Dendrites and axons are delicate neuronal membrane extensions that undergo degeneration after physical injuries. In neurodegenerative diseases, they often degenerate prior to neuronal death. Understanding the mechanisms of neurite degeneration has been an intense focus of neurobiology research in the last two decades. As a result, many discoveries have been made in the molecular pathways that lead to neurite degeneration and the cell-cell interactions responsible for the subsequent clearance of neuronal debris. Drosophila melanogaster has served as a prime in vivo model system for identifying and characterizing the key molecular players in neurite degeneration, thanks to its genetic tractability and easy access to its nervous system. The knowledge learned in the fly provided targets and fuel for studies in other model systems that have further enhanced our understanding of neurodegeneration. In this review, we will introduce the experimental systems developed in Drosophila to investigate injuryinduced neurite degeneration, and then discuss the biological pathways that drive degeneration. We will also cover what is known about the mechanisms of how phagocytes recognize and clear degenerating neurites, and how recent findings in this area enhance our understanding of neurodegenerative disease pathology.

Fabrication and Characterization of Chitosan-Silk Fibroin/Hydroxyapatite Composites via in situ Precipitation for Bone Tissue Engineering

Homogeneous chitosan-silk fibroin/hydroxyapatite （CS-SF/HA） composites were prepared by in situ precipitation method driven by a multiple-order template. The morphology of the composites was investigated by scanning electron microscope （SEM） and transmission electron microscope （TEM）. The compositional analysis was carried out by X-ray diffraction analysis （XRD） and Fourier transformed infrared spectroscopy （FTIR）. The mechanical properties and biocompatibility of the composites were also determined. The results indicated that the inorganic particles of uniform size （50 nm） were well-dispersed among the CS-SF matrices. The compressive modulus of the CS-SF/HA composites was enhanced with the increasing amount of SF. The in vitro results suggested that the MC3T3-E1 osteoblast-like cells on CS-SF/HA composite disks displayed strong bonding and spreading, and the cell proliferation cultured on each composite disk increased throughout the culture period for up to 7 days. Especially, the samples with higher content of SF had much better biological properties. The evidences proved that the CS-SF/HA composites possessed excellent biocompatibility. By using the freeze-drying technique, hierarchical porous scaffolds with pores ranging from 50 μm to 200 μm were obtained. This work presented the advantages of in situ precipitation method to prepare the organic/inorganic composites, and a multiple-order template was introduced in the system to improve the properties of the composites by combining the merits of each organic template.

In this study, recognition of 4 recombinant viral proteins (GST-NHA1) by the antibodies induced by multi-epitope vaccine was testified. Inhibitory activities of these antibodies were also investigated in vitro against four heterologous influenza A viruses (H3N2). Three epitope-specific antibodies purified by affinity chromatography could reduce the plaque formation. Interestingly, the three neutralizing antibodies in combination showed obvious enhancement of inhibitory activity, suggesting that the development of recombinant multi-epitope vaccine might be an effective way against viral mutation.

The increased prevalence of high temperatures （HTs） around the world is a major global concern, as they dramatically affect agronomic productivity. Upon HT exposure, plants sense the temperature change and initiate cellular and metabolic responses that enable them to adapt to their new environmental *** the mechanisms by which plants cope with HT will facilitate the development of molecular markers to enable the production of plants with improved thermotolerance. In recent decades, genetic, physiological, molecular, and biochemical studies have revealed a number of vital cellular components and processes involved in thermoresponsive growth and the acquisition of thermo- tolerance in plants. This review summarizes the major mechanisms involved in plant HT responses, with a special focus on recent discoveries related to plant thermosensing, heat stress signaling, and HT-regulated gene expression networks that promote plant adaptation to elevated environmental temperatures.

Proline plays an important role in regulating synaptic neurotransmission in both mammalian and insect nervous systems. The full-length complementary DNA sequences encoding the proline transporter gene （CsProT） and its alternative spliced transcript （CsProT-ST） were cloned from the striped stem borer （SSB）, Chilo suppressalis, one of the most important rice pests in Asia. The comparison of amino acid sequences showed that CsProT is highly similar to the proline transporter isolated from Manduca sexta （MasProT）. The developmental changes of the relative messenger RNA expression levels of CsProT and CsProT-ST genes were examined in SSB. The expression level of CsProT-ST is much higher than that of CsProT in all developmental periods, suggesting that the Y-end spliced variant is the major transcript instead of CsProT. The highest ex- pression level of both genes in the pupal stage means that proline may be involved in some unclear functions in the metamorphosis from pupa to adulthood.

