Inflammation is a primary defense process against various extracellular stimuli,such as viruses,pathogens,foods,and environmental *** cells respond to stimuli for short periods of time,it results in acute or physiological ***,if the stimulation is sustained for longer time or a pathological state occurs,it is known as chronic or pathological *** studies have shown that tumorigenesis in the gastrointestinal (GI) tract is closely associated with chronic inflammation,for which abnormal cellular alterations that accompany chronic inflammation such as oxidative stresses,gene mutations,epigenetic changes,and inflammatory cytokines,are shared with carcinogenic processes,which forms a critical cross-link between chronic inflammation and *** growth factor (TGF)-β is a multi-potent cytokine that plays an important role in regulation of cell growth,apoptosis and *** importantly,TGF-β is a strong anti-inflammatory cytokine that regulates the development of effector ***-β has a suppressive effect on carcinogenesis under normal conditions by inhibiting abnormal cell growth,but on the other hand,many GI cancers originate from uncontrolled cell growth and differentiation by genetic loss of TGF-β signaling molecules or perturbation of TGF-β *** a tumor has developed,TGF-β exerts a promoting effect on the tumor itself and stromal cells to enhance cell growth,alter the responsiveness of tumor cells to stimulate invasion and metastasis,and inhibited immune ***,novel development of therapeutic agents to inhibit TGF-β-induced progression of tumor and to retain its growth inhibitory activities,in addition to anti-inflammatory actions,could be useful in *** this review,we discuss the role of TGF-β in inflammation and carcinogenesis of the GI tract related to abnormal TGF-β signaling.

Terpenoids as potential chemopreventive and therapeutic agents in liver cancer

Despite significant advances in medicine, liver cancer, predominantly hepatocellular carcinoma remains a major cause of death in the United States as well as the rest of the world. As limited treatment options are currently available to patients with liver cancer, novel preventive control and effective therapeutic approaches are considered to be reasonable and decisive measures to combat this disease. Several naturally occurring dietary and non-dietary phytochemicals have shown enormous potential in the prevention and treatment of several cancers, especially those of the gastrointestinal tract. Terpenoids, the largest group of phytochemicals, traditionally used for medicinal purposes in India and China, are currently being explored as anticancer agents in clinical trials. Terpenoids (also called "isoprenoids") are secondary metabolites occurring in most organisms, particularly plants. More than 40 000 individual terpenoids are known to exist in nature with new compounds being discovered every year. A large number of terpenoids exhibit cytotoxicity against a variety of tumor cells and cancer preventive as well as anticancer efficacy in preclinical animal models. This review critically examines the potential role of naturally occurring terpenoids, from diverse origins, in the chemoprevention and treatment of liver tumors. Both in vitro and in vivo effects of these agents and related cellular and molecular mechanisms are highlighted. Potential challenges and future directions involved in the advancement of these promising natural compounds in the chemoprevention and therapy of human liver cancer are also discussed.

Objective:To determine the effect of rice bran extract(RBE)in combination with doxorubicin on 4 T1 triple-negative breast cancer cells and NIH-3 T3 ***:RBE was obtained by maceration with *** phytochemical profile of RBE was observed using highperformance liquid *** effect of RBE was evaluated through MTT *** addition,flow cytometry was used for cell cycle and apoptosis *** senescence was observed using SA-β-Gal assay and intracellular reactive oxygen species(ROS)levels were evaluated using DCFDA *** pro-oxidant property of RBE was also evaluated through 1-chloro-2,4-dinitrobenzene spectrophotometry and molecular ***:RBE was obtained with a yield of 18.42%w/w and contained tocotrienols as the major *** exerted no cytotoxic effect on 4 T1 and NIH-3 T3 ***,RBE in combination with doxorubicin decreased 4 T1 cell viability synergistically(combination index<0.9)and induced apoptosis and senescence on 4 T1 *** significantly decreased senescence in doxorubicin-treated NIH-3 T3 ***,RBE did not increase ROS levels in doxorubicin-treated 4 T1 ***,the combination of RBE and doxorubicin reduced ROS levels in NIH-3 T3 *** significantly reduced glutathione-S-transferase activity and alpha-tocotrienol interacted with glutathione-Stransferase in the glutathione binding ***:Rice bran may be used as a co-chemotherapeutic agent to improve the therapeutic effectiveness of doxorubicin while protecting against the cellular senescence effects of doxorubicin on healthy cells.

