Estimating Changes in Contact Patterns in China Over the First Year of the COVID-19 Pandemic:Implications for SARS-CoV-2 Spread—Four Cities,China,2020

Introduction:Previous studies have demonstrated significant changes in social contacts during the firstwave coronavirus disease 2019(COVID-19)in Chinese mainland.The purpose of this study was to quantify the time-varying contact patterns by age in Chinese mainland in 2020 and evaluate their impact on the transmission of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).Methods:Diary-based contact surveys were performed for four periods:baseline(prior to 2020),outbreak(February 2020),post-lockdown(March–May 2020),and post-epidemic(September–November 2020).We built a Susceptible-Infected-Recovered(SIR)model to evaluate the effect of reducing contacts on transmission.Results:During the post-epidemic period,daily contacts resumed to 26.7%,14.8%,46.8%,and 44.2%of the pre-COVID levels in Wuhan,Shanghai,Shenzhen,and Changsha,respectively.This suggests a moderate risk of resurgence in Changsha,Shenzhen,and Wuhan,and a low risk in Shanghai.School closure alone was not enough to interrupt transmission of SARS-CoV-2 Omicron BA.5,but with the addition of a 75%reduction of contacts at the workplace,it could lead to a 16.8%reduction of the attack rate.To control an outbreak,concerted strategies that target schools,workplaces,and community contacts are needed.Discussion:Monitoring contact patterns by age is key to quantifying the risk of COVID-19 outbreaks and evaluating the impact of intervention strategies.

Monitoring changes in the epidemiologic features between different severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants is key to understanding the evolution of viral fitness in the host population.Here,we analyzed a successfully contained local outbreak of the Delta variant that took place in Hunan,China,in July–August,2021.Detailed data on SARS-CoV-2 infections and their contacts were collected during the outbreak.By leveraging these data,we estimated key epidemiological parameters,including the incubation period,serial interval,and generation time.We constructed a generalized linear mixed-effects model(GLMM)to quantify risk factors for Delta infection and transmission.Between July 28 and August 15,2021,a total of 129 infections and their 2,118 close contacts were identified during the outbreak in Hunan Province.

Systematic analysis on expression quantitative trait loci identifies a novel regulatory variant in ring finger and WD repeat domain 3 associated with prognosis of pancreatic cancer

Background:Pancreatic adenocarcinoma(PAAD)is an extremely lethal malignancy.Identification of the functional genes and genetic variants related to PAAD prognosis is important and challenging.Previously identified prognostic genes from several expression profile analyses were inconsistent.The regulatory genetic variants that affect PAAD prognosis were largely unknown.Methods:Firstly,a meta-analysis was performed with seven published datasets to systematically explore the candidate prognostic genes for PAAD.Next,to identify the regulatory variants for those candidate genes,expression quantitative trait loci analysis was implemented with PAAD data resources from The Cancer Genome Atlas.Then,a two-stage association study in a total of 893 PAAD patients was conducted to interrogate the regulatory variants and find the prognostic locus.Finally,a series of biochemical experiments and phenotype assays were carried out to demonstrate the biological function of variation and genes in PAAD progression process.Results:A total of 128 genes were identified associated with the PAAD prognosis in the meta-analysis.Fourteen regulatory loci in 12 of the 128 genes were discovered,among which,only rs4887783,the functional variant in the promoter of Ring Finger and WD Repeat Domain 3(RFWD3),presented significant association with PAAD prognosis in both stages of the population study.Dualluciferase reporter and electrophoretic mobility shift assays demonstrated that rs4887783-G allele,which predicts the worse prognosis,enhanced the binding of transcript factor REST,thus elevating RFWD3 expression.Further phenotypic assays revealed that excess expression of RFWD3 promoted tumor cell migration without affecting their proliferation rate.RFWD3 was highly expressed in PAAD and might orchestrate the genes in the DNA repair process.Conclusions:RFWD3 and its regulatory variant are novel genetic factors for PAAD prognosis.

