Evaluation of water management on arsenic methylation and volatilization in arsenic-contaminated soils strengthened by bioaugmentation and biostimulation

Arsenic(As) fate in paddy fields has been one of the most significant current issues due to the strong As accumulation potential of rice plants under fooded conditions. However,no attempt was done to explore As methylation and volatilization under non-fooded conditions. Herein, we investigated the effects of water management on As methylation and volatilization in three arsenic-contaminated soils enhanced by biostimulation with strawderived organic matter and bioaugmentation with genetic engineered Pseudomonas putida KT2440(GE P. putida). Under fooded conditions, the application of biochar(BC), rice straw(RS)and their combination(BC+RS) increased total As in porewater. However, these effects were greatly attenuated under non-fooded conditions. Compared with RS amendment alone, the combination of GE P. putida and RS further promoted the As methylation and volatilization,and the promotion percentage under non-fooded conditions were significantly higher than that under fooded conditions. The combined GE P. putida and RS showed the highest efficiency in As methylation(88 μg/L) and volatilization(415.4 μg/(kg·year)) in the non-fooded soil with moderate As contamination. Finally, stepwise multiple linear regression analysis presented that methylated As, DOC and p H in porewater were the most important factors contributing to As volatilization. Overall, our findings suggest that combination of bioaugmentation with GE P. putida and biostimulation with RS/BC+RS is a potential strategy for bioremediation of arsenic-contaminated soils by enhancing As methylation and volatilization under non-fooded conditions.

Animal body size variation is of particular interest in evolutionary biology,but the genetic basis remains largely unknown.Previous studies have shown the presence of two parallel evolutionary genetic clusters within the fish genus Epinephelus with evident divergence in body size,providing an excellent opportunity to investigate the genetic basis of body size variation in vertebrates.Herein,we performed phylotranscriptomic analysis and reconstructed the phylogeny of 13 epinephelids originating from the South China Sea.Two genetic clades with an estimated divergence time of approximately 15.4 million years ago were correlated with large and small body size,respectively.A total of 180 rapidly evolving genes and two positively selected genes were identified between the two groups.Functional enrichment analyses of these candidate genes revealed distinct enrichment categories between the two groups.These pathways and genes may play important roles in body size variation in groupers through complex regulatory networks.Based on our results,we speculate that the ancestors of the two divergent groups of groupers may have adapted to different environments through habitat selection,leading to genetic variations in metabolic patterns,organ development,and lifespan,resulting in body size divergence between the two locally adapted populations.These findings provide important insights into the genetic mechanisms underlying body size variation in groupers and species differentiation.

Single-cell transcriptomic landscape of peripheral blood cells provides insights into adaptation of red-eared sliders(Trachemys scripta elegans)

Red-eared sliders(Trachemys scripta elegans),as one of the 100 most threatening aliens,have stronger immunity than the native species in response to environmental stress.Blood cells are an important component of immunity in the body.However,the blood cell researches of turtle are still in the traditional blood cell classification and morphological structure observation.Furthermore,turtle granulocytes cannot be accurately identified using traditional methods.Single-cell RNA sequencing techniques have been successfully implemented to study cells based on the mRNA expression patterns of each cell.The present study profiled the transcriptomes of peripheral blood cells in red-eared sliders to construct a single-cell transcriptional landscape of the different cell types and explored environmental adaptation mechanism from the perspective of hematology.All 14 transcriptionally distinct clusters(platelets,erythrocytes1,erythrocytes2,CSF1R monocytes,POF1B monocytes,neutrophils,GATA2high basophils,GATA2low basophils,CD4 T cells,CD7 T cells,B cells,ACKR4 cells,serotriflin cells,and ficolin cells)were identified in the peripheral blood cells of the red-eared sliders.In particular,a subtype of erythrocytes(erythrocytes1)that expressed immune signals was identified.Peripheral blood cells were grouped into three lineages:platelet,erythroid/lymphoid,and myeloid cell lineages.Furthermore,based on differentiation trajectory and up-regulated gene expression,ACKR4 cells were newly identified as lymphocytes,and serotriflin and ficolin cells as granulocytes.The single-cell transcriptional atlas of the peripheral blood cells in red-eared sliders provided in the present study will offer a comprehensive transcriptome reference for the exploration of physiological and pathological hematology in this species.