Administration of MCE could not inhibit hepatic carcinogenesis in DMBA-induced female *** was no difference in liver tissue histopathology profile between the extract treatment group and DMBA control group.

Combination of low-concentration of novel phytoestrogen(8,9)-furanyl-pterocarpan-3-ol from Pachyrhizus erosus attenuated tamoxifen-associated growth inhibition on breast cancer T47D cells

Objective:To investigate the estrogenic effect of(8,9)-furanyl-pterocarpan-3-ol(FPC)on growth of human breast cancer T47D cells and the interactions between the FPC and tamoxifen(TAM),on the growth of estrogen receptor-dependent breast cancer T47D ***:The proliferation effect of FPC were conducted on T47D cells in vitro by MTT test.T47D cells were treated with FPC alone(0.01-200μmol/L)or in combination with TAM 20 nmol/***,the expression of ERαor c-Myc were also determined by ***:The results indicated that administration of an anti-estrogen TAM showed growth inhibitory effect on T47D cells,wheraes co-administered with low concentration(less than 1μmol/L)of FPC attenuated to promote cell *** contrast,the combination of TAM with higher doses(more than 20μmol/L)of FPC showed growth *** result was supported by immunocytochemistry studies that the administration of 20 nmol/L TAM down-regulated ER-αand c-Myc,but the combination of 20 nmol/L TAM and 1μmol/L FPC robustly up-regulated expression of ER-α.Thus,the reduced growth inhibition of TAM 20 nmol/L by FPC 1μmol/L on T47D cells may act via the modulation of ER-α.Conclusions:The findings indicate and suggest that FPC had estrogenic activity at low concentrations and anti-estrogenic effect that are likely to be regulated by c-Myc and estrogen *** also confirm that low concentration of FPC attenuated the growth-inhibitory effects of TAM on mammary tumor ***,the present study suggests that caution is warranted regarding the consumption of dietary FPC by breast cancer patients while on TMA therapy.

DNA branched junctions with a unique molecular configuration existed naturally and function in specific biological processes.[1-3] The unique branched structures have been built in some works [4-6], exhibiting the slower electrophoretic mobility due to their extended arms [7-8]. In a confined environment such as cells, DNA branched junctions were conjectured to process the similar physical property. Herein we designed a target-induced catalytic assembly of DNA branched junctions and its application for in situ imaging of microRNAs. The precursors were delivered into cells, followed by in situ growth of DNA branched junctions. Their extended arms significantly impact their freedom of movement in cells, contributing to the in situ imaging of targets. Compare to the traditional fluorescence in situ hybridization（FISH） method, the proposed imaging technique is more sensitive. To our knowledge, DNA branched junctions were for the first time exploited for chemical biology applications, leading to an impressive method for in situ imaging of biomolecules within cells.

There are many biohazards existing in the blood that are potential health threats but cannot be *** create a novel avenue that uses an intelligently-designed safe biomaterial to precisely identify,tow and dump the endogenous biohazard into small *** we show that the positively-charged mesoporous silica nanoparticles（MSN） conjugated with EGFR-targeting aptamer（MSN-AP） can specifically bind blood negatively-charged oncogenic exosomes（A-Exo）,and tow A-Exo to pass hepatobiliary layers and the gate Oddi’s sphincter into small *** MSN-AP could specifically distinguish the oncogenic exosomes from the control exosomes in cell media and rat blood,or from blood of lung cancer *** MSN-AP bound A-Exo more tightly than the negatively-charged *** conjugate traversed hepatocyte,conlangiocyte,and endothelial monolayers but Kupffer cells engulfed some of *** MSN-AP significantly decreased circulating A-Exo and sequentially increased intestinal A-Exo,and attenuated A-Exo-induced lung metastasis in *** study exemplifies an innovative model to eliminate various life-threatening biohazards in blood.

