AIM: To determine the protective effect of triple viable probiotics on gastritis induced by Helicobacter pylori (H. pylori) and elucidate the possible mechanisms of protection. METHODS: Colonization of BIFICO strains in the mouse stomach was determined by counting colony-forming units per gram of stomach tissue. After treatment with or without BIFICO, inflammation and H. pylori colonization in the mouse stomach were analyzed by hematoxylin and eosin and Giemsa staining, respectively. Cytokine levels were determined by enzyme-linked immunosorbent assay and Milliplex. The activation of nuclear factor (NF)-kappa B and MAPK signaling in human gastric epithelial cells was evaluated by Western blot analysis. quantitative reverse transcription-polymerase chain reaction was used to quantify TLR2, TLR4 and MyD88 mRNA expression in the mouse stomach. RESULTS: We demonstrated that BIFICO, which contains a mixture of Enterococcus faecalis, Bifido-bacterium longum and Lactobacillus acidophilus, was tolerant to the mouse stomach environment and was able to survive both the 8-h and 3-d courses of administration. Although BIFICO treatment had no effect on the colonization of H. pylori in the mouse stomach, it ameliorated H. pylori -induced gastritis by significantly inhibiting the expression of cytokines and chemokines such as TNF-alpha, IL-1 beta, IL-10, IL-6, G-CSF and MIP-2 (P < 0.05). These results led us to hypothesize that BIFICO treatment would diminish the H. pylori-induced inflammatory response in gastric mucosal epithelial cells in vitro via the NF-kappa B and MAPK signaling pathways. Indeed, we observed a decrease in the expression of the NF-kappa B subunit p65 and in the phosphorylation of I kappa B-alpha, ERK and p38. Moreover, there was a significant decrease in the production of IL-8, TNF-alpha, G-CSF and GM-CSF (P < 0.05), and the increased expression of TLR2, TLR4 and MyD88 induced by H. pylori in the stomach was also significantly reduced following BIFICO treatmen

Traumatic brain injury often causes a variety of behavioral and emotional impairments that can develop into chronic disorders. Therefore, there is a need to shift towards identifying early symptoms that can aid in the prediction of traumatic brain injury outcomes and behavioral endpoints in patients with traumatic brain injury after early interventions. In this study, we used the Smart Cage system, an automated quantitative approach to assess behavior alterations in mice during an early phase of traumatic brain injury in their home cages. Female C57BL/6 adult mice were subjected to moderate controlled cortical impact（CCI） injury. The mice then received a battery of behavioral assessments including neurological score, locomotor activity, sleep/wake states, and anxiety-like behaviors on days 1, 2, and 7 after CCI. Histological analysis was performed on day 7 after the last assessment. Spontaneous activities on days 1 and 2 after injury were significantly decreased in the CCI group. The average percentage of sleep time spent in both dark and light cycles were significantly higher in the CCI group than in the sham group. For anxiety-like behaviors, the time spent in a light compartment and the number of transitions between the dark/light compartments were all significantly reduced in the CCI group than in the sham group. In addition, the mice suffering from CCI exhibited a preference of staying in the dark compartment of a dark/light cage. The CCI mice showed reduced neurological score and histological abnormalities, which are well correlated to the automated behavioral assessments. Our findings demonstrate that the automated Smart Cage system provides sensitive and objective measures for early behavior changes in mice following traumatic brain injury.

AIM： To determine allelic imbalance on chromosomal region 21q22-qter including trefoil factor family genes （TFF） in cholangiocarcinoma （CCA） patients and analyze the correlation between allelic imbalances and clinicopathological parameters. METHODS： quantitative PCR amplification was performed on four microsatellite markers and trefoil factor family genes （TFF1, TFF2, and TFF3） using a standard curve and SYBR Green I dye method. The relative copy number was determined by DNA copy number of tested locus to reference locus. The relative copy number was interpreted as deletion or amplification by comparison with normal reference range. Associations between allelic imbalance and clinicopathological parameters of CCA patients were evaluated by χ^2-tests. Kaplan-Meier method was used to analyze survival. RESULTS： The frequencies of amplification at D21S1890, D21S1893, and TFF3 were 32.5%, 30.0%, and 28.7%, respectively. Patients who had amplification at regions covering D21S1893, D21S1890, and TFF showed poor prognosis, whereas patients who had deletion showed favorable prognosis （mean： 51.7 wk vs 124.82 wk, P = 0.012）. Multivariate Cox regression analysis revealed that amplification of D21S1893, D21S1890 and TFF, blood vessel invasion, and staging were associated with poor prognosis. CONCLUSION： D21S1893-D21S1890 region may harbor candidate genes especially TFF and serine protease family, which might be involved in tumor invasion and metastasis contributing to poor survival. The amplification in this region may be used as a prognostic marker in the treatment of CCA patients.

