Radionuclides accidentally released from nuclear power plants,such as ^(137)Cs and ^(60)Co,can lead to severe contamination of marine *** macroalgae are effi cient in absorbing metal elements from seawater.A 10-day simulation was conducted to assess the potential of scavenging cesium(Cs)and cobalt(Co)with live U lva prolifera that was cultured in seawater medium containing Cs and Co in diff erent concentrations(0(the control),10,20,50,and 100 mg/L).In the experiment,5 kg(fresh weight)of *** was cultured in natural seawater in 90-L *** showed that after the experiment,the average bioconcentration factors(BCFs)of the control group were 247.2 and 1126.4 for Cs and Co,*** absolute absorption quantity of *** increased and the BCFs decreased with the increase in Cs and Co *** biosorption of the two metals on the fi rst day well fi t the pseudo-second-order equation(R^(2)>0.95),indicating that adsorption is the rate-limiting step in the total biosorption *** of both metal ions declined signifi cantly in the fi rst hour and decreased by 25.2%and 15.5%in 48 h,***,live *** is effi cient at scavenging Cs and Co in seawater,providing potential applications for the phytoremediation of radionuclides contaminated seawaters.

Pyropia species grow in the intertidal zone and are cold-water adapted. To date, most of the information about the whole plastid and mitochondrial genomes(ptDNA and mtDNA) of this genus is limited to Northern Hemisphere species. Here, we report the sequencing of the ptDNA and mtDNA of the Antarctic red alga Pyropia endiviifolia using the Illumina platform. The plastid genome(195 784 bp, 33.28% GC content) contains 210 protein-coding genes, 37 tRNA genes and 6 rRNA genes. The mitochondrial genome(34 603 bp, 30.5% GC content) contains 26 protein-coding genes, 25 tRNA genes and 2 rRNA genes. Our results suggest that the organellar genomes of Py. endiviifolia have a compact organization. Although the collinearity of these genomes is conserved compared with other Pyropia species, the genome sizes show significant differences, mainly because of the different copy numbers of rDNA operons in the pt DNA and group II introns in the mtDNA. The other Pyropia species have 2–3 distinct intronic ORFs in their cox 1 genes, but Py. endiviifolia has no introns in its cox 1 gene. This has led to a smaller mtDNA than in other Pyropia species. The phylogenetic relationships within Pyropia were examined using concatenated gene sets from most of the available organellar genomes with both the maximum likelihood and Bayesian methods. The analysis revealed a sister taxa affiliation between the Antarctic species Py. endiviifolia and the North American species Py. kanakaensis.

Aestivation is a common strategy of sea cucumbers(Apostichopus japonicus)in response to high-temperature *** studies have individually investigated the immune and physiological alterations at the aestivation ***,these studies have not evaluated the relationship between immunity and *** this study,we explored the transcriptome and metabolome of *** during the aestivation stage to study the *** transcriptome analysis of dormant(aestivation)and revived *** generated 2368 differentially expressed genes,including 927 downregulated genes and 1441 upregulated *** on Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses,the downregulated genes in the dormant group were found to be involved in DNA replication,RNA metabolic process,and protein metabolism,which results in the inhibition of motility,skeletal development,neural activity,cell proliferation,and development of *** contrast,the upregulated genes were found to be associated with fatty acid metabolism,carbohydrate hydrolysis,and *** the metabolome analysis,the downregulated metabolites were found to be associated with fatty acid metabolism,starch and sucrose metabolism,and TCA *** indicates that dormant sea cucumbers consume reserved carbohydrates and fatty acids to maintain low levels of energy *** protein-protein interaction network analysis further revealed that carbohydrate hydrolysis promoted phagocytosis activity in the dormant *** study provides new insights into potential molecular mechanisms of sea cucumber survival in high-temperature conditions,which is critical in aquaculture of sea cucumbers.