Fermented calcium butyrate supplementation in post-peak laying hens improved ovarian function and tibia quality through the“gut-bone” axis

The compromised egg quality and leg abnormality during the end of the laying cycle(after 40 weeks)have been leading to poor animal welfare and substantial economic losses. Therefore, the effects of fermented calcium(Ca) butyrate, produced by fermentation by Clostridium butyricum, on production,eggshell quality, and tibial property of hens were explored. A total of 192 Hy-line brown laying hens at50-week-old were assigned to a basal diet or the basal diet with 300 mg/kg of the fermented Ca butyrate from 50 to 58 weeks of age. Each treatment had 6 replicates with 16 hens each. The diet supplemented with 300 mg/kg fermented Ca butyrate notably increased egg weight, ovarian follicle number, and eggshell strength(P = 0.072) as compared to the basal diet, which were associated with cytokine secretion, toll-like receptor signaling pathways, and intestinal immunity based on the RNA-seq data from the granulosa. Dietary Ca butyrate inclusion decreased the expression of ileal tumor necrosis factor-alpha and serum pro-inflammatory cytokine concentration, as well as increased the content of serum immunoglobulin A when compared to the basal diet(both P 0.05). In addition, the diet with fermented Ca butyrate reduced the number of osteoclasts in the proximal tibia and the level of C-terminal cross-linked telopeptide of type I collagen, a bone resorption marker(P < 0.05), whereas it tended to increase the concentration of the procollagen type I N-terminal propeptide that reflects bone formation marker in serum. Moreover, the layers fed fermented Ca butyrate diets possessed higher(P < 0.05) bone area and trabecular number of the proximal tibia, yield load, and ultimate load than those that consumed basal diets. Species’ geographical range, environmental range and traits lead to specimen collection preference of dominant plant species of grasslands in Northern China

Many different factors,such as species traits,socio-economic factors,geographical and environmental factors,can lead to specimen collection preference.This study aims to determine whether grassland specimen collection in China is preferred by species traits(i.e.,plant height,flowering and fruiting period),environmental range(i.e.,the temperature and precipitation range)and geographical range(i.e.,distribution range and altitudinal range).Ordinary least squares models and phylogenetic generalized linear mixed models were used to analyze the relationships between specimen number and the explanatory variables.Random Forest models were then used to find the most parsimonious multivariate model.The results showed that interannual variation in specimen number between 1900 and 2020 was considerable.Specimen number of these species in southeast China was notably lower than that in northwest China.Environmental range and geographical range of species had significant positive correlations with specimen number.In addition,there were relatively weak but significant associations between specimen number and species trait(i.e.,plant height and flowering and fruiting period).Random Forest models indicated that distribution range was the most important variable,followed by flowering and fruiting period,and altitudinal range.These findings suggest that future floristic surveys should pay more attention to species with small geographical range,narrow environmental range,short plant height,and short flowering and fruiting period.The correction of specimen collection preference will also make the results of species distribution model,species evolution and other works based on specimen data more accurate.

Nest decoration:Black feathers prevent Crested Mynas from nest usurpation

Nests are important structures for birds to raise their offspring and for signaling.Many birds incorporate feathers into the nest,since feathers were traditionally thought to serve the function of insulation.Hypotheses in recent years have considered that some birds place feathers in conspicuous locations in the nest for decoration to trigger a fear response in the competitors.In this study,we investigated whether decorative feathers could deter nest usurpation by Crested Mynas(Acridotheres cristatellus)by manipulating nest box contents.The results revealed that Crested Mynas preferred black feathers to white feathers as decorations and occupied nest boxes decorated with black feathers significantly less than those decorated with white feathers,suggesting that black decorative feathers in the nest could be more effective in preventing nest usurpation by Crested Mynas and that white decorative feathers may have other functions.The black feathers in the nest are prominently placed at the edge of the nest to convey the message that“this nest is occupied”or“the owner of this nest has been preyed upon”to visitor Crested Mynas,thus effectively preventing them from usurping the nest at a later stage.

Total-factor energy efficiency of ten major global energy-consuming countries

Improving energy efficiency is regarded as a key path to tackling global warming and achieving the Sustainable Development Goals(SDGs).In 2020,the energy consumption of the world's ten major energy-consuming countries accounted for 66.8%of the global total.This paper applied data envelopment analysis(DEA)to calculate these ten major energyconsuming countries'total-factor energy efficiency(TFEE)at national and sectoral levels from 2001-2020,and explored the infuencing factors of total-factor energy efficiency with the Tobit regression model.The results showed significant difference in the ten countries'energy efficiency.The United States and Germany topped the list for total-factor energy efficiency,while China and India were at the bottom.Meanwhile,the energy efficiency of the industrial subsector has increased significantly over the past two decades,while that of the other subsectors has been relatively fat.The industrial structure upgrading,per capita GDP,energy consumption structure,and foreign direct investment had significant impacts on energy efficiency with national heterogeneity.Energy consumption structure and GDP per capita were determinative factors of energy efficiency.