The inclusion parameters,fracture surface morphology and void growth characteristics of ten- sile and fracture toughness specimens of 2.25Cr-1Mo steels with and without rare-earth (RE)additions have been investigated by quantitative metaltography(QTM),scanning elec- tron microscopy(SEM)and energy dispersive spectroscopy(EDS).There is a substantially higher density of inclusions in the RE-treated steel,which has lower values of fracture proper- ties including critical values of COD and J integral(δ_c and J_(IC)),fracture strain(ε_f) and Charpy V-notch energy(CVN).The fracture surface of the RE-treated steel comprises equiaxed dimples of diameters comparable with its inclusion spacing,whereas for the non-RE-treated steels,a wide range of dimple sizes is found with average diameter much smaller than the corresponding inclusion *** investigation indicates that the lower values of fracture properties for the steel with RE at room temperature may be ascribed to its large content of RE-containing inclusions.

NO is the earliest discovered gas signal molecule which is produced by normal healthy endothelial cells,and it has many functions,such as maintaining cardiovascular homeostasis,regulating vasodilation,inhibiting intimal hyperplasia and preventing atherosclerosis in the blood *** NO release is often observed in the pathological environment,for instance *** was discovered that NO could be released from the human endogenous NO donor by many compounds,and these methods can be used for the treatment of certain diseases in the blood *** this work,a series of copper-loaded polydopamine(PDA)coatings were produced through self-polymerization time for 24,48 and 72 *** chemical composition and structure,coating thickness and hydrophilicity of the different copper-loaded PDA coatings surfaces were characterized by phenol hydroxyl quantitative,X-ray photoelectron spectroscopy,ellipsometry atomic force microscopy and water contact *** results indicate that the thickness and the surface phenolic hydroxyl density of the PDA coatings increased with the polymerization *** copperloaded coating has glutathione peroxidase-like activity,and it has the capability of catalyzing NO releasing from *** surface of the coating showed desirable hemocompatibility,the adhesion and activation of platelets were inhibited on the copper-loaded *** the same time,the formation of the thrombosis was also *** copper-loaded PDA coatings could provide a promising platform for the development of blood contact materials.

AIM:To investigate trefoil factor(TFF) gene copy number,mRNA and protein expression as potential biomarkers in cholangiocarcinoma(CCA).METHODS:TFF mRNA levels,gene copy number and protein expression were determined respectively by quantitative reverse transcription polymerase chain reaction(PCR),quantitative PCR and immunohistochemistry in bile duct epithelium biopsies collected from individuals with CCA,precancerous bile duct dysplasia and from disease-free *** functional impact of recombinant human(rh) TFF2 peptide treatment on proliferation and epidermal growth factor receptor(EGFR) /mitogenactivated protein kinase(MAPK) signaling was assessed in the CCA cell line,KMBC,by viable cell counting and immunoblotting,***:TFF1,TFF2 and TFF3 mRNA expression was significantly increased in CCA tissue compared to disease-free controls,and was unrelated to gene copy ***1 immunoreactivity was strongly increased in both dysplasia and CCA,whereas TFF2 immunoreactivity was increased only in CCA compared to diseasefree *** contrast,TFF3 immunoreactivity was moderately decreased in dysplasia and further decreased in ***-Meier analysis found no association of TFF mRNA,protein and copy number with age,gender,histological subtype,and patient survival *** of KMBC cells with rhTFF2 stimulated proliferation,triggered phosphorylation of EGFR and downstream extracellular signal related kinase(ERK),whereas co-incubation with the EGFR tyrosine kinase inhibitor,PD153035,blocked rhTFF2-dependent proliferation and EGFR/ERK ***:TFF mRNA/protein expression is indicative of CCA tumor progression,but not predictive for histological sub-type or survival ***2 is mitogenic in CCA via EGFR/MAPK activation.

As a global staple,rice has an increasing demand for differentiation and authentication,considering both fraud and health aspects for *** complexity of rice urges a rapid,labor-saving,effective,and economical rice differentiation method.A semi-quantitative method using inductively coupled plasma mass spectrometry(ICP–MS)for fast and highthroughput analysis of rice entire elements with singular value decomposition(SVD)background correction(BC)was *** SVD BC method is the first time that it has been used with ICP–MS to minimize the effect of instrumental *** data preprocessing methods,normalization(NM),error scaling(ES),and standard normal variate(SNV)were evaluated by principal component analysis(PCA)and projected difference resolution(PDR).A fuzzy rule-building expert system(FuRES),super partial least squares discriminant analysis(sPLS-DA),support vector machine(SVM),and SVM classification tree(SVMTreeG)classifiers gave 98.1±0.0,99.8±0.4,99.8±0.4,and 100.0±0.0%prediction rates with all of the data preprocessing methods(NM,SVD BC with one component to reconstruct the background,ES,and SNV)for the differentiation of the nine rice-digested samples,respectively.
