Fruit wounding occurred at harvest and transportation requires rapid suberization as a major part of the healing process to prevent infection and desiccation. The focus of this work was to explore the mediation of abscisic acid(ABA) on woundinduced suberization and to determine expression profiles of specific genes involved in wound-induced suberization in tomato fruit. The measurements of weight loss and fruit firmness suggested wound-induced suberization started at 2 d after wounding. The suberization process with the accumulation of suberin polyphenolics(SPP) and polyaliphatics(SPA) observed through autofluorescence microscopy and Sudan IV staining was accelerated by ABA. Expressions of SlPAL5 and Sl4 CL involved in the synthesis of SPP reached the highest at 4 and 8 d after wounding following ABA application, respectively. Associated with SPA biosynthesis, SlLACS1 and SlLACS2 showed the most abundant transcripts at 8 and 6 d in ABA group, respectively. Transcript levels including SlKCSs, SlCYP86 B1, SlFAR3, and Sl GPATs were up-regulated at 2 d after wounding by ABA. Activities of polyphenol oxidase and lipoxygenase were also enhanced during wound-induced suberization following ABA application. The results in this study proved that ABA accelerated the wound-induced suberization progress by increasing the transcript levels of relevant genes in postharvest tomato fruit.

To determine the antioxidant activity of the water (WE), methanol (ME), ethanol (EE) and ethyl acetate extracts (EAE) from the leaves of Cistus monspeliensis, several methods such as 1,1-diphenyl-2-picryhydrazyl (DPPH) radical-scavenging assay, 2,2’-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation decolorization assay, reducing power assay, metal chelating assay, Thiobarbituric acid test (TBARS) and β-carotene linoleic acid system assay were investigated. Because of the important roles of total phenolics and total flavonoids as antioxidants, the amounts of total phenolics and total flavonoids in the extracts were also determined. The WE and ME showed a higher scavenging activity on the DPPH and ABTS radical. Both extracts also exhibited a strong chelating effect on Fe2+ and performed the best in the reducing power assay. On the other hand, the WE and ME were found to have the higher total phenolic contents (23.13 and 23.25 mg GAE/g of extract respectively) and total flavonoid contents (55.08 and 47.77 mg rutin/g of extract respectively). Thus, both extracts are promising alternatives to synthetic substances as food ingredients with antioxidant activity. The ME and the WE showed the similar levels in phenolic content.

Aim: The aim of the current study is to determine: (1) the prevalence of extended-spectrum β-lactamase-producing K. pneumoniae (ESBL-Kp) isolated from clinical samples and a hospital environment in Hassan II Hospital (Settat, Morocco);(2) the associated risk factors of ESBL-Kp infections;(3) the link between clinical and environmental isolates. Methods: During the study period (April 2010 to March 2011), all patients infected and hospital environment sites contaminated by K. pneumoniae were considered as the potential study population and environmental site. The clinical data were collected to identify risk factors for ESBL carriage of K. pneumoniae infection. Screening of ESBL-and carbapenemase-producing isolates was performed by using a double-disk synergy test and the modified Hodge test, respectively. ESBL-Kp isolates were tested for the presence of genes encoding β-lactamases and were investigated by PCR. The clonal relationship between ESBL-producing isolates was analysed by ERIC- and REP-PCR method. Results: The overall prevalence of ESBL-Kp among clinical and environmental K. pneumoniae isolates was 35.13% (13/37) and 4.04% (4/99), respectively. The main risk factors for carrying ESBL-Kp were renal disease (46.15%), recent surgery (53.84%), previous hospitalisation (76.92%), and the presence of many invasive devices (53.84%). All ESBL isolates were multidrug resistant. The blaCTX-M group1and blaSHV (70.58% for each) were the most prevalent followed by blaTEM (52.94%). Thirteen strains expressed at least two bla genes. One isolate was positive in the modified Hodge test and was a blaOXA-48 producer. ERIC and Rep-PCR methods revealed an epidemic clonal dissemination of these isolates. Conclusion: The emergence of OXA-48 carbapenemase, endemic clonal dissemination and multi-drug resistance of ESBL-Kp isolates in our institution is highly alarming.

The chemical composition of essential oils obtained from Artemisia herba-alba and Mentha pulegium were determined. The essential oils were analyzed by gas chromatography coupled with mass spectrometry （GC/MS）. Their antibacterial activity was studied in vitro against three standard strains： E. coli ATCC 25922, Staphylococcus aureus ATCC 29213, Pseudomonas aeruginosa ATCC 27853, and five clinical strains： Enterobacter cloacae, Staphylococcus aureus, Pseudomonas pyocyanique, Enterococcus faecium, and E. coli. Nineteen constituents were identified in A. herba-alba essential oil representing 99.57% of the total composition The major component was α-thujone （59.07%）. The bacterial strains were inhibited at concentrations ranging from 1.25 μL/mL to 5μL/mL and killed at concentrations ranging from 1.25 μL/mL to 10 μL/mL. M. pulegium resulted in the identification of eighteen constituents representing 99.48% of the total composition. The main component was pulegone （78.07%）. The minimal inhibitory （MIC） and bactericidal （MBC） concentrations were ranging from 1.25 μL/mL to 2.5 μL/mL.

