Polyphenols are the main active components of the anti-inflammatory compounds in dandelion,and chlorogenic acid(CGA)is one of the primary ***,the molecular mechanism underlying the transcriptional regulation of CGA biosynthesis remains ***-CoA:quinate hydroxycinnamoyl transferase(HQT2)is the last rate-limiting enzyme in chlorogenic acid biosynthesis in Taraxacum ***,using the TaHQT2 gene promoter as a probe,a yeast one-hybrid library was performed,and a basic helix-loop-helix(bHLH)transcription factor,TabHLH1,was identified that shared substantial homology with Gynura bicolor DC *** TabHLH1 transcript was highly induced by salt stress,and the TabHLH1 protein was localized in the *** and luteolin concentrations in TabHLH1-overexpression transgenic lines were significantly higher than those in the wild type,while CGA and luteolin concentrations in TabHLH1-RNA interference(RNAi)transgenic lines were significantly *** real-time polymerase chain reaction demonstrated that overexpression and RNAi of TabHLH1 in *** significantly affected CGA and luteolin concentrations by upregulating or downregulating CGA and luteolin biosynthesis pathway genes,especially TaHQT2,4-coumarate-CoA ligase(Ta4CL),chalcone isomerase(TaCHI),and flavonoid-3′-hydroxylase(TaF3′H).Dual-luciferase,yeast one-hybrid,and electrophoretic mobility shift assays indicated that TabHLH1 directly bound to the bHLH-binding motifs of proTaHQT2 and *** study suggests that TabHLH1 participates in the regulatory network of CGA and luteolin biosynthesis in *** and might be useful for metabolic engineering to promote plant polyphenol biosynthesis.

Background:Diabetic retinopathy(DR)is currently the leading cause of blindness in elderly individuals with *** Chinese medicine(tcm)prescriptions have shown remarkable effectiveness for treating *** study aimed to screen a novel tcm prescription against DR from patents and elucidate its medication rule and molecular mechanism using data mining,network pharmacology,molecular docking and molecular dynamics(MD)***:tcm prescriptions for treating DR was collected from patents and a novel tcm prescription was identified using data ***,the mechanism of the novel tcm prescription against DR was explored by constructing a network of core tcms-core active ingredients-core targets-core ***,molecular docking and MD simulation were employed to validate the findings from network ***:The tcms of the collected prescriptions primarily possessed bitter and cold properties with heat-clearing and supplementing effects,attributed to the liver,lung and kidney ***,a novel tcm prescription for treating DR was identified,composed of Lycii Fructus,Chrysanthemi Flos,Astragali Radix and Angelicae Sinensis *** core active ingredients and ten core targets of the novel tcm prescription for treating DR were ***,the novel tcm prescription played a crucial role for treating DR by inhibiting inflammatory response,oxidative stress,retinal pigment epithelium cell apoptosis and retinal neovascularization through various pathways,such as the AGE-RAGE signaling pathway in diabetic complications and the MAPK signaling ***,molecular docking and MD simulation demonstrated that almost all core active ingredients exhibited satisfactory binding energies to core ***:This study identified a novel tcm prescription and unveiled its multi-component,multi-target and multi-pathway characteristics for treating *** findings provide a scientific basis and novel insights into
