The development of RNA interference(RNAi) therapy offers a potential solution for Alzheimer's disease(AD). However, the brain-blood barrier(BBB) with its selective permeability and pharmacokinetic-related challenges poses restrictions on the delivery of small interfering RNA(siRNA) to the central nervous system(CNS). In this study, we demonstrate that the incorporation of 2'-fluoro(2'-F) substitutions and L-carnitine modification facilitates the self-assembly of siRNA through triple interaction, leading to the formation of nanorings, called LCSF-NR. Based on the enhanced cellular uptake and lysosomal escape by 2'-F substitution and the transport across the BBB promoted by L-carnitine, the nanorings realized the improved braintargeted delivery of siRNA, both in zebrafish and mice models. Moreover, our findings highlight the therapeutic potential of LCSF-NR formulation in an AD zebrafish model through a synergistic effect of downregulating the β-site APP cleavage enzyme1(BACE1) gene and L-carnitine-mediated neuroprotection, effectively inhibiting pathological processes. Overall, these results suggest that the chemical modification-based siRNA self-assembly strategy enables trans-BBB delivery and presents a concise approach for synergistic therapy of AD.

Laryngeal Cancers at the pathological Anatomical laboratory (ACP) in Dakar about 215 Cases

Introduction: Laryngeal cancer is the first cancer of upper aerodigestive tract. Dysphonia, dyspnea and dysphagia are evocative signs. Diagnosis is histological and squamous cell carcinoma is the most common type. The objective of our study was to discuss epidemiological and anatomo-pathological characteristics of laryngeal cancers diagnosed in different pathological anatomy and cytology (ACP) laboratories of Dakar hospitals. Material and method: This was a retrospective study spanning from January 2013 to December 2018 at the pathological anatomy laboratories of Fann, Aristide Le Dantec and Idrissa Pouye hospitals. All patients with laryngeal cancer confirmed at histology were included. Data collection was based on clinical records of the patients and archives of histological reports of the pathological laboratories. Data analysis was performed under the Excel software. Results: We collected 215 cases of laryngeal cancer. The average age was 58.01 years with extremes of 07 and 94 years. The male sex was predominant with 183 patients (85.12%) against 32 women (14.88%). The most noted risk factor was tobacco which affected 14 patients, or 60.87%. Three patients (13.04%) did not present any alcohol-smoking impregnation. Clinically, dysphonia was noted in 22 patients (26.51%). It was associated with dyspnea in 0.48% of cases and dysphagia in 3.61%. Laryngoscopy was performed in 62 patients or 28.83% of cases, nasofibroscopy in 6.45% of cases. Cord arythenoid fixity was noted in two patients (3.23%) and hypo-mobility in 1 patient (1.61%). The most frequent local extension was involvement of the piriform sinus with 11.29% of cases. Palpable lymphadenopathy reported in 17 patients (20.48%). pathological examination was performed in all patients after post endoscopic biopsy in 92 patients (42.79%), and after obtaining the operative specimen in 123 cases (57.21%). OBJECTIVE: To reach consensus on the diagnostic criteria of syndrome of dampness obstruction in idiopathic membranous nephropathy(IMN) patients by literature research and expert investigation(interviews and a Delphi method). METHODS: Our study was consistent with T/CACM 1336-2020. We searched the monographs and references published in the past 40 years(1983-2022), and established the diagnostic criteria pool of waterdampness syndrome and dampness-turbidity syndrome in Traditional Chinese Medicine(TCM) based on literature by using frequency statistics and correlation analysis. Expert investigation(interview method and two rounds of Delphi method) was used to form the diagnostic criteria of water-dampness syndrome and dampnessturbidity syndrome of idiopathic membranous nephropathy. Clinical diagnostic test research was carried out, and compared with “Diagnostic Criteria for dampness syndrome”(T/CACM 1454-2023) to evaluate the authenticity, reliability and clinical application value of the standard. RESULTS: A total of 122 relevant guides, standards, monographs and documents were included through searching books and Chinese databases. Four experts were interviewed and two rounds of delphi method(75 experts nationwide) were carried out. The experts' opinions are relatively concentrated and the differences are small. Based on the weight of each index, the diagnostic criteria indexes of water-dampness syndrome and dampness-turbidity syndrome were selected. After discussion by the core group members, the diagnostic model of "necessary symptoms and optional symptoms" was established, and the final diagnostic criteria of waterdampness syndrome and dampness-turbidity syndrome were established. One hundred and ninety-one inpatients and outpatients of Guangdong Provincial Hospital of Chinese Medicine from January 2021 to February 2023 were included in Diagnostic test study. Does Argan Oil Supplementation Affect Metabolic Parameters and Behavior in Wistar Rats?

