Bcl-2 down modulation in WEHI-3B/CTRES cells resistant to Cholera Toxin(CT)-induced apoptosis

The very different effects of Cholera Toxin (CT) on cell growth and proliferation may depend on the type of ganglio-side receptors in cell membranes and different signal transduction mechanisms triggered,but other functions relatedto the drug resistance mechanisms can not be *** effect of CT treatment on the "in vitro" clonogenicity,thePopulation Doubling Time (PDT),apoptosis,PKA activation and Bax and Bcl-2 expression was evaluated in WEHI-3Bcell line and its CT-resistant subclone (WEHI-3B/CTRES).In WEHI-3B parental cells the dramatic accumulation ofcAMP induced by CT correlated well with PKA activation,increased PDT value,inhibition of clonogenicity and apop-tosis.H-89 treatment inhibited PKA activation by CT but did not protect the cells from apoptosis and growth *** WEHI-3B/CTRES no significant CT-dependent accumulation of cAMP occurred with any increase of PKA activityand *** CT resistant cells (WEHI-3B/CTRES),Bcl-2 expression was down regulated by both CT or drug treatment(eg.,ciprofloxacin,CPX) although these cells were protected from CT-dependent apoptosis but not from *** from other cell models described,down regulation of Bcl-2 is proved to be independent on cAMPaccumulation and PKA *** observations support the implication of cAMP dependent kinase (PKA) in theinhibition of WEHI-3B cells growth and suggest that,in WEHI-3B/CTRES,Bcl-2 expression could be modulated byCT in the absence of cAMP *** in consideration of many contradictory data reported in literature,ourcell models(of one sensitive parental cell strain and two clones with different uncrossed specific resistance to CT andCPX)provides a new and interesting tool for better investigating the relationship between the CT signal transductionmechanisms and Bcl-2 expression and function.

The widespread presence of iron and sulfur compounds such as pyrite in marine waterlogged archeological wood(WAW)can cause irreversible damage to the safety of its *** issue has been a longstanding concern for cultural heritage conservation *** this study,we examined the distribution and phase composition of Fe and sulfur compounds in wood samples obtained from the Nanhai I shipwreck using ESEM-EDS,micro-Raman spectroscopy,and an X-ray *** removal of iron from WAW samples of the Nanhai I shipwreck using Acidithiobacillus ferrooxidans(***)was evaluated using conductivity and ICP-AES *** results showed that *** effectively improved the removal of iron from *** degradation of fresh healthy wood during treatment was also analyzed using infrared spectroscopy,and the results showed that the treatment had little effect on the samples over a short *** study demonstrates,for the first time,the feasibility of iron extraction from marine WAW by *** was also the first attempt in China to apply biological oxidation to the removal of iron from marine archeological materials.

Dementia is a comprehensive category of brain diseases that is great enough to affect a person＇s daily functioning. The most common type of dementia is Alzheimer＇s disease, which makes most of cases. New researches indicate that gastrointestinal tract microbiota are directly linked to dementia pathogenesis through triggering metabolic diseases and low-grade inflammation progress. A novel strategy is proposed for the management of these disorders and as an adjuvant for psychiatric treatment of dementia and other related diseases through modulation of the microbiota （e.g. with the use of probiotics）.

Objective:To formulate gentamicin liposphere by solvent-melting method using lipids and polyethylene glycol 4000(PEG-4000) for oral ***:Gentamicin lipospheres were prepared by melt-emulsification using 30%w/w Phospholipon(?) 90H in Beeswax as the lipid matrix containing *** lipospheres were characterized by evaluating on encapsulation efficiency,loading capacity,change in pH and the release profile. Antimicrobial activities were evaluated against Escherichia coli,Pseudomonas aeruginosa. Salmonella paratyphii and Staphylococcus aureus using the agar diffusion ***: Photomicrographs revealed spherical particles within a micrometer range with minimal growth after 1 *** release of gentamicin in vitro varied widely with the PEC-4000 contents. Moreover,significant(P>0.05) amount of gentamicin was released in vivo from the formulation. The encapsulation and loading capacity were all high,indicating the ability of the lipids to take up the *** antimicrobial activities were very high especially against Pseudomonas compare to other test *** strongly suggested that the formulation retain its bioactive ***:This study strongly suggest that the issue of gentamicin stability and poor absorption in oral formulation could be adequately addressed by tactical engineering of lipid drug delivery systems such as lipospheres.

Ectoine is a high-value stabilizer and protective agent with various applications in enzyme industry,cosmetics,and *** this study,rational engineering strategies have been implemented in Escherichia coli to efficiently produce ***,the synthetic pathway of ectoine was constructed in *** MG1655 by introducing an artificial thermal switch system harboring the ectABC cluster from Halomonas elongate,and the resulting strain produced 1.95 g/L ***,crr encoding the glucose-specific enzyme II domain A of phosphotransferase system and iclR encoding the glyoxylate shunt transcriptional repressor were deleted in *** for enhancing the oxaloacetate supply,leading to the increasement of the ectoine titer to 9.09 g/***,thrA encoding the bifunctional aspartokinase/homoserine dehydrogenase was removed from the genome to weaken the competitive pathway;simultaneously,an endogenous feedback-resistant lysC was overexpressed to complement the enzymatic activity deficiency of the aspartate kinase,leading to 30.36%increase of ectoine ***,the expression of phosphoenolpyruvate carboxylase was modulated with varying gradient strength promoters to accelerate the biosynthesis efficiency of ***,aspDH encoding aspartate dehydrogenase from Pseudomonas aeruginosa PAO1 was overexpressed to further improve the biosynthesis of *** final strain MWZ003/pFT28-ectABC-EclysC^(*)-aspDH-ppc3 produced 30.37 g/L ectoine after 36-h fed-batch fermentation with a yield of 0.132 g/g glucose and a productivity of 0.844 g/(L h).

Many studies have shown that the development of allergic asthma is associated with intestinal microbiota dysbiosis. Based on the gut-lung axis theory， probiotic intervention may be a potential strategy for respiratory diseases. Lacticaseibacillus paracasei K56 was reported to regulate gut microbiota in mice. However， its effect on allergic asthma has not been reported. In this study， we investigated the effect of the K56 on ovalbumin-induced asthma and its possible mechanisms. Our results showed that K56 reduced asthma symptoms and inflammatory cell infiltration in the lung of asthmatic mice. And K56 regulated the differentiation of helper T cells in the lung and intestine. Results from 16S rRNA gene sequencing showed that K56 prophylaxis significantly elevated the richness of Akkermansia and Burkholderia. Meanwhile， K56 abrogated the decrease in the levels of short-chain fatty acids (SCFAs) in the feces. In addition， a rebound in the mRNA expression of the SCFA receptors was observed after K56 prophylaxis. Notably， the regulatory T cell (Treg) frequencies and the FFAR3 levels were positively correlated. These results suggest that K56 could attenuate asthma， possibly by modulating the intestinal microbiota and regulating Treg differentiation through SCFA metabolized. Our study showed that K56 may be used as a probiotic to prevent pulmonary inflammation and dysbiosis of the intestinal microbiota in allergic asthma.
