To the Editor:Schizophrenia is a progressive,debilitating disease.[1]Safe and effective treatment performed during the acute phase is the first step to improve prognosis,and it is critically *** is known as a dopamine system stabilizer,and it exerts partial agonism with high affinity at dopamine D2 and serotonin-1A(5-HT1A)receptors as well as antagonism at serotonin-2A(5-HT2A)receptors.[2]It has been shown to be efficacious in amelioration of both positive and negative symptoms of acute schizophrenia,and it shows better tolerance than most antipsychotics.[3]We conducted a real-world study to further confirm the efficacy and safety of aripiprazole in patients with acute episodes of schizophrenia in China and tried to explore the clinical variables,including various drug regimens,which may be correlated with the efficacy of aripiprazole,and provide a basis for the optimization of aripiprazole treatment for patients with acute schizophrenia.

D<sub>1</sub>and TrkB Receptors Take Charge of the Molecular Antidepressant Action in Cultured Astroglial Cells

In psychopharmacology of depression, we observe two ways of research. One group is focused on catecholamines action. Second one fixes attention on neuronal morphogenesis and synaptic plasticity. The intimate connection of astrocytes, neurons and synaptic endings determines glial participation in neural homeostasis. Consequently this situation enlarges the role of astrocytes in the CNS synaptic plasticity. Brain Derived Neurotrophic factor and its receptor TrkB suppose to coordinate both of the above mentioning signaling pathways in depression disturbances. In our experiment, we have exploited striatal tissue because in our opinion this structure is misjudged in pathophysiology of depression alas;Several hypothesis proposed striatum as important in future intention activity structure. RT-PCR analysis was used to determine D1, BDNF and TrkB mRNA expression in cultured striatal astroglial cells. Administration of three representative antidepressants (ADs) like amitriptyline, moclobemide and sertraline to astroglial culture medium increase the D1, BDNF/TrkB mRNA expression. Our previous study showed that the stimulation of cAMP to CREB pathway after D1 receptors excitation constituted a common response to ADs. The present results signify that D1, BDNF/TrkB link which is next neural track (after cAMP/PKA) involved in the CNS adaptation to external conditions altered by chronic ADs treatment. Moreover, the striatum tissue appears to be important formation which takes an active part in antidepressant action thus essential in depression disorder etiology.

Background:Although many studies focus on investigating the new therapeutic target of acute lung injury(ALI),there still needs more works on exploring the role of other molecular in the pathology of *** specificity phosphatase(DUSP)8 has been reported to participate in the process of ***,the potential role of DUSP8 in lipopolysaccharide(LPS)-induced murine ALI is still ***:Firstly,murine ALI was established by LPS treatment and further measured by hematoxylin-eosin ***,the expression of DUSP8 in lung tissues was analyzed by real-time polymerase chain ***,DUSP8 overexpression vector was utilized and the protein level of DUSP8 was detected by western ***,the pathologic injury was measured by hematoxylin-eosin staining and wet/dry ***,we cultured bone-marrow-derived macrophages and detected the expression of DUSP8 by real-time polymerase chain reaction and western blot after LPS *** addition,DUSP8 overexpression vector was transfected into bone-marrow-derived macrophages and the levels of related inflammatory cytokines were measured by enzyme-linked immunosorbent ***:Compared with the control mice,DUSP8 significantly decreased in LPS-induced murine ***,DUSP8 overexpression could attenuate the pathology of ALI by altering lung inflammation and ***,DUSP8 was also reduced in LPS-treated BMDM and reached a peak at ***,DUSP8 overexpression could reduce the productions of related inflammatory cytokines,such as interleukin-1β,tumor necrosis factor-αand interleukin-6 in LPS-treated bone-marrow-derived ***:DUSP8 is reduced in LPS-induced murine acute lung injury and DUSP8 overexpression could ameliorate the pathologic injury of ALI by altering macrophage inflammation responses.

Consistent neglect of complex psychobiological processes involved in human health and diseases often leads to phar-macological discrepancies between preclinical and therapeutic observations. Numerous other pharmacological paradoxes arise from common practices of using dosing schedules in preclinical studies not identical to those in clinical studies. An Ayurveda centered holistic psychopharmacological strategy evolving from our efforts to identify plants potentially useful for preventing and combating illnesses and diseases could be a more rapid, rational, economically feasible, and realistic one for resolving such discrepancies and paradoxes. In this article, some observations made recently to experimentally very this conviction with the use of some well known medicinal plants are summarized and discussed in short. In addition, usefulness of the strategy for identifying edibles potentially useful for health care purposes, or for more objectively understanding “Ayurvedic biology” is pointed out.

BACKGROUND Subgrouping patients with major depressive disorder is a promising solution for the issue of ***,the link between available subtypes and distinct pathological mechanisms is weak and yields disappointing results in clinical *** To develop a novel approach for classification of patients with time-dependent prescription patterns at first onset in real-world *** Drug-naive patients experiencing their first major depressive episode(n=105)participated in this *** agents prescribed in the first 24 mo following disease onset were recorded monthly and categorized as antidepressants,augmentation agents,and *** cumulative doses of agents in each category were converted into relevant *** parameters were used to summarize the time-dependent prescription patterns for each psychotropic load:Stability,amount,frequency,and the time trend of monthly prescriptions.A K-means cluster analysis was used to derive subgroups of participants based on these input parameters of psychotropic agents across 24 *** validity of the resulting data-driven clusters was compared using relevant severity *** Four distinct clusters were derived from K-means analysis,which matches experts’consent:"Short-term antidepressants use","long-term antidepressants use","long-term antidepressants and sedatives use",and"long-term antidepressants,sedatives,and augmentation use".At the first 2 years of disease course,the four clusters differed on the number of antidepressants used at adequate dosage and duration,frequency of outpatient service use,and number of psychiatric *** the first 2 years following disease onset,depression severity was differed in the four *** Our findings suggested a new approach to optimize the subgrouping of patients with major depressive disorder,which may assist future etiological and treatment response studies.
