Analyses of fin ray types to detect strontium markers in juvenile blunt-snout bream Megalobrama amblycephala

Background Restocking by introducing hatchery-reared fish into wild habitats aids in the restoration of fishery aquatic ecosystems and reefs to increase the abundance of fish resources, restore the ecological balance of water bodies, and enhance ecosystem functioning. Accurately, rapidly, and effectively evaluating the success of restocking using chemical markers（e.g., strontium [Sr]） remains challenging for fisheries management. Consequently, for nonlethal fish sampling, hard tissues, such as fin rays, have received increasing attention as a target for marking method. However, data on the differences in marking different types of fin rays remain limited. Therefore, we exposed juvenile blunt snout bream individuals（Megalobrama amblycephala） to 0（control group） or 800 mg/L of SrCl2·6H2O（marked group） for 5 days and transferred them into normal aerated water for post-immersion culture. We sampled their pectoral, dorsal, ventral, anal, and caudal fin rays. The Sr marks among the fin types were sampled at 0 and 20 days post-immersion and evaluated using an electron probe micro-analyzer（EPMA） for the five-day Sr/Ca ratios, along with line transect and Sr mapping *** Sr marking signatures were observed in all fin types in the marked group, with a success rate of up to 100%. Although marking efficiency varied among the different fin ray types, the highest Sr/Ca ratios were most often detected in the dorsal fin. Cross-sectional Sr concentration maps of all fin rays sampled showed high-Sr domains in the marked group; in contrast, the entire cross-sections of the control group displayed low Sr contents, indicating successful marking *** Fin ray Sr marking is a successful method for juvenile M. amblycephala, with the advantages of nonlethality and negligible sampling injuries, facilitating the rapid and effective evaluation of Sr marking in restocking M. amblycephala.

Migration patterns and habitat use of the tapertail anchovy Coilia mystus in the Oujiang River Estuary and the Zhujiang River Estuary, China

Habitat use of the tapertail anchovy(Coilia mystus Linnaeus, 1758) from the Oujiang River Estuary and the Zhujiang(Pearl) River Estuary was studied by examining the environmental signatures of Sr and Ca in otoliths using electron probe microanalysis. Individuals from the Oujiang River had higher and varied Sr:Ca ratios(expressed as(Sr:Ca)×1 000, 3.83–13.0 average) in the otolith core regions, suggesting that they were born in brackish or sea waters, and that a freshwater habitat might not be necessary for egg hatching and larval ***, individuals from the Zhujiang River had lower Sr:Ca ratios(0.39–2.51 average) in the core regions,suggesting a freshwater origin. After hatching, anchovies from the Zhujiang River migrate downstream to the river estuary close to brackish water. Our results demonstrated varied habitat use for spawning during stages of early life history between the two populations, and suggested that such variations are promoting diversity of life history strategies of this species.

Unravelling habitat use of Coilia nasus from the Rokkaku River and Chikugo River estuaries of Japan by otolith strontium and calcium

The migratory history of the engraulid fish Coilia nasus in the Rokkaku and Chikugo River estuaries of the Ariake Sea, Japan was assessed using otolith strontium（Sr） X-ray intensity maps and strontium：calcium（Sr：Ca） ratio life history transects by an electron probe microanalyzer（EPMA）. The results showed that seven of the ten specimens from the Rokkaku River Estuary（LJC） and all 15 specimens collected in the Chikugo River Estuary（ZHC） had low Sr：Ca ratios（≤3） at the central otolith area, indicating their riverine origin and initial freshwater residence. After the first regime shift adjacent to natal regions, the Sr level mapping displayed a wide variety of color patterns, and the Sr：Ca ratios obtained by the line transect analysis could be divided into one to six significantly different phases indicative of gradual life history transition. The other three specimens from the Rokkaku River Estuary had high Sr：Ca ratios（3–6.7） at the central otolith area but showed alternating changes between low and high values outside the natal region, suggesting that estuarine-origin individuals occurred in the Rokkaku River Estuary. The two-dimensional maps of the Sr level and average of the otolith Sr：Ca ratios along the life-history transects could be used as effective tools for reconstruction of past habitat use of the tapertail anchovy in estuaries of the Ariake Sea, Japan.

Early life migration and population discrimination of the small yellow croaker Larimichthys polyactis from the Yellow Sea: inferences from otolith Sr/Ca ratios

The small yellow croaker Larimichthys polyactis is a benthic marine fish species of high ecological and commercial importance and is widely distributed in the northwestern Pacifi c Ocean,especially in the Chinese coastal waters of the Bohai,Yellow,and East China *** a highly migratory species,the whole life migration of *** has been intensively ***,knowledge about its early life migration is scarce,and population divisions are inconsistent,limiting the ability of fishery scientists and administrators to evaluate the design and potential benefits of thorough conservation and resource-management *** the present study,otolith Sr/Ca was analyzed to investigate the early migratory patterns and discriminate the populations of *** in the Yellow Sea,including two spawning groups and one overwintering *** variation in Sr/Ca ratios of ontogenetic growth zones,including the nucleus(N),larval(L),metamorphosis(M),juvenile(J),and edge(E)zones,was measured by electron probe *** variation in Sr/Ca ratios in early developmental growth zones was generally characterized by an evident downward trend from the N to J zone,which suggests that the early migratory pattern of *** might be from inshore to nearshore *** discriminant analysis,based on the otolith Sr/Ca ratios of the N,L,M,and J zones,allowed the successful discrimination of the two populations,namely,the northern and southern Yellow Sea groups,whose diff erences were mainly reflected in the L and J *** with previous studies,the traditional geographic boundaries(34°N)separating these two populations might be moving *** application of otolith Sr/Ca ratios based on ontogenetic stage could improve our understanding of the migration and population discrimination of *** from the Yellow Sea.

Pilot study to reconstruct life history of Diaphus thiollierei from the Arabian Sea by otolith microstructure and microchemistry

The lanternfishes are mesopelagic fish that are highly productive as common bycatch of deep-sea shrimp trawlers,but they are often neglected or *** being one of the dominant lanternfish species in the Arabian Sea,little is known about the life history of Diaphus thiollierei and its role in marine *** this study,103 *** were collected in the Arabian Sea during October-November 2020 to study population growth based on sagittal otolith daily ages;and 10 fish collected during April–May 2021 were subjected to otolith microchemistry analysis to reconstruct the vertical migration in their life history using LA-ICP-MS *** standard length–dry weight(SL-DW)relationships for *** revealed both negative allometric growth and a significant difference between the *** daily growth annuli counts on the sagittal section of otoliths,the von Bertalanffy growth equation for *** was *** pattern of four elemental ratios(Sr to Ca,Mg to Ca,Li to Ca,and Ba to Ca)in sagittal otolith suggested that,in general,*** descended continually after hatching until the post-larval(PL)stage when they reached a depth of approximately 200 ***,from the PL stage to the post-metamorphosisⅡ(PMⅡ)stage,*** likely further sank from 200 m to a depth of approximately 300 m,and then in the daytime they were at a depth of approximately 300–800 m to take refuge from *** pilot study explored to unravel the vertical migration during life history in *** from sagittal otoliths,whereas further investigation on otolith is needed to better delineate the population ecology in detail,and thus to provide basic information for the exploitation of the lanternfish resource and the understanding of their ecological roles.
