Quality evaluation of Nothofagus pumilio seeds linked to forest management and climatic events

Background Forest ecosystems undergo significant transformations due to harvesting and climate fluctuations, emphasizing the critical role of seeding in natural regeneration and long-term structural preservation. Climate change further amplifies these dynamics, affecting phenology across species and regions. In Tierra del Fuego(Argentina), Nothofagus pumilio(lenga) forests represent the most important timber resource, and it is managed through different silvicultural strategies. This species demonstrates notable post-disturbance regeneration, yet seed fall exhibits signifi-cant variability, leading to variations in seed quality(e.g., viability). This study aims to assess fluctuations in N. pumilio seed quality, determine how it varies concerning forest management strategies, annual productivity, and the co-occurrence of climatic phenomena including El Ni?o-Southern Oscillation(ENSO) and the Southern Annular Mode(SAM).Results Viable seeds represented 18.4% of the total, notably higher in unharvested than in managed areas. Con-versely, empty seeds were more prevalent in harvested areas(> 75%). Seed quality exhibited significant differences across silvicultural treatments, except for insect-predated seeds, which had similar proportions across all areas, though dispersed retention showed higher predation. When considering years with varying production levels, high-production years favoured full and viable seeds, particularly in unharvested forests and aggregated retention, while low-production years saw reduced viability across all treatments. Quadratic models revealed that viability increased with seed production, where unharvested forests achieved the highest values. Climate variability influenced seed proportions, where ENSO+/SAM+ promoting more full and viable seeds, while ENSO–/SAM+ favoured non-predated seeds, especially in unharvested *** Seed quality varies among treatments and years with different levels of seeding. Objective: To identify the main chemical classes of compounds from aqueous extract of Enterolobium contortisiliquum(E. contortisiliquum) seed bark and to evaluate its antibacterial activity, as well as its potential to increase the activity of antibiotics against strains of Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia ***: Different classes of compounds in the aqueous extract of E. contortisiliquum were evaluated based on the visual changes in the coloration and the formation of precipitate after the addition of specific reagents. The antibacterial activity of the extract and its potential to increase of antibiotic activity of antibiotics drugs, gentamicin and norfloxacin was determined by using the microdilution ***: Our results demonstrated that the following secondary metabolites were presented in E. contortisiliquum seed bark: flavones, flavonols, xanthones, flavononols,chalcones, aurones, flavones and catechins. The extract itself had very low antibacterial activity against all bacterial strains tested(MIC ≥ 1024 μg/m L), but there was an increase in the antibiotic activity of gentamicin and norfloxacin when combined in the subinhibitory concentration(i.e., MIC/8).Conclusions: Our data suggests that E. contortisiliquum seed bark may be an alternative source for new drugs with the potential to increase antibiotic activity against different strains of bacteria.

Pesticides have become an inevitable part of the modern environment as they are widely used in agriculture,household,and public health sectors and,hence,are extensively distributed throughout most ***,organophosphate pesticides are the most commercially favored group of pesticides,with large application areas all over the *** on their fate,these organophosphorus compounds may become bioavailable for microbial *** microbes,such as Aspergillus,Pseudomonas,Chlorella,and Arthrobacter,are capable of coupling a variety of physical and biochemical mechanisms for the degradation of organophosphate pesticides,including adsorption,hydrolysis of P–O alkyl and aryl bonds,photodegradation,and enzymatic ***,such as esterase,diisopropyl fluorophosphatase,phosphotriesterase,somanase,parathion hydrolase,and paraoxonase,have been isolated from microbes to study and understand the catabolic pathways involved in the biotransformation of these xenobiotic *** review highlights various aspects of biodegradation of organophosphate pesticides along with biological and molecular characterization of some organophosphate pesticide-degrading bacteria.

Multiple responses optimization in the development of a headspace gas chromatography method for the determination of residual solvents in pharmaceuticals

An efficient generic static headspace gas chromatography （HSGC） method was developed, optimized and validated for the routine determination of several residual solvents （RS） in drug substance, using a strategy with two sets of calibration. Dimethylsulfoxide （DMSO） was selected as the sample diluent and internal standards were used to minimize signal variations due to the preparative step. A gas chroma- tograph from Agilent Model 6890 equipped with flame ionization detector （FID） and a DB-624 （30 m × 0.53 mm i.d., 3.00 μm film thickness） column was used. The inlet split ratio was 5：1. The influ- encing factors in the chromatographic separation of the analytes were determined through a fractional factorial experimental design. Significant variables： the initial temperature （IT）, the final temperature （FT） of the oven and the carrier gas flow rate （F） were optimized using a central composite design. Response transformation and desirability function were applied to find out the optimal combination of the chromatographic variables to achieve an adequate resolution of the analytes and short analysis time. These conditions were 30 ℃ for IT, 158 ℃ for FT and 1.90 mL/min for F. The method was proven to be accurate, linear in a wide range and very sensitive for the analyzed solvents through a comprehensive validation according to the ICH guidelines.

