Masculinizer gene controls sexual differentiation in Hyphantria cunea

The Masculinizer gene,Masc,encodes a lepidopteran-specific novel CCCHtype zinc finger protein,which controls sex determination and dosage compensation in Bombyx mori.Considering the potential application of it in pest control,it is necessary to investigate the function of Masc gene in Hyphantria cunea,a globally invasive forest pest.In the present study,we identified and functionally characterized the Masc gene,HcMasc,in H.cunea.Sequence analysis revealed that HcMASC contained the conserved CCCH-type zinc finger domain,nuclear localization signal,and male determining domain,in which the last was confirmed to be required for its masculinization in BmN cell line.However,expression data showed that unlike male-biased expression in B.mori,Hc-Masc gene expresses in main all developmental stages or tissues in both sexes.Clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein 9-based disruption of the common exons 1 and 3 of the HcMasc gene resulted in imbalanced sex ratio and abnormal external genitalia of both sexes.Our results suggest that the HcMasc gene is required for both male and female sexual differentiation and dosage compensation in H.cunea and provide a foundation for developing better strategies to control this pest.

During our investigation of diatom biodiversity in Xizang,two species exhibited unique morphological features discriminative from all previously known genera.Herein we describe these two species and describe as new the genus,Spargeria gen.nov.The new genus features narrow to wide rectangular valves,narrow valve mantles,filiform raphe branches that occur on the valve face only,terminal raphe fissures straight or slightly deflected to same side,bow-tie shaped central areas,chambered striae present on the valve face only,being absent from the mantle,wider striae near the axial area and very narrow near the margin,multiseriate striae with small and round areolae that are occluded externally.Comparatively,Spargeria zhuii sp.***.has larger and robust valves,radiate striae,with one divergent stria near the apices,while Spargeria chenia sp.***.is smaller,with narrow valves,striae slightly radiate in the middle,becoming convergent or parallel near apices.This new genus belongs to the family Pinnulariaceae,and it was compared and contrasted with other genera of this family.Our work suggests the need for continued studies to document the biodiversity of diatoms in Xizang.

Plants of the Caprifoliaceae family are widely cultivated worldwide as ornamental plants owing to their numerous,sweet-smelling,beautiful flowers and fruits.Heptacodium miconioides Rehd.,a member of the family,is endemic to eastern China and is cultivated as a popular ornamental plant in North America and European countries.It has a rather novel and beautiful trait of high horticultural value,that is,its sepals persist and enlarge,turning purplish red.Here,we report the chromosome-level genome assembly of H.miconioides to understand its evolution and floral characteristics.The 622.28 Mb assembled genome harbored a shared whole-genome duplication with a related species,Lonicera japonica.Comparative genomic analysis suggested that chromosome fission events following genome duplication underlie the unusual chromosome number of these two species,as well as chromosome fission of another five chromosomes in H.miconioides,giving rise to a haploid chromosome number of 14(versus 9 in L.japonica).In addition,based on transcriptome and chloroplast genome analysis of 17 representative species in the Caprifoliaceae,we assumed that large structural variations in the chromosomes of H.miconioides were not caused by hybridization.Changes in the candidate genes of the MADS-box family were detected in the H.miconioides genome,including AP1-,AP3-,and SEPexpanded,which might underlie the sepal elongation and development in this species.The current findings provided a critical resource for genome evolution studies in Caprifoliaceae and it was an example of how multi-omics data can elucidate the regulation of important ornamental traits.

Examples of predator–prey interactions in which flies rob ants are uncommon.To date,this behavior has only been recorded in the genus Bengalia Robineau-Desvoidy(Bengaliinae,Diptera,Calliphoridae).These predatory flies ambush ants,and rob them of the food or offspring that they are carrying.However,because of the rarity of this behavior,the reasons and consequences(evolutionary advantages)are unknown,and indeed,the behavior has been sometimes considered anecdotal.In this study,we employed field investigations and behavioral analyses to investigate whether the sex of the fly Bengalia varicolor,or the weight and quality of the food carried by Pheidole nodus ants influenced fly–ant interactions in their natural habitats.We show that food weight and quality influenced the behavior of B.varicolor independent of the fly’s sex.Robbing behavior by the flies was more successful when the food robbed was of high-quality and light in weight.Furthermore,the weight of the food robbed modulated the escape distance the flies could carry it.This then may affect the food quality and weight transported by the ants.This is a novel example of deciphering the relationship between highwayman flies and their ant victims.Given the widespread distribution of Bengalia flies,we suggest that such interspecific predator–prey encounters may shape the robbery interactions and the carrying behavior of further ant species in nature.

Insects play important roles in the maintenance of ecosystem functioning and the provision of livelihoods for millions of people.However,compared with terrestrial vertebrates and angiosperms,such as the giant panda,crested ibis,and the metasequoia,insect conservation has not attracted enough attention,and a basic understanding of the geographical biodiversity patterns for major components of insects in China is lacking.Herein,we investigated the geographical distribution of insect biodiversity across multiple dimensions(taxonomic,genetic,and phylogenetic diversity)based on the spatial distribution and molecular DNA sequencing data of insects.Our analysis included 18 orders,360 families,5,275 genera,and 14,115 species of insects.The results revealed that Southwestern and Southeastern China harbored higher insect biodiversity and numerous older lineages,representing a museum,whereas regions located in Northwestern China harbored lower insect biodiversity and younger lineages,serving as an evolutionary cradle.We also observed that mean annual temperature and precipitation had significantly positive effects,whereas altitude had significantly negative effects on insect biodiversity in most cases.Moreover,cultivated vegetation harbored the highest insect taxonomic and phylogenetic diversity,and needleleaf and broadleaf mixed forests harbored the highest insect genetic diversity.These results indicated that human activities may positively contribute to insect spatial diversity on a regional scale.Our study fills a knowledge gap in insect spatial diversity in China.These findings could help guide national-level conservation plans and the post-2020 biodiversity conservation framework.