Dear Editor,Metastasis is the cause of most fatalities in cancer patients and remains the phenomenon poorly understood mechanistically. Deciphering of the regulatory networks underlying the cancer cell metastasis is urgently needed. Nucleolar protein 7 (NOL7) has been reported to function as a tumor suppressor in cervical cancer.1 Our current study reveals a novel tumor-promoting capacity of NOL7 in melanoma. We first detected that NOL7 expression is upregulated in metastatic melanoma as compared with its expression at the primary site through isobaric tag for relative and absolute quantitation proteomic screening, further confirming this finding with the analysis of NOL7 protein and messenger RNA (mRNA) levels (Supplementary Fig. S1). Importantly, NOL7 expression increased with the disease progression from benign nevus to primary melanoma and further to metastatic melanoma (Fig. 1a and Supplementary Fig. S1d). Previous studies have shown that melanoma is commonly associated with the amplification of the chromosome region 6p, particularly 6p21–23, where the NOL7 gene resides, and that this region frequently undergoes heterozygous loss in cervical cancer.2 It might therefore be predicted that NOL7 exhibits a different expression pattern and plays different roles in melanoma and cervical cancer.

Preparation and Characterization of Hydroxyapatite-Coated Selenium Nanoparticles and their Interaction with Osteosarcoma(SaOS-2) Cells

Nanotechnology has taken a firm step to revolutionize the field of orthopedic implants. Current research on bone implants focuses to develop implants with multifaceted functions viz., osteoinduction, chemoprevention, antimicrobial action etc., especially for cancerous bone resection. The objective of the present study was to synthesize a novel composite for bone implants, possessing the above properties. Selenium was selected owing to its chemopreventive and chemotherapeutic properties. Hydroxyapatite was selected owing to its bioactivity and similarity in composition to bone mineral properties. Selenium nanoparticles were prepared by chemical reduction method and coated with ***-coated selenium nanoparticle（HASnp） was characterized physico-chemically using fourier transform infrared spectroscopy, X-ray diffractometry, scanning electron microscope, and energy-dispersive X-ray *** was analysed in vitro using SaOS-2 cell line. Enhanced cell proliferation and alkaline phosphatase activity were observed in HASnp-treated cells. The results indicate that HASnp is highly suitable for the use in orthopedic applications.

The current role of neoadjuvant/adjuvant/chemoprevention therapy in partial hepatectomy for hepatocellular carcinoma:a systematic review

BACKGROUND: Following curative treatment for hepatocellular carcinoma (HCC), 50%-90% of postoperative death is due to recurrent disease. Intra-hepatic recurrence is frequently the only site of recurrence. Thus, any neoadjuvant or adjuvant therapy, which can decrease or delay the incidence of intra-hepatic recurrence, or any cancer chemoprevention which can prevent a new HCC from developing in the liver remnant, will improve the results of liver resection. This article systematically reviewed the current evidence of neoadjuvant, adjuvant, and chemoprevention in partial hepatectomy of HCC. DATA SOURCES: Studies were identified by searching MEDLINE and PubMed databases for articles from January 1990 to November 2008 using the keywords 'hepatocellular carcinoma', 'hepatectomy', 'adjuvant therapy', 'neoadjuvant therapy', and 'regional therapy'. Additional papers and book chapters were identified by a manual search of the references from the key articles. RESULTS: Neoadjuvant transarterial chemoembolization or adjuvant regional transarterial chemotherapy embolization+systemic chemotherapy did not add benefit. Both adjuvant transarterial radioembolization with (131)I-lipiodol and adjuvant systemic interferon showed promising results. However, there were only a limited number of such studies. CONCLUSIONS: Further randomized controlled studies need to be carried out. Currently, there is no consensus on a standard neoadjuvant/adjuvant/chemoprevention therapy in partial hepatectomy for HCC.