In this work,38 different organic emerging contaminants(ECs),belonging to various chemical classes such as pharmaceuticals(PhCs),endocrine-disrupting chemicals(EDCs),benzotriazoles(BTRs),benzothiazoles(BTHs),and perfluorinated compounds(PFCs),were initially identified and quantified in the biologically treated wastewater collected from Athens’(Greece)Sewage Treatment Plant(STP).Processes already used in existing STPs such as microfiltration(MF),nanofiltration(NF),ultrafiltration(UF),UV radiation,and powdered activated carbon(PAC)were assessed for ECs’removal,under the conditions that represent their actual application for disinfection or advanced wastewater *** results indicated that MF removed only one out of the 38 ECs and hence it was selected as pretreatment step for the other *** radiation in the studied conditions showed low to moderate removal for 5 out of the 38 *** showed better results than UF due to the smaller pore sizes of the filtration ***,this enhancement was observed mainly for 8 compounds originating from the classes of PhCs and PFCs,while the removal of EDCs was not statistically *** the various studied technologies,PAC stands out due to its capability to sufficiently remove most *** particular,removal rates higher than 70%were observed for 9 compounds,22 were partially removed,while 7 demonstrated low removal *** on our screening experiments,future research should focus on scaling-up PAC in actual conditions,combining PAC with other processes,and conduct a complete economic and environmental assessment of the treatment.

The encapsulation in a nutritive matrix is an effective technique which allows the conservation of vitro-derived explants and protects their viability. The encapsulated propagules are generated by submerging explants in a natural polysaccharide solution of sodium alginate and calcium chloride allowing to obtain uniform capsules. In this work, we present preliminary results of a study on the effect of an encapsulation protocol on any local fruit varieties, conserved in a Regional in vitro germplasm bank.

We estimated water-use efficiency and potential photosyn- thetic assimilation of Holm oak （Quercus ilex L.） on slopes of NW and SW aspects in a replicated field test examining the effects of intensifying drought in two Mediterranean coppice forests. We used standard tech- niques for quantifying gas exchange and carbon isotopes in leaves and analyzed total chlorophyll, carotenoids and nitrogen in leaves collected from Mediterranean forests managed under the coppice system. We pos- tulated that responses to drought of coppiced trees would lead to differ- ential responses in physiological traits and that these traits could be used by foresters to adapt to predicted warming and drying in the Mediterra- nean area. We observed physiological responses of the eoppiced trees that suggested acclimation in photosynthetic potential and water-use effi- ciency：（1） a significant reduction in stomatal conductance （p〈0.01） wasrecorded as the drought increased at the SW site; （2） foliar 813C increased as drought increased at the SW site （p〈0.01）; （3） variations in levels of carotenoids and foliar nitrogen, and differences in foliar morphology were recorded, and were tentatively attributed to variation in photosyn- thetic assimilation between sites. These findings increase knowledge of the capacity for acclimation of managed forests in the Mediterranean region of Europe.

Influence of Sanitization Process on Maintaining Sweet Potato Quality for Export

The whole cold-chain for exporting sweet potato(native variety“Abees”),to foreign market included immediate curing operation directly after harvest helped in healing skin texture,however,in order to reduce postharvest soft rot(Rhizopus stolonifer)incidence following trimming,and washing,ultraviolet light(UV-C)treatment was used as a main sanitizer for eliminating the soft *** of the roots to UV-C(254 nm)was applied in a UV-C room on freshly harvested and cured sweet potato while rolling up on a movable line at 20 cm distance for 1,2,and 3 *** combining UV-C treatment with chlorine(200 ppm)on roots,marked and significant reduction of the total microbial load and Rhizopus potential was achieved on root surfaces respectively compared with chlorine *** also reduced soft rot percentage to almost 0%*** 3 months of cold-storage,quality assessment of sweet potato showed that root characteristics were markedly *** ability of UV-C light to induce phenylalanine ammonia lyase(PAL)enzyme activity in root tissue and maintain the activities of peroxidase and polyphenol oxidase,however with slight increase,was ***-C caused an increase of phenol content in sweet potato tissue that made an activation of defense reaction against the rot causal *** the exposure time to UV-C light increased,a higher content of phenols ***,UV-C application caused decrease in sugar content of root tissue that is flavored by soft rot-causal pathogen.
