloticus).The results of bioluminescent imaging indicate that the 80 nm PS-MPs are more likely to invade the body.H&E staining shows severe damage on the intestinal villi and distinct hepatic steatosis in the 80 nm *** U labeling shows that the proliferation activity of intestinal and liver cells reduces significantly in the 80 nm *** gut microbiome analysis shows a severe imbalance of gut microbiota homeostasis in the 80 nm *** analysis of liver transcriptomics and metabolomics shows that the liver lipid metabolism is disordered in the 80 nm *** conclusion,this study confirms that the 80 nm PS-MPs are more likely to induce intestinal and liver *** the above lay the foundation for further study on the pathological damage of MPs to other organisms.

Both in nature and in aquaculture,fish may experience periods of food scarcity or *** metabolic regulation of fish when nutritional state changes is a complex process that involves many *** study glucose metabolism adaptability during fasting and re-feeding in the black carp(Mylopharyngodon piceus),we measured changes in some biochemical indicators related to glucose *** fish were sampled on days 0,1,3,5,and 10 of fasting(F,S1,S3,S5,and S10,respectively)and days 1,3,and 5 of re-feeding(RF1,RF3,and RF5,respectively).The serum glucose concentration decreased significantly at S1,reached the lowest point at S10,and increased significantly at RF1(P<0.05).The concentration of liver glycogen decreased significantly at S1 and reached the lowest level at S3,whereas the muscle glycogen level decreased significantly at S5 and reached the lowest value at S10(P<0.05).Both liver and muscle glycogen levels returned to the pre-fasting level at RF5(P<0.05).Regarding glycolysis,the concentrations of pyruvate kinase(PK)and hexokinase(HK)decreased significantly at S5 and increased significantly at RF5 and RF1,respectively(P<0.05).The concentrations of glucokinase(GCK)and insulin decreased significantly at S1 and increased significantly at RF1 and RF3,respectively(P<0.05).The mRNA expression levels of liver GCK and glucose transporter 2(GLUT2)decreased significantly at S1 and increased significantly at RF1 and RF5,respectively(P<0.05).As for gluconeogenesis,the concentration of glucose-6-phosphatase(G6PC)increased significantly at S1 and decreased significantly at RF1(P<0.05).The concentrations of glucagon and glucocorticoid(GC)increased significantly at S3 and significantly decreased at RF1 and RF5,respectively(P<0.05).The mRNA expression levels of liver G6PC and phosphoenolpyruvate carboxykinase(PEPCK)increased significantly at S3 and S1,and both decreased significantly at RF1(P<0.05).These results indicate that coordination between glycolysis and gluconeogen

Responding of zooplankton to environmental factor changes in the Changjiang River estuarine regions in spring-summer from 2016 to 2020

Coastal and estuarine protists are frequently exposed to salinity *** the tolerance and stress responses of microalgae to salinity have been extensively studied,there have been scarce studies on the physiological response of heterotrophic protists to salinity *** this study,we investigated the physiological response of the heterotrophic ciliate Gastrostyla setifera to a salinity of 3,via a transcriptomic *** first transcriptome of genus Gastrostyla was obtained utilizing a group of manually isolated ciliate individuals(cells)and RNA-seq *** completeness of the transcriptome was *** expressed gene(DEG)analysis was performed among the transcriptomes of *** acclimated in saline water(salinity 3)and those cultured in fresh *** results demonstrated a significant alternation in gene transcription,in which the ciliate exhibits a transcripttomic acclimation in responding salinity *** up-regulated DEGs were enriched in the pathways of cytoskeleton proteins,membrane trafficking,protein kinases and protein *** may represent enhanced functions of ion transport,stress response and cell *** involved in energy metabolism and biosynthesis were markedly down-regulated,reflecting decreased cell ***,we detected significantly down-regulated genes involved in several pathways of amino acid catabolism,which may lead to accumulation of amino acids in the ciliate *** acid could act as compatible solutes in the cytoplasm to maintain the osmotic balance in saline ***,this work is an initial exploration to the molecular basis of the heterotrophic protist responding to salinity *** result sheds light on the mechanisms of enhancement of cell protection,reduction of cell activity,and osmotic pressure regulation in ciliates acclimated to salinity.

The genus Silurus,an important group of catfish,exhibits heterogeneous distribution in Eurasian freshwater *** group includes economically important and endangered species,thereby attracting considerable scientific *** this interest,the lack of a comprehensive phylogenetic framework impedes our understanding of the mechanisms underlying the extensive diversity found within this ***,we analyzed 89 newly sequenced and 20 previously published mitochondrial genomes(mitogenomes)from 13 morphological species to reconstruct the phylogenetic relationships,biogeographic history,and species diversity of *** phylogenetic reconstructions identified eight clades,supported by both maximum-likelihood and Bayesian ***-based species delimitation analyses yielded multiple molecular operational taxonomic units(MOTUs)in several taxa,including the Silurus asotus complex(four MOTUs)and Silurus microdorsalis(two MOTUs),suggesting that species diversity is underestimated in the genus.A reconstructed time-calibrated tree of Silurus species provided an age estimate of the most recent common ancestor of approximately 37.61 million years ago(Ma),with divergences among clades within the genus occurring between 11.56 Ma and 29.44 Ma,and divergences among MOTUs within species occurring between 3.71 Ma and 11.56 *** reconstructions suggested that the ancestral area for the genus likely encompassed China and the Korean Peninsula,with multiple inferred dispersal events to Europe and Central and Western Asia between 21.78 Ma and 26.67 Ma and to Japan between 2.51 Ma and 18.42 *** factors such as the Eocene-Oligocene extinction event,onset and intensification of the monsoon system,and glacial cycles associated with sea-level fluctuations have likely played significant roles in shaping the evolutionary history of the genus Silurus.

Testosterone is closely associated with lipid metabolism and known to affect body fat composition and muscle mass in ***,the mechanisms by which testosterone acts on lipid metabolism are not yet fully understood,especially in *** this study,cyp17a1-/-zebrafish(Danio rerio)exhibited excessive visceral adipose tissue(VAT),lipid content,and up-regulated expression and activity of hepatic de novo lipogenesis(DNL)*** assay for transposase accessible chromatinwithsequencing(ATAC-seq)results demonstrated that chromatin accessibility of DNL genes was increased in cyp17a1-/-fish compared to cyp17a1+/+male fish,including stearoyl-CoA desaturase(scd)and fatty acid synthase(fasn).Androgen response element(ARE)motifs in the androgen signaling pathway were significantly enriched in cyp17a1+/+male fish but not in cyp17a1-/-*** androgen receptor(ar)-/-and wildtype(WT)zebrafish administered with Ar antagonist flutamide displayed excessive visceral adipose tissue,lipid content,and up-regulated expression and activity of hepatic de novo lipogenesis *** Ar agonist BMS-564929 reduced the content of VAT and lipid content,and down-regulated acetyl-CoA carboxylase a(acaca),fasn,and scd ***,the rescue effect of testosterone on cyp17a1-/-fish in terms of phenotypes was abolished when ar was additionally ***,these findings reveal that testosterone inhibits lipid deposition by down-regulating DNL genes via Ar in zebrafish,thus expanding our understanding of the relationship between testosterone and lipid metabolism in teleosts.