Many species exhibit color polymorphisms which have distinct physiological and behavioral characteristics.However,the consistency of morph trait covariation patterns across species,time,and ecological contexts remains unclear.This trait covariation is especially relevant in the context of invasion biology and urban adaptation.Specifically,physiological traits pertaining to energy maintenance are crucial to fitness,given their immediate ties to individual reproduction,growth,and population establishment.We investigated the physiological traits of Podarcis muralis,a versatile color polymorphic species that thrives in urban environments(including invasive populations in Ohio,USA).We measured five physiological traits(plasma corticosterone and triglycerides,hematocrit,body condition,and field body temperature),which compose an integrated multivariate phenotype.We then tested variation among co-occurring color morphs in the context of establishment in an urban environment.We found that the traits describing physiological status and strategy shifted across the active season in a morph-dependent manner—the white and yellow morphs exhibited clearly different multivariate physiological phenotypes,characterized primarily by differences in plasma corticosterone.This suggests that morphs have different strategies in physiological regulation,the flexibility of which is crucial to urban adaptation.The white-yellow morph exhibited an intermediate phenotype,suggesting an intermediary energy maintenance strategy.Orange morphs also exhibited distinct phenotypes,but the low prevalence of this morph in our study populations precludes clear interpretation.Our work provides insight into how differences among stable polymorphisms exist across axes of the phenotype and how this variation may aid in establishment within novel environments.

Based on a combination of morphology and molecular data of ribosomal DNA genes,a new diatom genus Lineaperpetua gen.nov.Yu,You,Kociolek&Wang is described.The features that help define Lineaperpetua at the level of genus include:a tangentially undulated valve face;continuous cribra areolae on the valve interior consisting of pores arranged as strips;single rimoportula located inside the ring of marginal fultoportulae.Additionally,phylogenetic analysis based on nuclear small subunit(SSU)rDNA sequences and nuclear large subunit(LSU)rDNA gene placed the three strains of L.lacustris in a single,monophyletic clade at a considerable sequence distance from the other genera(Thalassiosira,Conticribra,Planktoniella,Shinodiscus,and other genera)belonging to Thalassiosirales.Despite the similarities with some species of Thalassiosira,Conticribra,and Spicaticribra,the suite of features found in Lineaperpetua differentiate it from these other genera.These molecular data and morphological characters suggest an affinity of the new genus to the Thalassiosiraceae.

Role of climatic variability in shaping intraspecific variation of thermal tolerance in Mediterranean water beetles

The climatic variability hypothesis(CVH)predicts that organisms in more thermally variable environments have wider thermal breadths and higher thermal plasticity than those from more stable environments.However,due to evolutionary trade-offs,taxa with greater absolute thermal limits may have little plasticity of such limits(trade-off hypothesis).The CVH assumes that climatic variability is the ultimate driver of thermal tolerance variation across latitudinal and altitudinal gradients,but average temperature also varies along such gradients.We explored intraspecific variation of thermal tolerance in three typical Mediterranean saline water beetles(families Hydrophilidae and Dytiscidae).For each species,we compared two populations where the species coexist,with similar annual mean temperature but contrasting thermal variability(continental vs.coastal population).We estimated thermal limits of adults from each population,previously acclimated at 17,20,or 25℃.We found species-specific patterns but overall,our results agree with the CVH regarding thermal ranges,which were wider in the continental(more variable)population.In the two hydrophilid species,this came at the cost of losing plasticity of the upper thermal limit in this population,supporting the trade-off hypothesis,but not in the dytiscid one.Our results support the role of local adaptation to thermal variability and trade-offs between basal tolerance and physiological plasticity in shaping thermal tolerance in aquatic ectotherms,but also suggest that intraspecific variation of thermal tolerance does not fit a general pattern among aquatic insects.Overlooking such intraspecific variation could lead to inaccurate predictions of the vulnerability of aquatic insects to global warming.

Male competition conforms to a cost-benefit model,because while aggression may increase reproductive prospects,it can also increase the risk of injury.We hypothesize that an additional cost in aggressive males would be an increase in parasite load associated with a high energy investment into sexual competition.Some of these infections,in turn,may downmodulate the level of host aggression via energetic trade-offs.We staged dyadic male contests in the lab to investigate the relationships of multiple parasites with the agonistic behavior of lizard hosts,Sceloporus occidentalis.We also included both color and behavioral traits from opponents in the analyses because(1)color patches of lizards may serve as intraspecific signals used by conspecifics to assess the quality of opponents,and(2)contests between male lizards fit classical models of escalated aggression,where lizards increase aggression displays in response to an opponent's behavior.The results conform to our hypothesis because male lizards displayed more pushups when they had more ticks.Moreover,some parasites may modulate the levels of aggression because lizards infected by hematic coccidians performed fewer pushups.Interestingly,lizards also displayed fewer pushups when both the chroma and size of the opponent's blue patch were greater.The results thus also supported the role of the blue patch of s.occidentalis as a sexual armament,because it contributed to the deterrence of aggression from opponent lizards.We revealed that natural parasitic infections in lizard hosts can contribute to their agonistic behavior.We encourage future studies to account for parasites in behavioral testswithlizards.
