Backgrounds ATP is the major energy source for myotube contraction,and is quickly produced to compensate ATP consumption and to maintain sufficient ATP *** is consumed mainly in cytoplasm and produced in mitochondria during myotube *** understand the mechanism of ATP homeostasis during myotube contraction,it is essential to monitor mitochondrial ATP at single-cell level,and examine how ATP is produced and consumed in ***:We established C2C12 cell line stably expressing fluorescent probe of mitochondrial ATP,and induced differentiation into myotubes・We gave electric pulse stimulation to the differentiated myotubes,and measured mitochondrial *** constructed mathematical model of mitochondrial ATP at single-cell level,and analyzed kinetic parameters of ATP production and ***:We performed hierarchical clustering analysis of time course of mitochondrial ATP,which resulted in two *** 1 showed strong transient increase,whereas cluster 2 showed weak transient *** modeling at single-cell level revealed that the ATP production rate of cluster 1 was larger than that of cluster 2,and that both regulatory pathways of ATP production and consumption of cluster 1 were faster than those of cluster *** 1 showed larger mitochondrial mass than cluster 2,suggesting that cluster 1 shows the similar property of slow muscle fibers,and cluster 2 shows the similar property of fast muscle *** Cluster 1 showed the stronger mitochondrial ATP increase by larger ATP production rate,but not smaller *** 1 might reflect the larger oxidative capacity of slow muscle fiber.

The bipartite begomoviruses (Geminiviridae family), which are DNA viruses that replicate in the nucleus of infected cells, encode the nuclear shuttle protein (NSP) to facilitate the translocation of viral DNA from the nucleus to the cytoplasm via nuclear pores. This intracellular trafficking of NSP-DNA complexes is accessorized by the NSP-interacting guanosine triphosphatase (NIG) at the cytosolic side. Here, we report the nuclear redistribution of NIG by AtWWPI, a WW domain-containing protein that forms immune nuclear bodies (NBs) against begomoviruses. We demonstrated that AtWWPI relocates NIG from the cytoplasm to the nucleus where it is confined to AtWWP1-NBs, suggesting that the NIG-AtWWP1 interaction may interfere with the NIG pro-viral function associated with its cytosolic localization. Consistent with this assumpti on joss AtWWPI functi on cuased plants more susceptible to begomovirus infection, whereas overexpression of AWWP1 enhaneed plant resistance to begomovirus. Furthermore, we found that a mutant versi on of AtWWPI defective for NB formatio n was noIonger capable of interacti ng with and relocating NIG to the nucleus and lost its immune function against begomovirus. The antiviral function of AtWWP1-NBs, however, could be antagonized by viral infection that induced either the disruption or a decrease in the number of AtWWP1-NBs. Collectively, these results led us to propose that AtWWPI organizes nuclear structures into nuclear foci, which provide intrinsic immunity against begomovirus infection.

Flavones are among the major colorless pigments synthesized through branches of the flavonoid pathway in ***,due to the absence of a gene encoding flavone synthase(FNS)in the model plant Arabidopsis thaliana species,the regulatory mechanism of FNS-catalyzed flavone biosynthesis has rarely been studied in ***,it was found that flavones play a predominant role in the elimination of excess reactive oxygen species(ROS)at high temperatures in colorless plant organs.A novel atypical subgroup 7(SG7)R2R3-MYB transcription factor,CmMYB012,was found to be induced in response to prolonged high temperatures and to inhibit flavone biosynthesis by directly regulating ***,CmMYB012 was also found to inhibit anthocyanin biosynthesis by suppressing the expression of CmCHS,CmDFR,CmANS,and ***012 overexpression exerted a negative in fluence on plant fitness and pink flower color formation,while CmMYB012 suppression had the opposite effect in response to high *** findings provide new insights into the mechanisms by which high temperatures regulate the metabolism of flavones and anthocyanins to affect plant fitness and flower color formation.

Green fl uorescent protein(GFP)has been widely used for monitoring gene expression and protein localization in diverse ***,highly sensitive imaging equipment,like fl uorescence microscope,is usually required for the visualization of GFP,limitings its application to fi xed locations in samples.A reporter that can be visualized in realtime regardless the shape,size and location of the target samples will increase the fl exibility and ef fi ciency of research ***,we report the application of a GFP-like protein,called eYGFPuv,in both transient expression and stable transformation,in two herbaceous plant species(Arabidopsis and tobacco)and two woody plant species(poplar and citrus).We observed bright fl uorescence under UV light in all of the four plant species without any effects on plant growth or *** was shown to be effective for imaging transient expression in leaf and root *** a focus on in vitro transformation,we demonstrated that the transgenic events expressing 1x eYGFPuv could be easily identi fi ed visually during the callus stage and the shoot stage,enabling early and ef fi cient selection of ***,whole-plant level visualization of eYGFPuv revealed its ubiquitous stability in transgenic *** addition,our transformation experiments showed that eYGFPuv can also be used to select transgenic plants without *** work demonstrates the feasibility of utilizing 1x eYGFPuv in studies of gene expression and plant transformation in diverse plants.

自2010年4月新一轮的房地产市场宏观调控以来,我国房地产市场发生了巨大的变化。房价打破了单边上涨的规律,进入下降通道,国家对房地产业的政策也由支柱产业变成了民生产业。房企在新一轮调控中饱受各种危机,其中对房企影响最大的莫过于投资决策错误。导致投资决策失败有多方面的原因,包括投资阶段中市场、售价、去化能力定位失误,投资决策过程中财务分析不准确等。本文以杭州BY项目为例,从当前房地产市场宏观调控及市场特点分析,找出BY项目当前投资的合理市场、价格、去化能力,通过建立模型计算BY项目财务评价主要指标。
