Circulating tumor DNA(ctDNA)detection has found widespread applications in tumor diagnostics and treatment,where the key is to obtain accurate quantification of ***,this remains challenging due to the issue of background noise associated with existing *** this work,we developed a synthetic gene circuit-based assay with multilevel switch(termed CATCH)for background-free and absolute quantification of *** multilevel switch combining a small transcription activating RNA and a toehold switch was designed to simultaneously regulate transcription and translation processes in gene circuit to eliminate background ***,such a multilevel switch-based gene circuit was integrated with a Cas9 nickase H840A(Cas9n)recognizer and a molecular beacon reporter to form CATCH for ctDNA *** CATCH can be implemented in one-pot reaction at 35°C with virtually no background noise,and achieve robust absolute quantification of ctDNA when integrated with a digital chip(i.e.,digital CATCH).Finally,we validated the clinical capability of CATCH by detecting drug-resistant ctDNA mutations from the plasma of 76 non-small cell lung cancer(NSCLC)patients,showing satisfying clinical sensitivity and *** envision that the simple and robust CATCH would be a powerful tool for next-generation ctDNA detection.

Scientic Workow applications(SWFAs)can deliver collaborative tools useful to researchers in executing large and complex scientic ***,Scientic Workow Scheduling(SWFS)accelerates the computational procedures between the available computational resources and the dependent workow jobs based on the researchers’***,cost optimization is one of the SWFS challenges in handling massive and complicated tasks and requires determining an approximate(near-optimal)solution within polynomial computational *** by this,current work proposes a novel SWFS cost optimization model effective in solving this *** proposed model contains three main stages:(i)scientic workow application,(ii)targeted computational environment,and(iii)cost optimization *** model has been used to optimize completion time(makespan)and overall computational cost of SWFS in cloud computing for all considered scenarios in this research *** will ultimately reduce the cost for service *** the same time,reducing the cost has a positive impact on the protability of service providers towards utilizing all computational resources to achieve a competitive advantage over other cloud service *** evaluate the effectiveness of this proposed model,an empirical comparison was conducted by employing three core types of heuristic approaches,including Single-based(i.e.,Genetic Algorithm(GA),Particle Swarm Optimization(PSO),and Invasive Weed Optimization(IWO)),Hybrid-based(i.e.,Hybrid-based Heuristics Algorithms(HIWO)),and Hyper-based(i.e.,Dynamic Hyper-Heuristic Algorithm(DHHA)).Additionally,a simulation-based implementation was used for SIPHT SWFA by considering three different sizes of *** proposed model provides an efcient platform to optimally schedule workow tasks by handing data-intensiveness and computational-intensiveness of *** results reveal that the proposed cost optimization model attained an optimal Job completio

SoftwareDefined Networks(SDN)introduced better network management by decoupling control and data ***,communication reliability is the desired property in computer *** frequency of communication link failure degrades network performance,and service disruptions are likely to *** network applications,such as delaysensitive applications,suffer packet loss with higher Round Trip Time(RTT).Several failure recovery schemes have been proposed to address link failure recovery issues in ***,these schemes have various weaknesses,which may not always guarantee service *** paths differ in their roles;some paths are critical because of the higher frequency *** paths frequently share links between primary and *** the affected flows after failure occurrences without investigating the path roles can lead to post-recovery congestion with packet loss and system ***,there is a lack of studies to incorporate path criticality and residual path capacity to reroute the affected flows in case of link *** paper proposed Reliable Failure Restoration with Congestion Aware for SDN to select the reliable backup path that decreases packet loss and RTT,increasing network throughput while minimizing post-recovery *** affected flows are redirected through a path with minimal risk of failure,while Bayesian probability is used to predict post-recovery *** the former and latter path with a minimal score is *** simulation results improved throughput by(45%),reduced packet losses(87%),and lowered RTT(89%)compared to benchmarking works.

video object tracking is an important research topic of computer vision, whichfinds a wide range of applications in video surveillance, robotics, human-computerinteraction and so on. Although many moving object tracking algorithms have beenproposed, there are still many difficulties in the actual tracking process, such asillumination change, occlusion, motion blurring, scale change, self-change and so ***, the development of object tracking technology is still challenging. Theemergence of deep learning theory and method provides a new opportunity for theresearch of object tracking, and it is also the main theoretical framework for the researchof moving object tracking algorithm in this paper. In this paper, the existing deeptracking-based target tracking algorithms are classified and sorted out. Based on theprevious knowledge and my own understanding, several solutions are proposed for theexisting methods. In addition, the existing deep learning target tracking method is stilldifficult to meet the requirements of real-time, how to design the network and trackingprocess to achieve speed and effect improvement, there is still a lot of research space.