Transplantation of adult spinal cord tissue(aSCT)is a promising treatment for spinal cord injury(SCI)basing on various types of neural cells and matrix components inside ***,long-term systemic administration of immunosuppressors(***,TAC)is required for the survival of allogeneic tissue,which often associated with severe side effects such as infection,liver damageand renal *** this study,a triglycerol monostearate(TGM)-based TAC delivery system(***@TGM)with high drug loading concentration was developed,which possessed injectable properties as well as sustainable and immune-responsive drug release *** complete transected SCI model,locally injected TAC@TGM could reduce the infiltration of inflammation cells,enhance the survival of transplanted aSCT(***-1^(+)and NF^(+)neurons)and promote the recovery of locomotor ***,controlled release of TAC by TAC@TGM attenuated side effects of TAC on liver and kidneys compared with traditional systemic *** importantly,the developed TAC@TGM system provided a facile single dose of long-term immunosuppressive effect not just for aSCT transplantation,but also for other tissue/organ and cell transplantations.

Effects of simulated microgravity on nitric oxide level in cardiac myocytes and its mechanism

The depression of cardiac contractility induced by space microgravity is an important issue of aerospace medicine research, while its precise mechanism is still unknown. In the present study, we explored effects of simulated microgravity on nitric oxide (NO) level, inducible nitric oxide synthase (iNOS) expression and related regulative mechanism using electron spin resonance (ESR) spectroscopy, immunocytochemistry and in situ hybridization. We found a remarkable in-crease of NO level and up-regulation of iNOS and iNOS mRNA expression in rat cardiac myocytes under simulated microgravity. Staurosporine (a nonselective protein kinase inhibitor), calphostin C (a selective protein kinase C inhibitor), partially inhibited the effect of simulated microgravity. Thus regulative effect of simulated microgravity on iNOS expression is mediated at least partially via activation of protein kinase C. These results indicate that NO system in cardiac myocytes is sensi-tive to simulated microgravity and may play an important role in the depression of cardiac contrac-tility induced by simulated microgravity.

Supramolecular Assemblies in Salts of 2,2＇-Biimidazole with 5-Sulfosalicylic Acid and 3,4,5-Trihydroxybenzoic Acid

Supramolecular assemblies of 2,2＇-biimidazole with 5-sulfosalicylic acid and 3,4,5-trihydroxybenzoic acid, have been synthesized and characterized by single-crystal X-ray diffraction methods. Both the two proton-transfer compounds of 2,2＇-biimidazole with 3-carboxy-4-hydroxybenzenesulfonic acid （5-sulfosalicylic acid, 5-SSA） [namely bis（2-（2-1H-imidazolyl）-1H-imidazolium） 4-hydroxybenzene-3-carboxylate-1-sulfonate monohydrate, 2（C6HTN4）^＋· CTH4068^2-.H2O, （Ⅰ）] and 3,4,5-trihydroxybenzoic acid [namely 2,2＇-bi-1H-imidazolium bis（3,4,5-trihydroxybenzoate） tetrahydrate, C6H8N4^2＋ ·2（C7H5O5）^-·4（H20）, （Ⅱ）] feature extensively hydrogen-bonded three-dimensional network structures having significant interlayer n-n interactions between the cation and anion species. In Ⅰ, a 5-SSA^2- dianionic species results from deprotonation of both the sulfonic and the carboxylic acid groups, all available O-atom acceptors interact with all cation and water molecule donors by hydrogen bonds. In Ⅱ, the formula unit displays a crystallographic inversion symmetry. The structural information about the two complexes between 2,2＇-biimidazole compound and benzenecarboxylic acids obtained in this work will be particularly important for the rational design of supramolecular organic functional materials.

The effects of G protein and cGMP on phytochrome mediated amaranthin biosynthesis in Amaranthus caudatus seedlings were studied. It was shown that G protein agonist cholera toxin induced amaranthin synthesis in darkness, whereas G protein antagonist pertussis toxin inhibited red light induced amaranthin synthesis. Amaranthin synthesis was also induced by exogenous cGMP, while the amaranthin biosynthesis induced by cholera toxin, red light and exogenous cGMP was inhibited by genistein. LY 83583, an inhibitor of guanylyl cyclase, inhibited the amaranthin synthesis induced both by red light and cholera toxin, while it was not able to inhibit the amaranthin synthesis induced by exogenous cGMP. These results suggest that G protein, guanylyl cyclase and cGMP were the candidates in phytochrome signal transduction chain for red light induced amaranthin biosynthesis and the red light signal transduction chain might be as follows: red light→phytochrome→G protein→guanylyl cyclase→cGMP.
