Probiotic Pediococcus pentosaceus restored gossypol-induced intestinal barrier injury by increasing propionate content in Nile tilapia

Background Intestinal barrier is a dynamic interface between the body and the ingested food components, however, dietary components or xenobiotics could compromise intestinal integrity, causing health risks to the host. Gossypol, a toxic component in cottonseed meal(CSM), caused intestinal injury in fish or other monogastric animals. It has been demonstrated that probiotics administration benefits the intestinal barrier integrity, but the efficacy of probiotics in maintaining intestinal health when the host is exposed to gossypol remains unclear. Here, a strain(YC) affiliated to Pediococcus pentosaceus was isolated from the gut of Nile tilapia(Oreochromis niloticus) and its potential to repair gossypol-induced intestinal damage was *** A total of 270 Nile tilapia(2.20 ± 0.02 g) were allotted in 3 groups with 3 tanks each and fed with 3 diets including CON(control diet), GOS(control diet containing 300 mg/kg gossypol) and GP(control diet containing 300 mg/kg gossypol and 10^(8) colony-forming unit(CFU)/g P. pentosaceus YC), respectively. After 10 weeks, addition of P. pentosaceus YC restored growth retardation and intestinal injury induced by gossypol in Nile tilapia. Transcriptome analysis and si RNA interference experiments demonstrated that NOD-like receptors(NLR) family caspase recruitment domain(CARD) domain containing 3(Nlrc3) inhibition might promote intestinal stem cell(ISC) proliferation, as well as maintaining gut barrier integrity. 16S r RNA sequencing and gas chromatography-mass spectrometry(GC-MS) revealed that addition of P. pentosaceus YC altered the composition of gut microbiota and increased the content of propionate in fish gut. Effect of red clover isoflavones on hormone, immune, inflammatory,and plasma biochemistry in lactating dairy cows

This study was to conducted to investigate the effect of red clover isoflavones on the health indicated by immune status and blood biochemistry in dairy cows. Sixty-eight healthy Holstein lactating cows were randomly divided into four treatments(n = 17 per treatment) from 5 blocks according to milk yield using a randomized complete block design. No initial differences in parity(2.13 ± 1.21), days in milk(165 ± 21 d), and milk yield(33.93 ± 3.81 kg/d) between groups. Cows were fed the basal diet supplemented with 0, 2, 4, or 8 g/kg red clover extract(RCE) in diet(dry matter based). Feeding, refusal feed weights, and milk yield were recorded three consecutive days in weeks 0, 4, 8, and 12. Blood was collected from the tail vein of the cows on the last day of weeks 4, 8 and 12, 1 h after the morning feeding,and analyzed for hormones, immunoglobulins, inflammatory markers, and markers of liver and kidney activities. The dry matter intake was significantly decreased by 3.7% in the 8 g/kg group(P 0.05). These results recommend the supplementation of RCE at a level from 2 to4 g/kg DM.

Dietary copper requirement of broilers fed a corn-soybean meal diet during 22—42 d of age

This research was to assess the dietary copper(Cu) requirement of broiler chickens fed a practical cornsoybean meal diet during 22—42 d of age. A total of 288 numbered Arbor Acres male broilers at 22 d of age were randomly allotted 6 treatments with 8 replicate cages(6 broilers per cage) per *** were fed a Cu-unsupplemented corn-soybean meal basal diet(control, containing 7.36 mg Cu/kg) or the basal diet added with 3, 6, 9, 12, or 15 mg Cu/kg from CuSO4·5H2O for 21 d. Quadratic,asymptotic and broken-line models were fitted and the best fitted models were selected to determine dietary Cu requirements. The results revealed that the contents of Cu in serum and liver, m RNA expression levels of Cu-and zinc-containing superoxide dismutase(CuZnSOD) in liver and monoamine oxidase b(MAO B) in heart, as well as protein expression level of CuZnSOD in liver were affected(P < 0.05) by supplemental Cu levels, and the above indices varied linearly and quadratically(P < 0.05)with increasing Cu levels. Dietary Cu requirements assessed according to the best fitted broken-line models(P < 0.05) of the above indexes were 10.45—13.81 mg/kg. It was concluded that mRNA expression levels of CuZnSOD in liver and MAO B in heart, as well as liver CuZnSOD protein expression level were new specific sensitive biomarkers for estimating dietary Cu requirements, and the dietary Cu requirement was recommended to be 14 mg/kg to support Cu metabolic needs related to key Cucontaining enzymes in broilers fed the corn-soybean meal diet during 22—42 d of age, which was higher than the dietary Cu requirement(8 mg/kg) for broilers at the corresponding stage suggested by the Chinese Feeding Standard of Chicken.