Argan oil is renowned for its particular biochemical profile: high-fat oleic and linoleic acids, tocopherols, sterols, polyphenols. This composition gives it nutritional, therapeutic and preventive properties against dermatological, metabolic and proliferative diseases. The composition of argan oil assigns its benefits to mental health;it would be provided with possible effects on the prevention and/or cure of stress related disorder. This work aims to evaluate the impact of argan oil dietary on the behavioral response, biochemical and hematological constants and histological profiles of adrenal involved in emotional responses to stress. The variation of these parameters was evaluated in Wistar rats receiving dietary 10 ml/Kg/day of argan oil, starting from weaning, for 13 weeks. Our results show that supplementation has resulted in an increase in locomotor activity, reduced sensitivity to frightening environments with sex dependent variation. Moreover, lipid markers, corticosterone and lymphocytes show a rising trend. If the important role of argan oil diet in cardio-metabolic health is generally well recognized;for mental health, it is the first study that needs further investigation linking between the nervous system, inflammation parameters and metabolism.

Multi-omics analysis and experimental verification reveal testicular fatty acid metabolism disorder in non-obstructive azoospermia

Increasing evidence implicates disruptions in testicular fatty acid metabolism as a contributing factor in nonobstructive azoospermia(NOA), a severe form of male infertility. However, the precise mechanisms linking fatty acid metabolism to NOA pathogenesis have not yet been fully elucidated. Multi-omics analyses, including microarray analysis, single-cell RNA sequencing(scRNA-seq), and metabolomics, were utilized to investigate disruptions in fatty acid metabolism associated with NOA using data from public databases. Results identified ACSL6, ACSBG2, and OLAH as key genes linked to fatty acid metabolism dysregulation, suggesting their potential causative roles in NOA. A marked reduction in omega-3 polyunsaturated fatty acids, especially docosahexaenoic acid(DHA), was observed, potentially contributing to the pathological process of NOA. Sertoli cells in NOA patients exhibited apparent fatty acid metabolic dysfunction, with PPARG identified as a key transcription factor(TF) regulating this process. Functional analyses demonstrated that PPARG is crucial for maintaining blood-testis barrier(BTB) integrity and promoting spermatogenesis via regulation of fatty acid metabolism. These findings reveal the pivotal role of fatty acid metabolism in NOA and identify PPARG as a potential therapeutic target.

Network pharmacology and animal experiments revealed the protective effects of Guilong prescription(归龙方) on chronic prostatitis and its possible mechanisms

OBJECTIVE: To investigate the protective effects of Guilong prescription(归龙方, GL) on chronic prostatitis(CP) and unravel the underlying mechanisms of its pharmacological effects. METHODS: The composition of GL was determined via linear ion trap/electrostatic field orbital trap tandem highresolution mass spectrometry, and the identified compounds were performed network pharmacological analysis to predict possible pathways of the effects of GL on CP. A CP rat model was established by carrageenan, and rats were randomly assigned into a Control group, Sham group, CP group, GL low dose(3.5 g/kg) group, GL medium dose(7 g/kg) group, and GL high dose(14 g/kg) group. Hematoxylin-eosin staining of the prostate, and prostate blood-perfusion measured by laser speckle contrast analysis were used to evaluate the efficacy of GL. Expression of intercellular cell adhesion molecule-1(ICAM-1) and induce nitric oxide synthase(i NOS) were determined by immunohistochemistry, and the content of interferon-γ(IFN-γ), interleukin-1β(IL-1β), interleukin-4(IL-4), interleukin-10(IL-10), chemokine ligand 1(CXCL1) and tumor necrosis factor-α(TNF-α) were determined by electro-chemiluminescence assays. The expression of p38 mitogen-activated protein kinase(p38 MAPK), phosphatidylinositol 3-kinase(PI3K), ribosomeassociated complex-alpha serine/threonine-protein kinase(Akt), nuclear factor-κ-gene binding p65(NF-κB p65), inhibitor of NF-κB-α(IκBα), glycogen synthase kinase-3β(GSK-3β), and their phosphorylated forms were tested by Western blot. RESULTS: In GL, a total of 48 compounds were identified, including 14 flavonoids, 14 alkaloids, 11 carboxylic acids, 4 lactones, 2 glycosides, 2 terpenoids and 1 aldehyde. Network pharmacological analysis suggested that the mechanism of GL may be related to PI3K-Akt signaling pathway and cytokine expression.