BACKGROUND The high prevalence and persistence of Helicobacter pylori(H. pylori) infection, as well as the diversity of pathologies related to it, suggest that the virulence factors used by this microorganism are varied. Moreover, as its proteome contains 340hypothetical proteins, it is important to investigate them to completely understand the mechanisms of its virulence and survival. We have previously reported that the hypothetical protein HP0953 is overexpressed during the first hours of adhesion to inert surfaces, under stress conditions, suggesting its role in the environmental survival of this bacterium and perhaps as a virulence *** To investigate the expression and localization of HP0953 during adhesion to an inert surface and against gastric(AGS) *** Expression analysis was performed for HP0953 during H. pylori adhesion. HP0953 expression at 0,3, 12, 24, and 48 h was evaluated and compared using the Kruskal-Wallis equality-of-populations rank test. Recombinant protein was produced and used to obtain polyclonal antibodies for immunolocalization. Immunogold technique was performed on bacterial sections during adherence to inert surfaces and AGS cells, which was analyzed by transmission electron microscopy. HP0953 protein sequence was analyzed to predict the presence of a signal peptide and transmembrane helices, both provided by the ExPASy platform, and using the GLYCOPP platform for glycosylation sites. Different programs, via, I-TASSER, RaptorX, and HHalign-Kbest, were used to perform three-dimensional *** HP0953 exhibited its maximum expression at 12 h of infection in gastric epithelium *** technique revealed HP0953 localization in the cytoplasm and accumulation in some peripheral areas of the bacterial body, with greater expression when it is close to AGS *** analysis revealed the presence of a signal peptide that interacts with the transmembrane region and then allows the release of the protein

Orbit orientation in didelphid marsupials （Didelphimorphia： Didelphidae）

Usually considered a morphologically conservative group, didelphid marsupials present consider- able variation in ecology and body size, some of which were shown to relate to morphological structures. Thus, changes on orbit morphology are likely and could be related to that variation. We calculated orbit orientation in 873 specimens of 16 Didelphidae genera yielding estimates of orbits convergence （their position relative to midsagittal line） and verticality （their position relative to frontal plane）. We then compared similarities in these variables across taxa to ecological, morpho- logical and phylogenetic data to evaluate the influencing factors on orbit orientation in didelphids. We found an inverse relation between convergence and verticality. Didelphids orbits have low ver- ticality but are highly convergent, yet orbit orientation differs significantly between taxa, and that variation is related to morphological aspects of the cranium. Rostral variables are the only morpho- logical features correlated with orbit orientation： increasing snout length yields more convergent orbits, whereas increase on snout breadth imply in more vertical orbits. Size and encephalization quotients are uncorrelated with orbit orientation. Among ecological data, diet showed significant correlation whereas locomotion is the factor that less affects the position of orbits. Phylogeny is uncorrelated to any orbital parameters measured. Ecological factors seemingly play a more import- ant role on orbit orientation than previously expected, and differentiation on orbit orientation seems to be more functional than inherited. Thus, despite the apparent homogeneity on didelphid morphology, there is subtle morphological variability that may be directly related to feeding behavior.

Communities of saprobic fungi on leaf litter of Vismia guianensis in remnants of the Brazilian Atlantic Forest

We examined the mycobiota associated with Vismia guianensis leaf litter in three Atlantic Forest remnants of Brazil＇ s semiarid region. Among the study sites, two remnants were protected forest reserves, whereas the third was influenced by major anthropogenic activities. Eighteen litter samples were collected in wet and dry seasons and were processed by particle filtration technique. A total of 4750 fungal isolates of 142 taxa were identified. Species richness was higher in litter samples collected during wet season. Nonmetric multidimensional scaling multivariate analysis showed differences in the composition of fungal communities among the sampling sites and the seasons. Analysis of similarity showed that the differences were statistically significant （R = 0.85; P = 0.0001）. Our findings revealed that spatial and temporal heterogeneity, and human activities had significant impacts on the saprobic fungi of V. guianensis leaf litter.