Maternal resveratrol improves the intestinal health and weight gain of suckling piglets during high summer temperatures: The involvement of exosome-derived microRNAs and immunoglobin in colostrum

Previous studies have shown that maternal resveratrol improved growth performance and altered the microbial composition of suckling piglets under hot summer conditions. However, it remains unclear how maternal resveratrol improves growth performance of suckling piglets during high summer temperatures. A total of 20 sows(Landrace × Large White; three parity) were randomly assigned to 2 groups(with or without 300 mg/kg resveratrol) from d 75 of gestation to d 21 of lactation during high ambient temperatures(from 27 to 30℃). The results showed that maternal resveratrol supplementation increased total daily weight gain of piglets under hot summer conditions, which is consistent with previous studies. Furthermore, we found that maternal resveratrol improved the intestinal morphology and intestinal epithelial proliferation in suckling piglets. Dietary resveratrol supplementation affected the characteristics of exosome-derived microRNAs(miRNAs) in sow colostrum, as well as the genes targeted by differentially produced miRNAs. MiRNAs are concentrated in the tight junction pathway. Effects of biochanin A on lactational performance,nitrogen metabolism,and blood metabolites in dairy cows

Optimizing nitrogen utilization efficiency and mitigating nitrogen losses in cows plays a pivotal role in fostering economic sustainability within contemporary agricultural *** A(BCA),a natural component in red clover,has the potential to improve nitrogen metabolism in dairy *** primary objective of this study was to probe the impact of biochanin A supplementation on lactational performance,nitrogen metabolism,and blood metabolites in dairy cows.A complete randomized block design experiment was conducted over 28 d,involving 36 multiparous Holstein cows(comparable milk yield=37.1±2.90 kg,BW=642±70.0 kg,days in milk=92±8.0 d,and parity=2.4±0.50),which were allocated to three treatment groups:the Control group(with 0 g/d BCA),the Low group(with 10 g/d per cow BCA),and the High group(with 40 g/d per cow BCA).Biochanin A supplementation improved the lactational performance of cows by increasing milk yield by 6.3%(P=0.007)and feed efficiency by 12.7%(P=0.009).Total intestinal apparent digestibility was unaffected by BCA supplementation(P>0.05),but microbial nitrogen was increased by 30.0%(P=0.002)for promoting nitrogen utilization efficiency by 20.7%(P=0.004).Milk competent yields(protein,lactose,and non-fat milk solid)were increased with increasing BCA supplementation(P 0.05).BCA did not affect body health of dairy ***,none of the plasma endocrine hormones were affected(P>0.05).A total of 95 significantly different metabolites were screened from the plasma metabolites of cows in the BCA-added and non-added *** performing an enrichment analysis of the metabolic pathways associated with the different metabolites,six specific pathways were identified:bile acid biosynthesis,aspartate metabolism,pyrimidine metabolism,arginine and proline metabolism,the urea cycle,and ammonia *** inclusion of BCA is suggested to enhance milk yield and modulate nitrogen metabolism by influencing relevant metabolites within the metabolic pathwa

Sesamin is an effective spleen tyrosine kinase inhibitor against IgE-mediated food allergy in computational， cell-based and animal studies

Food allergy has become a global concern. Spleen tyrosine kinase (SYK) inhibitors are promising therapeutics against allergic disorders. In this study， a total of 300 natural phenolic compounds were firstly subjected to virtual screening. Sesamin and its metabolites， sesamin monocatechol (SC-1) and sesamin dicatechol (SC-2)， were identified as potential SYK inhibitors， showing high binding affinity and inhibition efficiency towards SYK. Compared with R406 (a traditional SYK inhibitor)， sesamin， SC-1， and SC-2 had lower binding energy and inhibition constant (Ki) during molecular docking， exhibited higher bioavailability， safety， metabolism/clearance rate， and distribution uniformity ADMET predictions， and showed high stability in occupying the ATP-binding pocket of SYK during molecular dynamics simulations. In anti-dinitrophenyl-immunoglobulin E (Anti-DNP-IgE)/dinitrophenyl-human serum albumin (DNP-HSA)-stimulated rat basophilic leukemia (RBL-2H3) cells， sesamin in the concentration range of (5 – 80) μmol/L influenced significantly the degranulation and cytokine release， with 54.00% inhibition against β-hexosaminidase release and 58.45% decrease in histamine. In BALB/c mice， sesamin could ameliorate Anti-DNP-IgE/DNP-HSA-induced passive cutaneous anaphylaxis and ovalbumin (OVA)-induced active systemic anaphylaxis reactions， reduce the levels of allergic mediators (immunoglobulins and pro-inflammatory cytokines)， partially correct the imbalance of T helper (Th) cells differentiation in the spleen， and inhibit the phosphorylation of SYK and its downstream signaling proteins， including p38 mitogen-activated protein kinases (p38 MAPK)， extracellular signal-regulated kinases (ERK)， and p65 nuclear factor-κB (p65 NF-κB) in the spleen. Thus， sesamin may be a safe and versatile SYK inhibitor that can alleviate IgE-mediated food allergies.