Field-based evidence for the enrichment of intrinsic antibiotic resistome stimulated by plant-derived fertilizer in agricultural soil

Animal manures have been demonstrated to enhance antibiotic resistance in agricultural soils.However,little is known about the effects of plant-derived fertilizer on soil antibiotic resistome.Herein,metagenomic sequencing was used to investigate the effects of a plantderived fertilizer processed from sugarcane and beet on soil antibiotic resistance genes(ARGs)in a soybean field along crop growth stages.ARG profiles in the soils amended by plant-derived fertilizer were compared with those in the soils amended by chicken manure.The abundance and diversity of total ARGs in the soils amended by plant-derived fertilizer were significantly(P<0.05)elevated at the sprout stage,to a level comparable to that in the manured soils.Whereas,unlike chicken manure mainly introducing manure-borne ARGs to soil,the plant-derived fertilizer was indicated to mainly enrich multidrug resistance genes in soil by nourishing indigenous bacteria.ARGs with abundances in amended soils significantly(P<0.05)higher than in unamended soils at the sprout stage of soybean were considered as enriched ARGs.Decrease in the abundance of the enriched ARGs was observed in both the amended soils from the sprout to the harvest.Network analysis further identified Proteobacteria and Bacteroidetes as the primary bacterial taxa involved in the temporal variation of the enriched ARGs in the soils amended by plant-derived fertilizer,while in manured soils were Firmicutes and Actinobacteria.As revealed by multivariate statistical analyses,variation of the enriched ARGs in the soils amended by plant-derived fertilizer was majorly attributed to the response of co-occurred bacteria to depleting nutrients,which was different from the failed establishment of manure-borne bacteria in the manured soils.Our study provided field-based evidence that plant-derived fertilizer stimulated the intrinsic antibiotic resistome,and proposed attention to the un-perceived risk since some clinically relevant ARGs originate and evolve from natural resistome.

More tree growth reduction due to consecutive drought and its legacy effect for a semiarid larch plantation in Northwest China

Extreme climate has increasingly led to negative impacts on forest ecosystems globally,especially in semiarid areas where forest ecosystems are more vulnerable.However,it is poorly understood how tree growth is affected by different drought events.In 2006–2009,the larch plantations in the semiarid areas of Northwest China were negatively affected by four consecutive dry years,which was a very rare phenomenon that may occur frequently under future climate warming.In this study,we analyzed the effect of these consecutive dry years on tree growth based on the data of the tree rings in the dominant layer of the forest canopy on a larch plantation.We found that the tree-ring width index(RWI)in dry years was lower than that in normal years,and it experienced a rapidly decreasing trend from 2006 to 2009(slope=-0.139 year^(-1),r=-0.94)due to water supply deficits in those dry years.Drought induced legacy effects of tree growth reduction,and consecutive dry years corresponded with greater growth reductions and legacy effects.Growth reductions and legacy effects were significantly stronger in the third and fourth consecutive dry years than that of single dry year(p<0.05),which might have been due to the cumulative stress caused by consecutive dry years.Our results showed that larch trees experienced greater tree growth reduction due to consecutive dry years and their legacy effect,and the trees had lower recovery rates after consecutive dry years.Our results highlight that consecutive dry years pose a new threat to plantations under climate warming,and thus,the effect of climate extremes on tree growth should be considered in growth models in semiarid areas.

Climate change filtered out resource-acquisitive plants in a temperate grassland in Inner Mongolia,China

Global climate change has led to the decline of species and functional diversity in ecosystems,changing community composition and ecosystem functions.However,we still know little about how species with different resource-use strategies(different types of resource usage and plant growth of plants as indicated by the spectrum of plant economic traits,including acquisitive resource-use strategy and conservative resource-use strategy)would change in response to climate change,and how the changes in the diversity of species with different resource-use strategies may influence community-level productivity.Here,using long-term(1982–2017)observatory data in a temperate grassland in Inner Mongolia,we investigated how climate change had affected the species richness(SR)and functional richness(FRic)for the whole community and for species with different resource-use strategies.Specifically,based on data for four traits representing leaf economics spectrum(leaf carbon concentration,leaf nitrogen concentration,leaf phosphorus concentration,and specific leaf area),we divided 81 plant species appearing in the grassland community into three plant functional types representing resource-acquisitive,medium,and resource-conservative species.We then analyzed the changes in community-level productivity in response to the decline of SR and FRic at the community level and for different resource-use strategies.We found that community-level SR and FRic decreased with drying climate,which was largely driven by the decline of diversity for resource-acquisitive species.However,community-level productivity remained stable because resource-conservative species dominating this grassland were barely affected by climate change.Our study revealed distinctive responses of species with different resource-use strategies to climate change and provided a new approach based on species functional traits for predicting the magnitude and direction of climate change effects on ecosystem functions.