BMP2 plays crucial roles in vertebrate developmental process and acts as a bone inducer during osteogenesis. We present here the molecular cloning of bmp2 cDNA from the marine flatfish Cynoglossus semilaevis, and the analysis of bmp2 expression profiling and promoter function. The full length of bmp2 cDNA sequence is 2 048 bp,which encodes a protein of 422 amino acids. Tissue expression distribution of bmp2 was examined in 14 tissues of mature individuals by quantitative real time PCR（qRT-PCR）. The results revealed that bmp2 was expressed ubiquitously, and the highest expression level was detected in the spinal cord. Moreover, bmp2 expression levels were detected at 15 sampling time points of early developmental stages（egg, larva, juvenile and fingerling stages）.The highest expression level of bmp2 was observed at the gastrula stage, which was about ten times higher than those at the other three embryo stages. Whole-mount in situ hybridization showed that the bmp2 signal was strongly detected at the location of the crown-like larval fin, heart and liver, and slightly expressed in the notochord at one day post hatch（dph）; then the expression of bmp2 started to be concentrated in notochord at three dph. Subsequently, we characterized the 5′-flanking region of bmp2 by testing the promoter activity by Luciferase reporter assays. Positive regulatory region was detected at the location of –179 to ＋109. The predicted transcription factor binding sites（E-box binding factors, zinc finger transcription factor, etc.） in this region might participate in the transcriptional regulation of the bmp2 gene.

Meiotic bouquet formation(known as crescent formation in Tetrahymena thermophila) is indispensable for homologous pairing and recombination, but the regulatory mechanism of bouquet formation remains largely unknown. As a conjugation specific cyclin gene, CYC2 knockout mutants failed to form an elongated crescent structure and aborted meiosis progress in T. thermophila. γ-H2 A.X staining revealed fewer micronuclear DNA double-strand breaks(DSBs) in cyc2Δ cells than in wild-type cells. Furthermore, cyc2Δ cells still failed to form a crescent structure even though DSBs were induced by exogenous agents,indicating that a lack of DSBs was not completely responsible for failure to enter the crescent stage. Tubulin staining showed that impaired perinuclear microtubule structure may contribute to the blockage in micronuclear elongation. At the same time,expression of microtubule-associated kinesin genes, KIN11 and KIN141, was significantly downregulated in cyc2Δ ***, micronuclear specific accumulation of heterochromatin marker trimethylated H3 K23 abnormally increased in the cyc2Δ mutants. Together, these results show that cyclin Cyc2 p is required for micronuclear bouquet formation via controlling microtubule-directed nuclear elongation in Tetrahymena.

A new species of bubblegum coral, Paragorgia rubra sp. nov., discovered from a seamount at a water depth of 373 m near the Yap Trench is studied using morphological and molecular approaches. Paragorgia rubra sp. nov. is the fourth species of the genus found in the tropical Western Pacific. The new gorgonian is red-colored, uniplanar, and measures approximately 530 mm high and 440 mm wide, with autozooids distributed only on one side of the colony. Paragorgia rubra sp. nov. is most similar to P. kaupeka Sainchez, 2005, but differs distinctly in the polyp ovals with large and compound protuberances （vs. small and simple conical protuberances） and the medullar spindles possessing simple conical protuberances （vs. compound protuberances）. Moreover, P. rubra sp. nov. differs from P. kaupeka in the smaller length/ width ratio of surface radiates （1.53 vs. 1.75 ）. The genetic distance of the mtMutS gene between P. rubra sp. nov. and P. kaupeka is 0.66%, while the intraspecific distances within Paragorgia Milne-Edwards ＆ Haime, 1857 except the species P. regalis complex are no more than 0.5%, further supporting the establishment of the new species. Furthermore, the ITS2 secondary structure ofP. rubra sp. nov. is also different from those of congeners. Phylogenetic analyses indicate Paragorgia rubra sp. nov. and P. kaupeka form a clade, which branched early within Paragorgia and diversified approximately 15 Mya.
