Here we review the literature on the effects of molecular hydrogen (H2) on normal human subjects and patients with a variety of diagnoses, such as metabolic, rheumatic, cardiovascular and neurodegenerative and other diseases, infections and physical and radiation damage as well as effects on aging and exercise. Although the effects of H2 have been studied in multiple animal models of human disease, such studies will not be reviewed in depth here. H2 can be administered as a gas, in saline implants or infusions, as topical solutions or baths or by drinking H2-enriched water. This latter method is the easiest and least costly method of administration. There are no safety issues with hydrogen;it has been used for years in gas mixtures for deep diving and in numerous clinical trials without adverse events, and there are no warnings in the literature of its toxicity or long-term exposure effects. molecular hydrogen has proven useful and convenient as a novel antioxidant and modifier of gene expression in many conditions where oxidative stress and changes in gene expression result in cellular damage.

目的:探讨间歇运动对大鼠骨骼肌细胞增殖和白血病抑制因子(Leukemia inhibitory factor,LIF)及其下游信号分子表征的影响。方法:3月龄雄性SD大鼠,随机分为安静对照组(C)、间歇运动组(CE),每组15只。运动组采用小动物跑台运动,运动方式为先以10 m/min×10 min,再以25 m/min×7 min和15m/min×3 min,依次交替,60 min/d,5 d/周×8周。运动结束后次日,分离切取大鼠腓肠肌组织,称重。组织学方法测定腓肠肌细胞横截面积(cross-section area,CSA);Western blot检测腓肠肌LIF及其受体(Leukemia inhibitory factor receptor,LIFR)、信号转导与转录激活因子(signal transducer and activator of transcription,STAT3)及其磷酸化水平(p-STAT3)和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)蛋白表达;免疫荧光化学观察分析腓肠肌LIF、LIFR、PCNA和细胞增殖抗原标记物(ki67)表达;RT-q PCR检测腓肠肌LIF/LIFR mRNA水平。结果:与C组比较,CE组大鼠腓肠肌重量和细胞CSA增大(P<0.01),LIF/LIFR基因和蛋白表达均显著增加(P<0.01,P<0.05),STAT3磷酸化水平显著升高(P<0.01),PCNA和ki67蛋白表达显著增加(P<0.01)。结论:8周间歇运动可促进正常大鼠骨骼肌细胞肥大,激活LIF-LIFR-STAT3信号通路,诱导骨骼肌细胞增殖。

Metabolic regulation of adult stem cell-derived neurons

The discovery of continuous generation of functional neurons throughout fife has emerged as a major contributor to plasticity in defined regions of the adult mammalian brain. Work over the past decades identified cellular constituents of the distinct adult neurogenic niches as well as numerous signaling pathways, transcriptional and epigenetic regulators that exert tight control over the production of new neurons from resident stem cells. Recent studies uncovered developmental stage-specific adaptations of metabolic circuits and have provided evidence for their central regulatory function in the adult neurogenic lineage. Moreover, there is increasing evidence for a regulatory impact of a wide range of systemic metabolic factors including exercise induced metabolic changes and diet on the development of adult-born neurons. Here, we will summarize current knowledge and emerging principles underlying the metabolic control of neuronal maturation in adult neurogenesis.

Highly sensitive troponin I assay in the diagnosis of coronary artery disease in patients with suspected stable angina

Purpose: The purpose of the present controlled cross-sectional study was to investigate proximal femur and whole-body bone mineral density(BMD), as well as bone turnover profile, in lifelong trained elderly male football players and young elite football players compared with untrained age-matched ***: One hundred and forty healthy, non-smoking men participated in the study, including lifelong trained football players(FTE, n = 35)aged 65—80 years, elite football players(FTY, n = 35) aged 18—30 years, as well as untrained age-matched elderly(UE, n = 35) and young(UY,n = 35) men. All participants underwent a regional dual-energy X-ray Absorptiometry(DXA) scan of the proximal femur and a whole-body DXA scan to determine BMD. From a resting blood sample, the bone turnover markers(BTMs) osteocalcin, carboxy-terminal type-1 collagen crosslinks(CTX-1), procollagen type-1 amino-terminal propeptide(P1NP), and sclerostin were ***: FTE had 7.3%—12.9% higher(p < 0.05) BMD of the femoral neck, wards, shaft, and total proximal femur in both legs compared to UE,and 9.3%—9.7% higher(p < 0.05) BMD in femoral trochanter in both legs compared to UY. FTY had 24.3%—37.4% higher(p < 0.001) BMD in all femoral regions and total proximal femur in both legs compared to UY. The whole-body DXA scan confirmed these results, with FTE showing similar whole-body BMD and 7.9% higher(p < 0.05) leg BMD compared to UY, and with FTY having 9.6% higher(p < 0.001) wholebody BMD and 18.2% higher(p < 0.001) leg BMD compared to UY. AIM:To investigate the impact of MK-801 on gene expression patterns genome wide in rat brain regions. METHODS:Rats were treated with an intraperitoneal injection of MK-801 [0.08(low-dose) and 0.16(highdose) mg/kg] or NaC l(vehicle control). In a first series of experiment,the frontoparietal electrocorticogram was recorded 15 min before and 60 min after injection. In a second series of experiments,the whole brain of each animal was rapidly removed at 40 min post-injection,and different regions were separated:amygdala,cerebral cortex,hippocampus,hypothalamus,midbrain and ventral striatum on ice followed by DNA microarray(4 × 44 K whole rat genome chip) ***:Spectral analysis revealed that a single systemic injection of MK-801 significantly and selectively augmented the power of baseline gamma frequency(30-80 Hz) oscillations in the frontoparietal electroencephalogram. DNA microarray analysis showed the largest number(up- and down- regulations) of gene expressions in the cerebral cortex(378),midbrain(376),hippocampus(375),ventral striatum(353),amygdala(301),and hypothalamus(201) under low-dose(0.08 mg/kg) of MK-801. Under high-dose(0.16 mg/kg),ventral striatum(811) showed the largest number of gene expression changes. Gene expression changes were functionally categorized to reveal expression of genes and function varies with each brain ***:Acute MK-801 treatment increases synchrony of baseline gamma oscillations,and causes very early changes in gene expressions in six individual rat brain regions,a first report.

The Physiological Effects of a Walking to Music Intervention in Adults with Intermediate Hyperglycemia

Purpose: Overweight individuals are at risk of intermediate hyperglycemia (IHG) [1]. Increasing physical activity (PA) in IHG is one method to reduce the risk of glucometabolic and cardiovascular (CV) complications [2]. This study investigates walking to music as a modality to increase moderate intensity PA and regulate glucometabolic disturbances. Patients and Methods: Participants were randomized to usual care (UC) or intervention group (IG) who completed a 6 month walking to music program. Physiological assessments for a range of variables (DEXA, flow mediated dilatation (FMD) and glucometabolic biomarkers) were completed at baseline, 4, 6 and 9 months (follow-up). Results: For IG group, walking compliance decreased with time however 71.4%, 79.5% and 73% of walking completed was moderate intensity at 4, 6 and 9 months. At 6 months IG FMD was significantly lower than UC and HbA1Cwas 5% lower. Other physiological markers were not altered. Conclusion: Walking to music may be a novel method to encourage moderate intensity PA. However, the majority of results demonstrate that this intervention was not more effective than UC in managing glucometabolic and CV biomarkers in IHG. Future interventions should include additional support for the entire study duration however this has cost implications.