Introduction:Minimizing the importation and exportation risks of coronavirus disease 2019(COVID-19)is a primary concern for sustaining the“Dynamic COVID-zero”strategy in China.Risk estimation is essential for cities to conduct before relaxing border control measures.Methods:Informed by the daily number of passengers traveling between 367 prefectures(cities)in China,this study used a stochastic metapopulation model parameterized with COVID-19 epidemic characteristics to estimate the importation and exportation risks.Results:Under the transmission scenario(R0=5.49),this study estimated the cumulative case incidence of Changchun City,Jilin Province as 3,233(95%confidence interval:1,480,4,986)before a lockdown on March 14,2022,which is close to the 3,168 cases reported in real life by March 16,2022.In a total of 367 prefectures(cities),127(35%)had high exportation risks according to the simulation and could transmit the disease to 50%of all other regions within a period from 17 to 94 days.The average time until a new infection arrives in a location in 1 of the 367 prefectures(cities)ranged from 26 to 101 days.Conclusions:Estimating COVID-19 importation and exportation risks is necessary for preparedness,prevention,and control measures of COVID-19—especially when new variants emerge.

Short-term impacts of air pollution on the platelet-lymphocyte ratio and neutrophil-lymphocyte ratio among urban adults in China

The short-term impacts of urban air pollution on the platelet-lymphocyte ratio (PLR) and neutrophil-lymphocyte ratio (NLR) remain obscure.In this study,we included 3487 urban adults from the Wuhan-Zhuhai cohort.Individual inhalation exposure to air pollutants was estimated by combining participants’daily breath volume and ambient concentrations of six air pollutants (includingfine particulate matter (PM_(2.5)),inhalable particulate matter(PM_(10)),nitrogen dioxide (NO_(2)),sulfur dioxide (SO_(2)),carbon monoxide (CO) and ozone (O_(3))).The cumulative impacts were assessed by applying lag structures of up to 7 days before the survey date.Associations of air pollutants with PLR and NLR were assessed using a linear mixed model and Bayesian kernel machine regression (BKMR) model.We found that PLR was negatively related to PM_(2.5)(lag02-lag06),PM_(10)(lag02-lag07),NO_(2)(lag02-lag07),and SO_(2)(lag03-lag05) and NLR was negatively related to PM_(10)(lag05 and lag07).In the BKMR model,a negative joint association between the six-air-pollutant mixture and PLR and NLR was observed,whereas PM_(10)and NO_(2)appeared to be more important than the other pollutants in the mixture.The negative impact of air pollutants was stronger in males,participants with lower body mass index (health impact of air pollutants.

Background The impact of pediatric body mass index(BMI)trajectories on the risk of adolescent hypertension(HTN)determined by three separate visits remains unclear.This longitudinal study aims to identify potential pediatric sex-specific BMI trajectories and to assess their associations with HTN and HTN subtypes.Methods Based on the health Promotion Program for Children and Adolescents(HPPCA)in Suzhou,China,a total of 24,426 participants who had initial normal blood pressure(BP)and had at least four BMI measurements during 2012-2020 were included.HTN was defined as simultaneously having three separate visits of elevated BP in 2020.Latent class growth models were used to explore sex-specific BMI trajectories,whose associations with HTN and HTN subtypes were further examined by logistic regression.Results The incidence of HTN determined through three separate visits was 3.34%.Four trajectories were identified for both sexes:low BMI increasing,medium BMI increasing,high BMI increasing,and highest BMI increasing.Compared to the medium BMI increasing group,the odds ratio(95%confidential interval)for developing adolescent HTN of the low,high,and highest BMI increasing groups among boys were 0.54(0.39,0.75),1.90(1.44,2.51),and 2.89(1.90,4.39),respectively;and the corresponding values for girls were 0.66(0.48,0.90),2.30(1.72,3.09),and 4.71(3.06,7.26).Similar gradually elevated associations between different trajectories with isolated systolic hypertension,systolic and diastolic hypertension were observed.Conclusion Current results emphasized the adverse effects of stable high BMI on HTN and the benefits of maintaining normal weight throughout childhood.
